Intensive cropping with conventional tillage results in a decline of soil organic carbon (SOC) with consequent deterioration of soil physical properties. Some studies indicate that this decline in ...SOC can be arrested by way of organic manure application and improved nutrient management practices. This study was conducted to find out the long-term effects of inorganic fertilizer, manure and lime application on organic carbon content and physical properties of an acidic Alfisol (Typic Haplustalf) under an annual soybean–wheat crop rotation. Six treatments namely, control (CON), nitrogen fertilization (NIT), nitrogen and phosphorus (NP), nitrogen, phosphorus and potassium (NPK), NPK plus manure (NPKM) and NPK plus lime (NPKL) from a long-term fertilizer experiment continuing at Ranchi, India, were chosen for this study. Soil samples were collected from the selected treatments after 29 crop cycles and analyzed for physical and chemical properties. The results indicated that SOC content in all the treatments decreased from initial levels, but the decrease was considerably less in NPKM (8.7%) and NPKL (10.9%) treatments than that in NIT (28.3%) treatment. The SOC at 0–15 and 15–30 cm depth was lowest in NIT and CON. The NPKM, NPKL and NPK treatments up to 30 cm soil depth recorded significantly higher SOC than NIT and CON. Application of balanced fertilizer along with manure (NPKM) or lime (NPKL) improved soil aggregation, soil water retention, microporosity and available water capacity and reduced bulk density of the soil in 0–30 cm depth over CON. In contrast, soil aggregate stability, microporosity and available water capacity were significantly lower in the NIT plots than that in CON. The study thus suggests that soil management practices in acidic Alfisols should include integrated use of mineral fertilizer and organic manure or lime to maintain the organic carbon status and physical environment of soil.

Transition from traditional high-fiber to Western diets in urbanizing communities of Sub-Saharan Africa is associated with increased risk of non-communicable diseases (NCD), exemplified by colorectal ...cancer (CRC) risk. To investigate how urbanization gives rise to microbial patterns that may be amenable by dietary intervention, we analyzed diet intake, fecal 16 S bacteriome, virome, and metabolome in a cross-sectional study in healthy rural and urban Xhosa people (South Africa). Urban Xhosa individuals had higher intakes of energy (urban: 3,578 ± 455; rural: 2,185 ± 179 kcal/d), fat and animal protein. This was associated with lower fecal bacteriome diversity and a shift from genera favoring degradation of complex carbohydrates (e.g., Prevotella) to taxa previously shown to be associated with bile acid metabolism and CRC. Urban Xhosa individuals had higher fecal levels of deoxycholic acid, shown to be associated with higher CRC risk, but similar short-chain fatty acid concentrations compared with rural individuals. Fecal virome composition was associated with distinct gut bacterial communities across urbanization, characterized by different dominant host bacteria (urban: Bacteriodota; rural: unassigned taxa) and variable correlation with fecal metabolites and dietary nutrients. Food and skin microbiota samples showed compositional differences along the urbanization gradient. Rural-urban dietary transition in South Africa is linked to major changes in the gut microbiome and metabolome. Further studies are needed to prove cause and identify whether restoration of specific components of the traditional diet will arrest the accelerating rise in NCDs in Sub-Saharan Africa.
Pleural mesothelioma is an aggressive malignancy with limited effective therapies. In order to identify therapeutic targets, we integrated SNP genotyping, sequencing and transcriptomics from tumours ...and low-passage patient-derived cells. Previously unrecognised deletions of SUFU locus (10q24.32), observed in 21% of 118 tumours, resulted in disordered expression of transcripts from Hedgehog pathways and the T-cell synapse including VISTA. Co-deletion of Interferon Type I genes and CDKN2A was present in half of tumours and was a predictor of poor survival. We also found previously unrecognised deletions in RB1 in 26% of cases and show sub-micromolar responses to downstream PLK1, CHEK1 and Aurora Kinase inhibitors in primary mesothelioma cells. Defects in Hippo pathways that included RASSF7 amplification and NF2 or LATS1/2 mutations were present in 50% of tumours and were accompanied by micromolar responses to the YAP1 inhibitor Verteporfin. Our results suggest new therapeutic avenues in mesothelioma and indicate targets and biomarkers for immunotherapy.

(concentration of drug causing 50% inhibition of cell growth) values comparable to adriamycin. At the concentration of 50 μM, complex 3 is more effective in the inhibition of cyclooxygenase and lipooxygenase enzymes, followed by complex 2 and complex 1 in comparison to their respective free NSAID ligands indicating a possible correlation between the inhibition of COX and/or LOX and anticancer properties. Molecular docking studies with COX-2 reveal that complexes 1 and 2 having naproxen and diclofenac ligands exhibit stronger interactions with COX-2 than their respective free NSAIDs and these results are in good agreement with their relative experimentally observed COX inhibition as well as anti-proliferative activities.
B-cell CLL/lymphoma 6 (BCL6) exerts oncogenic effects in several human hematopoietic malignancies including chronic myeloid leukemia (CML), where BCL6 expression was shown to be essential for CML ...stem cell survival and self-renewal during imatinib mesylate (IM) treatment. As several lines of evidence suggest that interferon γ (IFNγ) production in CML patients might have a central role in the response to tyrosine kinase inhibitor (TKI) therapy, we analyzed if IFNγ modulates BCL6 expression in CML cells. Although separate IFNγ or IM treatment only slightly upregulated BCL6 expression, combined treatment induced remarkable BCL6 upregulation in CML lines and primary human CD34+ CML stem cells. We proved that during combined treatment, inhibition of constitutive signal transducer and activator of transcription (STAT) 5 activation by IM allowed the specific enhancement of the STAT1 dependent, direct upregulation of BCL6 by IFNγ in CML cells. By using colony-forming assay, we found that IFNγ enhanced the ex vivo colony or cluster-forming capacity of human CML stem cells in the absence or presence of IM, respectively. Furthermore, inhibition of the transcriptional repressor function of BCL6 in the presence of IM and IFNγ almost completely blocked the cluster formation of human CML stem cells. On the other hand, by using small interfering RNA knockdown of BCL6, we demonstrated that in an IM-treated CML line the antiapoptotic effect of IFNγ was independent of BCL6 upregulation. We found that IFNγ also upregulated several antiapoptotic members of the BCL2 and BIRC gene families in CML cells, including the long isoform of MCL1, which proved to be essential for the antiapoptotic effect of IFNγ in an IM-treated CML line. Our results suggest that combination of TKIs with BCL6 and MCL1 inhibitors may potentially lead to the complete eradication of CML stem cells.

Here we present neutrino oscillation in the framework of quantum walks. Starting from a one spatial dimensional discrete-time quantum walk we present a scheme of evolutions that will simulate ...neutrino oscillation. The set of quantum walk parameters which is required to reproduce the oscillation probability profile obtained in both, long range and short range neutrino experiment is explicitly presented. Our scheme to simulate three-generation neutrino oscillation from quantum walk evolution operators can be physically realized in any low energy experimental set-up with access to control a single six-level system, a multiparticle three-qubit or a qubit–qutrit system. We also present the entanglement between spins and position space, during neutrino propagation that will quantify the wave function delocalization around instantaneous average position of the neutrino. This work will contribute towards understanding neutrino oscillation in the framework of the quantum information perspective.

Posterior polymorphous corneal dystrophy (PPCD, also known as PPMD) is a rare disease involving metaplasia and overgrowth of corneal endothelial cells. In patients with PPCD, these cells manifest in ...an epithelial morphology and gene expression pattern, produce an aberrant basement membrane, and, sometimes, spread over the iris and nearby structures in a way that increases the risk for glaucoma. We previously mapped PPCD to a region (PPCD3) on chromosome 10 containing the gene that encodes the two-handed zinc-finger homeodomain transcription factor TCF8. Here, we report a heterozygous frameshift mutation in TCF8 that segregates with PPCD in the family used to map PPCD3 and four different heterozygous nonsense and frameshift mutations in TCF8 in four other PPCD probands. Family reports of inguinal hernia, hydrocele, and possible bone anomalies in affected individuals suggest that individuals with TCF8 mutations should be examined for nonocular anomalies. We detect transcripts of all three identified PPCD genes (VSX1, COL8A2, and TCF8) in the cornea. We show presence of a complex (core plus secondary) binding site for TCF8 in the promoter of Alport syndrome gene COL4A3, which encodes collagen type IV α3, and we present immunohistochemical evidence of ectopic expression of COL4A3 in corneal endothelium of the proband of the original PPCD3 family. Identification of TCF8 as the PPCD3 gene provides a valuable tool for the study of critical gene regulation events in PPCD pathology and suggests a possible role for TCF8 mutations in altered structure and function of cells lining body cavities other than the anterior chamber of the eye. Thus, this study has identified TCF8 as the gene responsible for approximately half of the cases of PPCD, has implicated TCF8 mutations in developmental abnormalities outside the eye, and has presented the TCF8 regulatory target, COL4A3, as a key, shared molecular component of two different diseases, PPCD and Alport syndrome.

In this work a strategy to grow diamond on β-Ga2O3 has been presented. The ζ-potential of the β-Ga2O3 substrate was measured and it was found to be negative with an isoelectric point at pH ∼ 4.6. The ...substrates were seeded with mono-dispersed diamond solution for growth of diamond. The seeded substrates were etched when exposed to diamond growth plasma and globules of gallium could be seen on the surface. To overcome problem ∼100 nm of SiO2 and Al2O3 were deposited using atomic layer deposition. The nanodiamond seeded SiO2 layer was effective in protecting the β-Ga2O3 substrate and thin diamond layers could be grown. In contrast Al2O3 layers were damaged when exposed to diamond growth plasma. The thin diamond layers were characterised with scanning electron microscopy and Raman spectroscopy. Raman spectroscopy revealed the diamond layer to be under compressive stress of 1.3–2.8 GPa.

ABSTRACT
We present an in-depth spectral and timing analysis of the black hole binary 4U 1630−472 during 2016 and 2018 outbursts as observed by AstroSat and MAXI. The extensive coverage of the ...outbursts with MAXI is used to obtain the hardness intensity diagram (HID). The source follows a ‘c’-shaped profile in agreement with earlier findings. Based on the HIDs of previous outbursts, we attempt to track the evolution of the source during a ‘super’-outburst and ‘mini’-outbursts. We model the broad-band energy spectra (0.7–20.0 keV) of AstroSat observations of both outbursts using phenomenological and physical models. No Keplerian disc signature is observed at the beginning of 2016 outburst. However, the disc appears within a few hours after which it remains prominent with temperature (Tin) ∼ 1.3 keV and increase in photon index (Γ) from 1.8 to 2.0, whereas the source was at a disc dominant state throughout the AstroSat campaign of 2018 outburst. Based on the HIDs and spectral properties, we classify the outbursts into three different states – the ‘canonical’ hard and soft states along with an intermediate state. Evolution of rms along different states is seen although no quasi-periodic oscillations are detected. We fit the observed spectra using a dynamical accretion model and estimate the accretion parameters. Mass of the black hole is estimated using inner disc radius, bolometric luminosity, and two-component flow model to be 3–9 M⊙. Finally, we discuss the possible implications of our findings.
