Background
Chronic Prostatitis/Chronic Pelvic Pain Syndrome (CP/CPPS) is a common disease of urology, of which the pathogenesis and therapy remain to be further elucidated. Quercetin has been ...reported to improve the symptoms of CP/CPPS patients. We aimed to verify the therapeutic effect of quercetin on CP/CPPS and identify the mechanism responsible for it.
Methods
A novel CP/CPPS model induced with Complete Freund Adjuvant in Sprague Dawley rats was established and the prostates and blood specimens were harvested for further measurement after oral administration of quercetin for 4 weeks.
Results
Increased prostate index and infiltration of lymphocytes, up‐regulated expression of IL‐1β, IL‐2, IL‐6, IL‐17A, MCP1, and TNFα, decreased T‐SOD, CAT, GSH‐PX, and increased MDA, enhanced phosphorylation of NF‐κB, P38, ERK1/2, and SAPK/JNK were detected in CP/CPPS rat model. Quercetin was identified to ameliorate the histo‐pathologic changes, decrease the expression of pro‐inflammatory cytokines IL‐1β, IL‐2, IL‐6, IL‐17A, MCP1, and TNFα, improve anti‐oxidant capacity, and suppress the phosphorylation of NF‐κB and MAPKs.
Conclusions
Quercetin has specific protective effect on CP/CPPS, which is mediated by anti‐inflammation, anti‐oxidation, and at least partly through NF‐κB and MAPK signaling pathways.

Hyman hematoma cells of the line SMMC-7721 were cultured and divided into two groups: octreotide group, co-cultured with octreotide 4,7, and 10 microg/ml respectively for 24 h, 48 h, or 72 h, and control group. TUNEL was used to detect the apoptosis of the cells. Flow cytometry was used with terminal deoxynucleotidyl transferase mediated dUTP nick end labeling (TUNEL) technique to detect the apoptosis of the cells. FC was used with direct labeling of monoclonal antibody (CD95/CD178) to detect the expression of Fas and FasL, and the Fas/FasL ratio.
Octreotide increased the apoptotic rate of the cells dose- and time-dependently (all P < 0.05). Octreotide increased the Fas expression rate, FasL expression rate, and Fas/FasL ratio dose- and time-dependently (all P < 0.05).

The field of gene therapy has been increasingly studied in the last four decades, and its clinical application has become a reality in the last 15 years. Traditional Chinese medicine (TCM), an ...important component of complementary and alternative medicine, has evolved over thousands of years with its own unique system of theories, diagnostics and therapies. TCM is well-known for its various roles in preventing and treating infectious and chronic diseases, and its usage in other modern clinical practice. However, whether TCM can be applied alongside gene therapy is a topic that has not been systematically examined. Here we provide an overview of TCM theories in relation to gene therapy. We believe that TCM theories are congruent with some principles of gene therapy. TCM-derived drugs may also act as gene therapy vehicles, therapeutic genes, synergistic therapeutic treatments, and as co-administrated drugs to reduce side effects. We also discuss in this review some possible approaches to combine TCM and gene therapy.

AIM:To assess the corneal sensitivity and the incidences of dry eye after small incision lenticule extraction(SMILE) and femtosecond laser-assisted in situ keratomileusis(FSLASIK).METHODS:The ...Meta-analysis was performed using Rev Man 5.3.We searched on Pub Med from inception to March 2016.Summary weighted mean difference(WMD) and 95% confidence intervals(CIs) were used to analyze the datum.Random-effects or fixed-effects models were chosen up to between-study heterogeneity.The main outcomes were composed of the Ocular Surface Disease Index(OSDI) scores,tear film break-up time(TBUT),Schirmer Test and corneal sensitivity.RESULTS:Eight eligible studies including 772 eyes(386 in SMILE group and 386 in FS-LASIK group) were identified.The parameters have no significiant difference heterogeneity between SMILE and FS-LASIK group preoperatively.There were significant differences between the two groups in OSDI scores at one and three months postoperatively,in TBUT at one and three months postoperatively,in corneal sensitivity at one week,about one month and three months postoperatively.However,there was no significant difference observed in Schirmer Test at the follow-up periods.CONCLUSION:Compare to FS-LASIK,dry eye and the corneal sensitivity recover better in the SMILE group,in first three months after the surgery.
The present study was planned to explore the effect of Lactobacillus (L.) acidophilus on the T helper-17 (Th17) immune response in a mouse model of β-lactoglobulin (β-lg) allergy. Bovine β-lg ...sensitised BALB/c mice were orally administered with different doses of heat-killed L. acidophilus (low, 5×107 colony forming unit (CFU); medium, 5×108 CFU; high, 5×109 CFU) in 200 µL of phosphate buffered saline (PBS) three times a week, starting from 1 week before β-lg sensitisation for 4 weeks. After the allergen challenge, the numbers of blood eosinophils and neutrophils were examined by light microscope; the levels of cytokine (interleukin (IL)-12, IL-4, tumor growth factor (TGF)-β, IL-10, IL-6 and IL-17A), total immunoglobulin E (IgE) and β-lg-specific IgE contents in the serum were measured with enzyme-linked immunosorbent assay (ELISA); The mRNA expression levels of TGF-β, IL-17A,CD25, Foxp3, retinoic acid-related orphan receptor γt (RORγt) and IL-10 were analyzed using real-time polymerase chain reaction (PCR). The results showed that oral administration of L. acidophilus suppressed hypersensitivity responses, attenuated the numbers of inflammatory cells and inhibited IgE production. We found up-regulation of TGF-β and down-regulation of IL-17A in the serum of L. acidophilus-treated group, along with IL-6 levels was significantly decreased than that of the allergy group (p<0.05). Moreover, the mRNA expression levels of CD25, forkhead box P3 and TGF-β were significantly higher in the spleen of L. acidophilus-treated group, while the mRNA expression levels of IL-17A, RORγt and IL-10 were significantly lower than that in the allergy group (p<0.05). In conclusion, the suppression of major allergic symptoms by oral administration of L.acidophilus was probably due to improve the regulatory T (Treg)/Th17 balance and inhibit the IL-6 production.
This study investigated whether the Th17/Treg balance was impaired in mice with β-lactoglobulin (β-Lg) allergies. BALB/c mice were intraperitoneally sensitised with β-Lg. The Th17/Treg ratios of ...allergic mice and healthy controls were calculated, and their correlation with specific transcription factors and cytokines were analysed. The results indicated that sensitisation with β-Lg led to the development of an allergic response, characterised by inflammatory cell infiltration in the intestine and high levels of IgE in the serum. The frequencies of Th17 cells, Th17-related cytokines and RORγt expression were increased, whereas the frequencies of Treg cells, plasma TGF-β level and Foxp3 expression were decreased in β-Lg-sensitised mice. In addition, the Th17/Treg ratios were positively correlated with the serum concentrations of Th2- and Th17-related cytokines. These results suggest that a Th17/Treg imbalance in combination with an increase in the Th2 cell response may be responsible for the development and progression of β-Lg allergy.

Melittin and its derivative have been developed to support effective gene delivery systems. Their ability to facilitate endosomal release enhances the delivery of nanoparticle-based gene therapy. ...Nevertheless, its potential application in the context of viral vectors has not received much attention. Therefore, we would like to optimize the rAAV vector by Melittin analog to improve the transduction efficiency of rAAV in liver cancer cells and explore the mechanism of Melittin analog on rAAV.
Various melittin-derived peptides were inserted into loop VIII of the capsid protein in recombinant adeno-associated virus vectors. These vectors carrying either gfp or fluc genes were subjected to quantitative polymerase chain reaction assays and transduction assays in human embryonic kidney 293 (HEK293T) cells to investigate the efficiency of vector production and gene delivery. In addition, the ability of a specific p5RHH-rAAV vector to deliver genes was examined through in vitro transduction of different cultured cells and in vivo tail vein administration to C57BL/6 mice. Finally, the intricate details of the vector-mediated transduction mechanisms were explored by using pharmacological inhibitors of every stage of the rAAV2 intracellular life cycle.
A total of 76 melittin-related peptides were identified from existing literature. Among them, CMA-3, p5RHH and aAR3 were found to significantly inhibit transduction of rAAV2 vector crude lysate. The p5RHH-rAAV2 vectors efficiently transduced not only rAAV-potent cell lines but also cell lines previously considered resistant to rAAV. Mechanistically, bafilomycin A1, a vacuolar endosome acidification inhibitor, completely inhibited the transgene expression mediated by the p5RHH-rAAV2 vectors. Most importantly, p5RHH-rAAV8 vectors also increased hepatic transduction in vivo in C57BL/6 mice.
The incorporation of melittin analogs into the rAAV capsids results in a significant improvement in rAAV-mediated transgene expression. While further modifications remain an area of interest, our studies have substantially broadened the pharmacological prospects of melittin in the context of viral vector-mediated gene delivery.

Sorafenib has been extensively used for the treatment of advanced hepatocellular carcinoma (HCC), and Chinese herbal medicine has also been used to manage advanced HCC. The present work evaluates the ...effectiveness and safety of Jiedu (JD) Granule, a compound of traditional Chinese herbal medicine, side-by-side with sorafenib for the treatment of advance HCC.
Patients with advanced HCC receiving treatment with JD Granule or sorafenib were enrolled from December 2014 to March 2018. The primary endpoint was overall survival (OS). The secondary endpoints were progression-free survival (PFS) and safety. Propensity score matching (PSM) analysis was used to control for possible selection bias from the study group allocation process.
Of the 325 patients included, 161 received JD Granule and 164 received sorafenib. No significant differences were found in OS or PFS among patients receiving JD Granule compared to sorafenib (P > 0.05). Median OS of the two study groups was 6.83 months (95% confidence interval CI: 5.83–9.47) in the group receiving JD Granule and 8 months (95% CI: 6.67–9.80) in the group receiving sorafenib, with half-, 1- and 2-year survival rates of 53.6%, 31.2% and 13.2% vs 60.1%, 35.5% and 14.2%, respectively. Even after PSM, the median survival time did not differ between the JD Granule group (9.03 months; 95% CI: 6.37–14.2) and the sorafenib group (7.93 months; 95% CI: 6.5–9.97), with comparable half-, 1- and 2-year survival rates. The most common adverse events (AEs) were diarrhea (13.7%) and fatigue (5.6%) in the JD Granule group, and hand-foot skin reaction (46.3%) and diarrhea (36.6%) in the sorafenib group. The JD Granule was more cost-effective than sorafenib treatment for advanced HCC.
Compared to sorafenib, JD Granule was more cost-effective and caused fewer AEs for the treatment of Chinese patients with advanced HCC.

Herein, we propose a concept on the nanopolymer that is a nanoscale polymer chain with the repeat units of nanomonomers, In ...this article, a soluble organic nanopolymer of wide bandgap semiconductors was synthesized by the Yamamoto polymerization of nanogrid monomer as the repeat units with the rectangle size of -1.7 nm × 1.2 nm. The alkyl side chain substituent at 9-position of fluorenes guarantees the polygrid with excellent solubility. Tetrafluorenes in the conjugation-interrupted backbones of polygrid acts as the active light-emitting centers without obvious green band in the fluorescence spectra of the films after 10 h annealing at 180 ℃, indicating this nanopolymer exhibits excellent spectral stability. Such soluble nanopolymers will be the fifth- generation of macromolecular materials with a potential character of overall performance improvement.
