Gold nanorods are an important kind of nanoparticles characterized by peculiar plasmonic properties. Despite their widespread use in nanotechnology, the synthetic methods for the preparation of gold ...nanorods are still not fully optimized. In this paper we describe a new, highly efficient, two-step protocol based on the use of hydroquinone as a mild reducing agent. Our approach allows the preparation of nanorods with a good control of size and aspect ratio (AR) simply by varying the amount of hexadecyl trimethylammonium bromide (CTAB) and silver ions (Ag(+)) present in the "growth solution". By using this method, it is possible to markedly reduce the amount of CTAB, an expensive and cytotoxic reagent, necessary to obtain the elongated shape. Gold nanorods with an aspect ratio of about 3 can be obtained in the presence of just 50 mM of CTAB (versus 100 mM used in the standard protocol based on the use of ascorbic acid), while shorter gold nanorods are obtained using a concentration as low as 10 mM.
At present, mammary carcinoma is the second most common type of malignant tumor in adult women after lung cancer, as more than one million women are diagnosed with breast cancer every year. Despite ...advances in diagnosis and treatment, which have resulted in a decrease in mortality in recent decades, breast cancer remains a major public health problem. One of the most significant unresolved clinical and scientific problems is the occurrence of resistance to clinical treatments and their toxicity (and how to predict, prevent and overcome them). However, the heterogeneity of human breast cancer in terms of genetic features, molecular profiles and clinical behavior represents a constraint obstructing the discovery of a solution to the disease. It is currently considered that the chances of success of therapy may increase if the tumor cells are selectively removed before they can evolve to their mature stages up to metastases production. Therefore, novel and more sensitive diagnostic tools are being developed, with the aim of improving the early and noninvasive detection of rising malignancies and the accuracy of tumor tissue localization. Meanwhile, there is an emerging use of targeted therapies in oncology, depending on the expression of specific proteins or genes present in tumor cells. Among the molecular targets considered for the treatment of breast cancer cells so far, we chose to focus on examples involving overexpression and/or gene amplification of “Human Epidermal growth factor Receptor 2” (HER2) protein. In current studies, various types of nanoparticles conjugated with the anti-HER2 monoclonal antibody, the so-called “trastuzumab”, are investigated extensively due to promising results in biological and preclinical applications aimed at improving the treatment of breast cancer. In this paper, we present a critical review of the preparation and use of different kinds of trastuzumab-functionalized nanoparticles, with an emphasis on the therapeutic and diagnostic (theranostic) potential of this generation of hybrid nanoparticles, exploiting the multifaceted mechanisms of action of trastuzumab against malignant cells.

The nanocrystals were transferred to water by exploiting the amphiphilic character of the copolymer backbone. Hydrolyzed MAPS units contributed to improve the solubility of QDs in water, whereas NAS exhibited reactivity toward biomolecules. A solution of streptavidin in phosphate buffer exhibited good dispersion ability leading to a clear and transparent colloidal suspension, indicative of good QD dispersion during phase transfer and purification. Unlike most of the published methods, the proposed functionalization approach does not require coupling agents and multistep reactions.

In this paper, a straightforward method based on elastic light scattering is shown to provide a sensitive and reliable tool for the quantitative determination of protein–ligand interactions that ...occur at the surface of suitably designed core–shell nanoparticles. The assay makes use of monodisperse nanocolloids that have minimal optical contrast with the aqueous environment. By properly coating the particles with avidin and oligo(ethylene glycol)‐based amphiphiles, we developed a hybrid system that combines the availability of standard ligands with the necessary bioinvisibility towards the accidental adsorption of nonspecific macromolecules. This probe was employed to detect interactions between different kinds of biotinylated proteins, and it revealed high specificity and affinities in the low nanomolar range. In particular, we obtained an efficient avidin anchorage of biotinylated protein A on the surface of the nanoparticles, which we exploited as a functional probe for the rapid, quantitative, picomolar detection of human IgG antibodies. Overall, these light‐scattering‐based nanosensors appear as a simple and highly informative tool for proteomics studies.
Invisible sensors! Elastic light scattering enables the quantitative determination of protein–ligand binding that occurs at the surface of suitably designed nanoparticles with a core–shell morphology. The core material is an index‐matched perfluoropolymer, which has been surface‐functionalized with a few native avidin molecules embedded in an oligo(ethylene glycol) amphiphile monolayer. The resulting nanosensor was used to detect protein–ligand and protein–antibody interactions.

Highly fluorinated materials are being widely investigated due to a number of peculiar properties, which are potentially useful for various applications, including use as lubricants, anti‐adhesive ...films, and substitutes for biological fluids for biomedical utilization. However, at present such potential is still poorly exploited. One of the major drawbacks that hampers the rapid development of nanoscale fluoro‐hybrid devices is the remarkable inertness of perfluoropolymeric materials that lack reactive functionalities, as they do not offer any functional groups that can be employed to covalently anchor organic molecules on their surface. In this paper, a convenient method for the stable biofunctionalization of strongly unreactive perfluoropolymer nanoparticles (PnPs) is reported. PnPs are easily coated with newly synthesized asymmetric diacetylenic monomer compounds (ADMs), thanks to PnP’s high propensity to interact with hydrophobic moieties. Once monomerically adsorbed onto PnPs, such suitably designed ADMs enable the formation of a robust polymeric shell around the perfluoroelastomer core via a clean UV‐promoted localized photopolymerization. Given the peculiar optical characteristics of PnPs, the coating of the particles can be monitored step by step using light scattering, which also allows estimation of the fraction of reacted monomers by competitive adsorption with smaller particles. The potential of this method for the biofunctionalization of PnPs is demonstrated with representative proteins and carbohydrates. Among them, the extension to avidin–biotin technology may broaden the scope and applicability of this strategy to potentially a large number of molecules of biomedical interest.
Making the unreactive reactive: A smart method for the biofunctionalization of strongly inert perfluoropolymer nanoparticles (PnPs) is presented, using a stable coating with novel diacetylenic compounds followed by clean UV photopolymerization to generate reactive functionalities on the PnP surface. This method further allows fine tuning of the amount of conjugated biomolecules, which can be sensitively and straightforwardly quantified.

A new light‐scattering‐based method to detect molecular interactions at the surface of low‐refractive‐index nanoparticles was recently proposed. Water‐dispersed nanoparticles functionalized with ...receptors typical of immature bacteria cell walls were used to study the activity of the antibiotic vancomycin. This method subtly depends on the specific properties of the nanoparticles. Here we discuss, by comparative experiments and through theoretical evaluation, the effects of size, refractive index, electric charge, and dilution on the reliability and accuracy of the method. Quite surprisingly, perfect index matching and minimal size (i.e., maximum surface), which is almost attained in one of the colloids here employed, do not represent the ideal conditions. Rather, we show that a nanoparticle radius of 100 nm and a refractive index slightly below that of water yields the best signal/background amplitude. We also show that repulsive interactions can lead to artifacts in the adsorption isotherm, thus indicating that electrostatic stabilization should be kept at a minimum. The close agreement between the interaction strengths, as measured with two different nanoparticle systems, testifies to the reliability of the method.

Hydrophobic, transparent, electrically stabilized nanospheres (20–100 nm) with a refractive index lower than water were used to study the interaction of a glycopeptide antibiotic with its bacterial wall receptor.

We present here the surface-enhanced Raman spectroscopic (SERS)-based detection of the Wilm’s tumor gene (WT1) sequence using dye-labeled reporter oligonucleotide and magnetic core @ gold shell ...nanoparticles. Thiolated single-stranded DNA (ssDNA) complementers of the WT1 sequence were used to functionalize the gold shell with capture oligonucleotides in a facile and fast two-step method. The signal amplification performance of the core @ shell colloidal SERS substrate was tested using malachite green as label dye. The Raman signal enhancing efficacy of the magnetic core @ gold shell nanomaterial was compared with the efficacy of spherical gold particles produced using the conventional citrate reduction method. The core @ shell particles were found to be superior both regarding robustness in SERS and facile separation in a heterogeneous reaction system. The core @ shell particles functionalized with target specific oligonucleotide were able to capture the WT1 target and worked as Raman signal amplifiers in our assay system. The good physicochemical characteristics of these particles and the sensitivity observed in SERS experiments allow us to expect good performance in the further development steps of a novel, fast and reliable spectroscopic method for WT1 detection in minimal residual disease patients.
