A fundamental principle in biology is that the program for early development is established during oogenesis in the form of the maternal transcriptome. How the maternal transcriptome acquires the ...appropriate content and dosage of transcripts is not fully understood. Here we show that 3' terminal uridylation of mRNA mediated by TUT4 and TUT7 sculpts the mouse maternal transcriptome by eliminating transcripts during oocyte growth. Uridylation mediated by TUT4 and TUT7 is essential for both oocyte maturation and fertility. In comparison to somatic cells, the oocyte transcriptome has a shorter poly(A) tail and a higher relative proportion of terminal oligo-uridylation. Deletion of TUT4 and TUT7 leads to the accumulation of a cohort of transcripts with a high frequency of very short poly(A) tails, and a loss of 3' oligo-uridylation. By contrast, deficiency of TUT4 and TUT7 does not alter gene expression in a variety of somatic cells. In summary, we show that poly(A) tail length and 3' terminal uridylation have essential and specific functions in shaping a functional maternal transcriptome.

Growing evidence has shown that brain activity at rest slowly wanders through a repertoire of different states, where whole-brain functional connectivity (FC) temporarily settles into distinct FC ...patterns. Nevertheless, the functional role of resting-state activity remains unclear. Here, we investigate how the switching behavior of resting-state FC relates with cognitive performance in healthy older adults. We analyse resting-state fMRI data from 98 healthy adults previously categorized as being among the best or among the worst performers in a cohort study of >1000 subjects aged 50+ who underwent neuropsychological assessment. We use a novel approach focusing on the dominant FC pattern captured by the leading eigenvector of dynamic FC matrices. Recurrent FC patterns - or states - are detected and characterized in terms of lifetime, probability of occurrence and switching profiles. We find that poorer cognitive performance is associated with weaker FC temporal similarity together with altered switching between FC states. These results provide new evidence linking the switching dynamics of FC during rest with cognitive performance in later life, reinforcing the functional role of resting-state activity for effective cognitive processing.

Structural health monitoring (SHM) on railway tunnels is increasingly demanding inspection methodologies based on full‐field image‐based techniques. The relevance of adequate inspection ...methodologies to signal potential problems that need increased attention is at the forefront of any monitoring system for problems that can disturb tunnel's integrity. Detecting defects is decisive for an efficient inspection, investigating tunnel status, and rescheduling maintenance operations, when and where required. This paper introduces a methodology design, based on acquisition of tunnel's 3D geometry and monitoring it through time to signal the possible emergence of structural defects and return the exact position where monitoring equipment will have to be deployed to. A demonstrator is built, and the applicability and accuracy of the system is assessed through implementation of a model prototype, carried out on a scaled tunnel model. At a secondary stage inspection, imposed geometrical changes and defects of the tunnel are monitored and characterised by an optical nondestructive inspection technique based on the digital image correlation (DIC). It enables the appraisal of structural integrity on the tunnel lining through displacement and strain fields acquisition. The chief advantage of the deployed inspection system, amongst others, lies on its robust functionality in both geometrical and structural integrity inspection aspects, contributing to the establishment of a robust SHM methodology for railway tunnels over time. In conclusion, experimental results are presented for evaluating scaled tunnel condition and monitoring the tunnel profile, and further DIC data are presented inferring displacement field progress of an introduced structural defect.

Obesity has been linked to autonomic dysfunction, which is thought to be one of the main contributors for hypertension, cardiac remodelling and death. Exercise and diet-based weight loss are the ...mainstay therapy for obesity, but there is a paucity of data regarding the effect of weight changes in autonomic nervous system (ANS) activity.
To describe the impact of weight changes in autonomic nervous system.
A systematic literature search of four biomedical databases was performed evaluating effects of weight changes, thorough diet and/or exercise-based interventions, in the following ANS outcomes: heart rate variability, namely low frequency (LF)/high frequency (HF) ratio (LF/HF ratio), normalized units of LF (LFnu) and HF (HFnu), muscle sympathetic nerve activity (MSNA), noradrenaline spillover rate (NA-SR), standard deviation of normal-to-normal intervals (SDNN), root mean square of successive differences (RMSSD), baroreflex sensitivity and pupillometry. Quality appraisal was performed using the GRADE methodology and, where fitting, studies with comparable outcomes were pooled for meta-analysis.
Twenty-seven studies - 7 controlled clinical trials and 20 observational studies - were included. Weight gain was reported in 4 studies and weight loss in all the other studies. Interventions inducing weight changes included: hypocaloric or hypercaloric diets, exercise (strength, endurance or aerobic training) and hypocaloric diet coupled with exercise programs. Most studies which resulted in weight loss reported decreases in LF/HF ratio, LFnu, MSNA burst frequency and incidence, NA-SR, and an increase of baroreflex sensitivity, HF, HFnu and RMSSD, pointing to a parasympathetic nervous system activation. Meta-analysis regarding weight loss interventions showed a significant pooled effect size (95% CI) with a decreased of MSNA burst frequency −5.09 (−8.42, −1.75), MSNA incidence −6.66 (−12.40, −0.62), however this was not significant for SDNN 14.32 (−4.31, 32.96). Weight gain was associated with an increase in LF/HF, LFnu, MSNA burst frequency and incidence. The weight loss effects were potentiated by the association of hypocaloric diet with exercise. Nevertheless, weight changes effects in these outcomes were based in low or very low quality of evidence.
Diet and exercise based weight loss appears to increase parasympathetic and decrease sympathetic activity, the opposing effects being observed with weight gain. These findings are not uniformly reported in the literature, possibly due to differences in study design, methodology, characteristics of the participants and techniques used to estimate autonomic nervous activity.

As Missões Culturais do Secretariado de Propaganda Nacional (SPN) constituíram uma importante iniciativa no início dos anos 40 e tinham como objetivo a realização de vários concertos musicais, com ...uma palestra temática e recitação de poesia, em várias cidades e vilas de Portugal. O presente artigo procura abordar as missões culturais na sua dimensão política, na ação do SPN de chegar às classes médias dos pequenos centros urbanos, e qual o papel desempenhado pelo pianista e compositor Eurico Tomás de Lima enquanto chefe daquelas missões entre 1940 e 1941. A pesquisa permitiu, a partir de diferentes fontes documentais, perceber: a organização e orgânica das Missões Culturais, contribuindo para um conhecimento mais profundo no âmbito dos objetivos propagandísticos do Estado Novo; o papel central desempenhado por Eurico Tomás de Lima, que tinha várias responsabilidades na implementação e execução desta iniciativa; os intuitos artísticos e musicais dos programas escolhidos.
Over the last decades, several compliant contact force models have been proposed. However, no complete and systematic comparison has been done on these models, which provides information on their ...range of application and accuracy for use in different contact scenarios. Thus, the selection of an appropriate model for a given contact problem is still an important and challenging issue to be addressed. The Hertzian contact theory remains the foundation for almost all of the available force models, but by itself, it is not appropriate for most impacts in practice, due to the amount of energy dissipated during the impact. A good number of contact force models have been offered that augment the Hertzian law with a damping term to accommodate the energy loss during the impact process for small or moderate impact velocities. In this work, the main issues associated with the most common compliant contact force models of this type are analyzed. Results in terms of the dynamic simulations of multibody systems are presented, which allow for the comparison of the similarities and differences among the models considered.

Epoxy resins are one of the most widely used thermosets in different engineering fields, due to their chemical resistance and thermo-mechanical properties. Recently, bio-based thermoset resin systems ...have attracted significant attention given their environmental benefits related to the wide variety of available natural resources, as well as the resulting reduction in the use of petroleum feedstocks. During the last two decades, considerable improvement on the properties of bio-sourced resins has been achieved to obtain performances comparable to petroleum-based systems. This paper reviews recent advances on new bio-based epoxy resins, derived from natural oils, natural polyphenols, saccharides, natural rubber and rosin. Particular focus has been given to novel chemical formulations and resulting mechanical properties of natural derived- epoxies, curing agents or entire systems, constituting an interesting alternative for a large variety of engineering applications, including the aviation sector. The present work is within the scope of the ECO-COMPASS project, where new bio-sourced epoxy matrixes for green composites are under investigation.

Cadmium is a toxic heavy metal that has been implicated in breast cancer etiology, albeit with inconsistent results.
To investigate the shape of the relation between cadmium exposure and breast cancer incidence and mortality in cohort studies.
Following a literature search through April 14, 2020, we carried out a systematic review and dose-response meta-analysis to investigate the shape of the relation between cadmium exposure (assessed either through diet or urine excretion) and disease incidence and mortality.
For inclusion, a study had to report incidence or mortality for breast cancer according to baseline cadmium exposure category; be a prospective cohort, case-cohort or nested case-control study with a minimum one-year follow-up, and reporting effect estimates for all exposure categories.
Studies were evaluated using the ROBINS-E risk of bias tool. The effects in humans were assessed quantitatively using one-stage dose-response meta-analysis in a random effects meta-analytical model.
We identified 10 studies eligible for inclusion in the dose-response meta-analysis, six based on cadmium dietary intake, and four on urinary excretion levels. We found a marginal and imprecise positive relation between dietary cadmium intake and breast cancer, and no association when urinary cadmium excretion was used for exposure assessment. Compared to no exposure, at 20 µg/day of cadmium intake the summary risk ratio was 1.12 (95% confidence interval 0.80–1.56), while at 2 µg/g creatinine of cadmium excretion the summary risk ratio was 0.89 (95% confidence interval 0.38–2.14). Analysis restricted to post-menopausal women showed no association between either dietary or urinary cadmium and subsequent breast cancer incidence and mortality.
Overall, we found scant evidence of a positive association between cadmium and breast cancer. Available data were too limited to carry out stratified analyses according to age, smoking and hormone receptor status. Therefore, possible associations between cadmium exposure and breast cancer in selected subgroups cannot be entirely ruled out.
