In this paper, we will review both past and recent progresses in the generation, detection and application of intense terahertz (THz) radiation. We will restrict the review to laser based intense ...few-cycle THz sources, and thus will not include sources such as synchrotron-based or narrowband sources. We will first review the various methods used for generating intense THz radiation, including photoconductive antennas (PCAs), optical rectification sources (especially the tilted-pulse-front lithium niobate source and the DAST source, but also those using other crystals), air plasma THz sources and relativistic laser-plasma sources. Next, we will give a brief introduction on the common methods for coherent THz detection techniques (namely the PCA technique and the electro-optic sampling), and point out the limitations of these techniques for measuring intense THz radiation. We will then review three techniques that are highly suited for detecting intense THz radiation, namely the air breakdown coherent detection technique, various single-shot THz detection techniques, and the spectral-domain interferometry technique. Finally, we will give an overview of the various applications that have been made possible with such intense THz sources, including nonlinear THz spectroscopy of condensed matter (optical-pump THz-probe, THz-pump THz-probe, THz-pump optical-probe), nonlinear THz optics, resonant and non-resonant control of material (such as switching of superconductivity, magnetic and polarization switching) and controlling the nonlinear response of metamaterials. We will also provide a short perspective on the future of intense THz sources and their applications.
In high-order harmonic generation, resonant harmonics (RH) are sources of intense, coherent extreme-ultraviolet radiation. However, intensity enhancement of RH only occurs for a single harmonic ...order, making it challenging to generate short attosecond pulses. Moreover, the mechanism involved behind such RH was circumstantial, because of the lack of direct experimental proofs. Here, we demonstrate the exact quantum paths that electron follows for RH generation using tin, showing that it involves not only the autoionizing state, but also a harmonic generation from dressed-AIS that appears as two coherent satellite harmonics at frequencies ±2Ω from the RH (Ω represents laser frequency). Our observations of harmonic emission from dressed states open the possibilities of generating intense and broadband attosecond pulses, thus contributing to future applications in attosecond science, as well as the perspective of studying the femtosecond and attosecond dynamics of autoionizing states.
High harmonics from laser-ablated plumes are mostly generated from ionic species. We demonstrate that with ultrashort infrared (∼1.82 μm) driving lasers, high harmonics from laser-ablated manganese ...are predominantly generated from neutral atoms, a transition metal atom with an ionization potential of 7.4 eV. Our results open the possibility to advance laser-ablation technique to study the dynamics of neutral atoms of low ionization potential. Moreover, as manganese contains giant autoionizing resonance, intense and broadband high harmonics have been demonstrated from this resonance at energies from 49 to 53 eV. This opens the possibility to generate intense attosecond pulses directly from the giant resonances, as well as to study these resonances using high-harmonic spectroscopy.

Perturbative optical nonlinearities induced by static electric fields1 have proven useful in visualizing dynamical function in systems including operating circuits2,3, electric and magnetic domain ...walls4, and biological matter5, and in controlling light for applications in silicon photonics6. Here, we extend field-induced second-harmonic generation to the non-perturbative regime. We demonstrate that static or transient fields up to terahertz (THz) frequencies applied to silicon and ZnO crystals generate even-order high harmonics. Images of the even harmonics confirm that static fields delivered with conventional electronics control the spatial properties of the high-harmonic emission. Extending our methodology to higher-harmonic photon energies7,8 paves the way for realizing active optics in the extreme ultraviolet and will allow imaging of operating electronic circuits9, of Si-photonic devices10 and of other functional materials11,12, with higher spatio-temporal resolution than perturbative methods. For THz spectroscopy, our method has the bandwidth to allow measurement of attosecond transients imprinted on THz waveforms.

The characteristics of terahertz (THz) radiation generated from large-aperture photoconductive antennas (LAPCAs) were investigated. The antennas were fabricated using different wide-bandgap ...semiconductor crystals (ZnSe, GaN, 6H-SiC, 4H-SiC and β-Ga2O3) as the substrate. We used an amplified sub-picosecond KrF excimer laser for illumination of the LAPCAs. THz emission scaling was studied as a function of the bias field and the pump laser energy. It was found that the radiated THz energy scales quadratically as a function of the bias field and sub-linearly as a function of the optical fluence for most of the substrates. Further, we demonstrate that SiC, and especially 4H-SiC LAPCAs offer the best THz generation performances. In order to generate intense THz radiation, we fabricated both 6H- and 4H-SiC LAPCAs with an interdigitated structure. From the field autocorrelation trace, it was found that the spectra lie in the sub-THz regime, extending up to 400 GHz, with a peak frequency at 50 GHz, making the bridge between the microwaves band and the THz band. The maximum generated THz energy was 11 J, which is to date the highest THz energy measured from LAPCA sources, with a corresponding peak electric field of 115 kV cm−1 and a corresponding ponderomotive potential of 60 eV. Nonlinear THz experiments were performed using these energetic THz pulses, and open aperture Z-scan experiments in an n-doped InGaAs layer revealed a transmission enhancement of 1.7. It was also shown that in order to have efficient THz generation, the energy contrast of the laser must be kept high.
Summary Objective To investigate the mechanism of mechanical stress-induced expression and regulation of aggrecanases and examine the role of runt-related transcription factor 2 (RUNX-2) in ...chondrocyte-like cells. Methods SW1353 cells were seeded onto stretch chambers at a concentration of 5 × 104 cells/chamber, and a uni-axial cyclic tensile strain (CTS) (0.5 Hz, 10% stretch) was applied for 30 min. Total RNA was extracted, reverse transcribed, and analyzed by polymerase chain reaction (PCR) and real-time PCR. RUNX-2 overexpression and small interfering RNA (siRNA) targeting RUNX-2 were used to investigate the role of RUNX-2 in CTS-induced gene expression. The involvement of diverse mitogen-activated protein kinase (MAPK) pathways in the activation of RUNX-2, MMP-13 and ADAMTS-5 during CTS was examined by Western blotting. Results CTS induced expression of RUNX-2 , MMP-13 , ADAMTS-4 , -5 , and -9 . Overexpression of RUNX-2 up-regulated expression of MMP-13 and ADAMTS-5 , whereas RUNX-2 siRNA resulted in significant down-regulation of mechanically-induced MMP-13 and ADAMTS-5 expression. CTS induced activation of p38 MAPK, and CTS induction of RUNX-2 , MMP-13 and ADAMTS-5 mRNA was down-regulated by the selective p38 MAPK inhibitor SB203580 but not by the p44/42 MAPK inhibitor U0126, or the JNK MAPK inhibitor JNK inhibitor II. Conclusions RUNX-2 might have a role as a key downstream mediator of p38’s ability to regulate mechanical stress-induced MMP-13 and ADAMTS-5 expression.

Methods Human chondrocytes were seeded in stretch chambers at a concentration of 5 × 104 cells/chamber. Cells were pre-incubated with or without HDAC inhibitors (MS-275 or trichostatin A; TSA) for 12 h, followed by uniaxial cyclic tensile strain (CTS) (0.5 Hz, 10% elongation), which was applied for 30 min using the ST-140-10 system (STREX, Osaka, Japan). Total RNA was extracted and the expression of RUNX-2, ADAMTS-5, matrix metalloproteinase (MMP)-3, and MMP-13 at the mRNA and protein levels were examined by real-time polymerase chain reaction (PCR) and immunocytochemistry, respectively. The activation of diverse mitogen-activated protein kinase (MAPK) pathways with or without HDAC inhibitors during CTS was examined by western blotting. Results HDAC inhibitors (TSA: 10 nM, MS-275: 100 nM) suppressed CTS-induced expression of RUNX-2, ADAMTS-5, and MMP-3 at both the mRNA and protein levels within 1 h. CTS-induced activation of p38 MAPK (p38), extracellular signal-regulated kinase (ERK), and c-Jun N-terminal kinase (JNK) MAPKs was downregulated by both HDAC inhibitors. Conclusion The CTS-induced expression of RUNX-2 and ADAMTS-5 was suppressed by HDAC inhibitors via the inhibition of the MAPK pathway activation in human chondrocytes. The results of the current study suggested a novel therapeutic role for HDAC inhibitors against degenerative joint disease such as osteoarthritis.
