Data on breakthrough SARS-CoV-2 Delta variant infections in vaccinated individuals are limited.
We studied breakthrough infections among Oxford-AstraZeneca vaccinated healthcare workers in an ...infectious diseases hospital in Vietnam. We collected demographic and clinical data alongside serial PCR testing, measurement of SARS-CoV-2 antibodies, and viral whole-genome sequencing.
Between 11th–25th June 2021 (7-8 weeks after the second dose), 69 staff tested positive for SARS-CoV-2. 62 participated in the study. Most were asymptomatic or mildly symptomatic and all recovered. Twenty-two complete-genome sequences were obtained; all were Delta variant and were phylogenetically distinct from contemporary viruses obtained from the community or from hospital patients admitted prior to the outbreak. Viral loads inferred from Ct values were 251 times higher than in cases infected with the original strain in March/April 2020. Median time from diagnosis to negative PCR was 21 days (range 8–33). Neutralizing antibodies (expressed as percentage of inhibition) measured after the second vaccine dose, or at diagnosis, were lower in cases than in uninfected, fully vaccinated controls (median (IQR): 69.4 (50.7-89.1) vs. 91.3 (79.6-94.9), p=0.005 and 59.4 (32.5-73.1) vs. 91.1 (77.3-94.2), p=0.043). There was no correlation between vaccine-induced neutralizing antibody levels and peak viral loads or the development of symptoms.
Breakthrough Delta variant infections following Oxford-AstraZeneca vaccination may cause asymptomatic or mild disease, but are associated with high viral loads, prolonged PCR positivity and low levels of vaccine-induced neutralizing antibodies. Epidemiological and sequence data suggested ongoing transmission had occurred between fully vaccinated individuals.

The insect bacterium Wolbachia pipientis is being introgressed into Aedes aegypti populations as an intervention against the transmission of medically important arboviruses. Here we compare Ae. ...aegypti mosquitoes infected with wMelCS or wAlbB to the widely used wMel Wolbachia strain on an Australian nuclear genetic background for their susceptibility to infection by dengue virus (DENV) genotypes spanning all four serotypes. All Wolbachia-infected mosquitoes were more resistant to intrathoracic DENV challenge than their wildtype counterparts. Blocking of DENV replication was greatest by wMelCS. Conversely, wAlbB-infected mosquitoes were more susceptible to whole body infection than wMel and wMelCS. We extended these findings via mosquito oral feeding experiments, using viremic blood from 36 acute, hospitalised dengue cases in Vietnam, additionally including wMel and wildtype mosquitoes on a Vietnamese nuclear genetic background. As above, wAlbB was less effective at blocking DENV replication in the abdomen compared to wMel and wMelCS. The transmission potential of all Wolbachia-infected mosquito lines (measured by the presence/absence of infectious DENV in mosquito saliva) after 14 days, was significantly reduced compared to their wildtype counterparts, and lowest for wMelCS and wAlbB. These data support the use of wAlbB and wMelCS strains for introgression field trials and the biocontrol of DENV transmission. Furthermore, despite observing significant differences in transmission potential between wildtype mosquitoes from Australia and Vietnam, no difference was observed between wMel-infected mosquitoes from each background suggesting that Wolbachia may override any underlying variation in DENV transmission potential.

In this study, mixed culture (microalgae:activated sludge) of a photobioreactor (PBR) were investigated at different inoculation ratios (1:0, 9:1, 3:1, 1:1, 0:1 wt/wt). This work was not only to determine the optimal ratio for pollutant remediation and biomass production but also to explore the role of microorganisms in the co-culture system. The results showed high total biomass concentrations were obtained from 1:0 and 3:1 ratio being values of 1.06, 1.12 g L-1, respectively. Microalgae played a dominant role in nitrogen removal via biological assimilation while activated sludge was responsible for improving COD removal. Compared with the single culture of microalgae, the symbiosis between microalgae and bacteria occurred at 3:1 and 1:1 ratio facilitated a higher COD removal by 37.5–45.7 %. In general, combined assessment based on treatment performance and biomass productivity facilitated to select an optimal ratio of 3:1 for the operation of the co-culture PBR.

Addition of flow fields to carbon paper electrodes in a vanadium redox flow battery (VRFB) can improve the peak power density through uniform distribution of electrolyte in the electrodes. However, ...it is unclear whether flow fields have a similar effect with graphite felt electrodes, as VRFBs with felt electrodes reported in literature show a large anomaly in obtained power density. In this work, we evaluate three flow fields; viz. serpentine, interdigitated and conventional (without flow pattern) type with felt electrodes and compare their performance with a serpentine flow field using carbon paper electrodes under identical experimental conditions. The conventional flow field provides highest energy efficiency (75%) followed by serpentine (64%) and interdigitated (55%) at 0.2 A cm−2 attributable to the deteriorating electrolyte distribution in the electrodes. Computation fluid dynamic simulations confirm the experimental finding of worsening electrolyte distribution (conventional < serpentine < interdigitated). A power density of 0.51 W cm−2 at 60 mL min−1 flow rate is obtained for serpentine and conventional flow fields with felt electrodes; comparable to the highest power density reported in literature for high performing zero-gap flow field architecture. This paper gives comprehensive insights on flow fields for VRFBs that can be extended to other flow batteries.

The wMel strain of Wolbachia can reduce the permissiveness of Aedes aegypti mosquitoes to disseminated arboviral infections. Here, we report that wMel-infected Ae. aegypti (Ho Chi Minh City ...background), when directly blood-fed on 141 viremic dengue patients, have lower dengue virus (DENV) transmission potential and have a longer extrinsic incubation period than their wild-type counterparts. The wMel-infected mosquitoes that are field-reared have even greater relative resistance to DENV infection when fed on patient-derived viremic blood meals. This is explained by an increased susceptibility of field-reared wild-type mosquitoes to infection than laboratory-reared counterparts. Collectively, these field- and clinically relevant findings support the continued careful field-testing of wMel introgression for the biocontrol of Ae. aegypti-born arboviruses.

In this study, ZnO‐doped carbon aerogel is synthesized from cellulose in coconut peat with sodium alginate as a binder via both freeze‐drying and pyrolysis processes. Of those, zinc nitrate is used ...as not only a crosslinking agent to form gel but also a precursor source to dope ZnO in the carbon aerogel matrix. The effect of precursor ratios on the characterization and energy storage capacity of composite aerogel is investigated. As a result, it is indicated that the formed ZnO‐doped carbon aerogel possesses a highly porous structure which is typical for aerogel structure shown by SEM images, density, and porosity. Besides, via the XRD patterns, the confirmation of the ZnO crystal structure is found within the carbon aerogel lattice. In terms of energy storage, based on the specific capacitance results, the ZCA‐4 sample with a sodium alginate and cellulose weight ratio of 1:20 shows the best energy storage with a specific capacitance of 105 F/g in the voltage range of 0–0.5 V and scan‐rate speed of 0.005 V. Therewithal, the ZCA‐4 also performs high durability, high scanning speed tolerance, and stable storage performance with efficiency reaching more than 99% after 500 consecutive scan cycles. These results demonstrate that the ZnO‐doped carbon aerogel has potential applications as electrode materials in supercapacitors.

Dengue shock (DS) is the most severe complication of dengue infection; endothelial hyperpermeability leads to profound plasma leakage, hypovolaemia and extravascular fluid accumulation. At present, ...the only treatment is supportive with intravenous fluid, but targeted endothelial stabilising therapies and host immune modulators are needed. With the aim of prioritising potential therapeutics, we conducted a prospective observational study of adults (≥16 years) with DS in Vietnam from 2019-2022, comparing the pathophysiology underlying circulatory failure with patients with septic shock (SS), and investigating the association of biomarkers with clinical severity (SOFA score, ICU admission, mortality) and pulmonary vascular leak (daily lung ultrasound for interstitial and pleural fluid). Plasma was collected at enrolment, 48 hours later and hospital discharge. We measured biomarkers of inflammation (IL-6, ferritin), endothelial activation (Ang-1, Ang-2, sTie-2, VCAM-1) and endothelial glycocalyx breakdown (hyaluronan, heparan sulfate, endocan, syndecan-1). We enrolled 135 patients with DS (median age 26, median SOFA score 7, 34 required ICU admission, 5 deaths), together with 37 patients with SS and 25 healthy controls. Within the DS group, IL-6 and ferritin were associated with admission SOFA score (IL-6: βeta0.70, p<0.001 & ferritin: βeta0.45, p<0.001), ICU admission (IL-6: OR 2.6, p<0.001 & ferritin: OR 1.55, p<0.001) and mortality (IL-6: OR 4.49, p = 0.005 & ferritin: OR 13.8, p = 0.02); both biomarkers discriminated survivors and non-survivors at 48 hours and all patients who died from DS had pre-mortem ferritin ≥100,000ng/ml. IL-6 most strongly correlated with severity of pulmonary vascular leakage (R = 0.41, p<0.001). Ang-2 correlated with pulmonary vascular leak (R = 0.33, p<0.001) and associated with SOFA score (β 0.81, p<0.001) and mortality (OR 8.06, p = 0.002). Ang-1 was associated with ICU admission (OR 1.6, p = 0.005) and mortality (OR 3.62, p = 0.006). All 4 glycocalyx biomarkers were positively associated with SOFA score, but only syndecan-1 was associated with ICU admission (OR 2.02, p<0.001) and mortality (OR 6.51, p<0.001). This study highlights the central role of hyperinflammation in determining outcomes from DS; the data suggest that anti-IL-1 and anti-IL-6 immune modulators and Tie2 agonists may be considered as candidates for therapeutic trials in severe dengue.

In this study, the zinc oxide-decorated graphitic carbon nitride composite (ZnO/gC3N4 – ZCN) was synthesized for photocatalytic degradation of organic dyes in aqueous media. Particularly, ZnO was ...fabricated by the calcination method with the assistance of Garcinia mangostana pericarp extract as a stabilizer and reducing agent, while graphitic carbon nitride (gC3N4 – CN) was prepared by the calcination method and exfoliated using a strong oxidizer mixture. Characterization results showed ZnO possesses a spherical shape with an average diameter of 30–80 nm and is distributed evenly on the CN sheets. Subsequently, ZCN was subjected to photodegradation towards methylene blue (MB), which indicated that under optimal conditions of 50 mg of catalyst, pH 9, and 10 mg/L of MB concentration, the overall efficiency reached up to 99.16 % after 180 min of visible irradiation. Additionally, ZCN also possesses the degradation ability for other organic dyes, for instance, malachite green (MG) and methyl orange (MO) with an overall efficiency of 96.42 and 57.57 %, respectively. Moreover, the radical scavenging experiments elucidated that the major responsible component for the photodegradation was the photogenerated holes, while other radicals also facilitated the process. Besides, the reusability of the material was excellent as only 10 % of the yield decreased after five cycles. The aforementioned findings reveal the notable potential applications of the ZCN material in the environmental remedial field.
