The experimental infection of a mouse lung with influenza A virus has proven to be an invaluable model for studying the mechanisms of viral adaptation and virulence. The mouse adaption of human ...influenza A virus can result in mutations in the HA and other proteins, which is associated with increased virulence in mouse lungs. In this study, a mouse-adapted seasonal H1N1 virus was obtained through serial lung-to-lung passages and had significantly increased virulence and pathogenicity in mice. Genetic analysis indicated that the increased virulence of the mouse-adapted virus was attributed to incremental acquisition of three mutations in the HA protein (T89I, N125T, and D221G). However, the mouse adaption of influenza A virus did not change the specificity and affinity of receptor binding and the pH-dependent membrane fusion of HA, as well as the in vitro replication in MDCK cells. Notably, infection with the mouse adapted virus induced severe lymphopenia and modulated cytokine and chemokine responses in mice. Apparently, mouse adaption of human influenza A virus may change the ability to replicate in mouse lungs, which induces strong immune responses and inflammation in mice. Therefore, our findings may provide new insights into understanding the mechanisms underlying the mouse adaption and pathogenicity of highly virulent influenza viruses.

The novel pandemic A (H1N1) virus was first identified in Mexico in April 2009 and quickly spread worldwide. Like all influenzas, the H1N1 strain-specific properties of replication, virulence, and ...pathogenicity are a result of the particular genomic sequence and concerted expression of multiple genes. Thus, specific mutations may support increased virulence and may be useful as biomarkers of potential threat to human health. We performed comparative genomic analysis of ten strains of the 2009 pandemic A (H1N1) influenza viruses to determine whether genotypes associated with clinical phenotypes, which ranged from mild to severe illness and up to lethal. Virus replication capacity was tested for each strain in vitro using cultured epithelial cells, while virulence and pathogenicity were investigated in vivo using the BALB/c mouse model. The results indicated that A/Sichuan/1/2009 strain had significantly higher replication ability and virulence than the other strains, and five unique non-synonymous mutations were identified in important gene-encoding sequences. These mutations led to amino acid substitutions in HA (L32I), PA (A343T), PB1 (K353R and T566A), and PB2 (T471M), and may be critical molecular determinants for replication, virulence, and pathogenicity. Our results suggested that the replication capacity in vitro and virulence in vivo of the 2009 pandemic A (H1N1) viruses were not associated with the clinical phenotypes. This study offers new insights into the transmission and evolution of the 2009 pandemic A (H1N1) virus.

The integration of distributed energy resources (DERs) has revolutionized the power distribution grids, transitioning them into a decentralized mode. While DERs bring numerous advantages, they also ...introduce the complexity and uncertainties to the distribution system operation, necessitating adaptable operational strategies to enhance system resilience. This paper presents a pioneering approach that exploits the load and DERs in outage managements to enhance the distribution grid restoration. The proposed network equivalent model aggregates DERs and loads, preserving controllability information for effective real-time system outage management and restoration. A data-driven load model that combines an artificial neural network-based short-term load forecasting model and a delay exponential cold load pickup (CLPU) model is developed to compute the reliable safety margin for restoration and pick up the maximum number of loads during the restoration. Furthermore, an optimal restoration strategy is presented, encompassing restored network topology, controllable utility loads, and DERs set-point determination.
Stroke imposes a substantial burden worldwide. With the rapid economic and lifestyle transition in China, trends of the prevalence of stroke across different geographic regions in China remain ...largely unknown. Capitalizing on the data in the National Health Services Surveys (NHSS), we assessed the prevalence and risk factors of stroke in China from 2003 to 2018. In this study, data from 2003, 2008, 2013, and 2018 NHSS were collected. Stroke cases were based on participants’ self-report of a previous diagnosis by clinicians. We estimated the trends of stroke prevalence for the overall population and subgroups by age, sex, and socioeconomic factors, then compared across different geographic regions. We applied multivariable logistic regression to assess associations between stroke and risk factors. The number of participants aged 15 years or older were 154,077, 146,231, 230,067, and 212,318 in 2003, 2008, 2013, and 2018, respectively, among whom, 1435, 1996, 3781, and 6069 were stroke patients. The age and sex standardized prevalence per 100,000 individuals was 879 in 2003, 1100 in 2008, 1098 in 2013, and 1613 in 2018. Prevalence per 100,000 individuals in rural areas increased from 669 in 2003 to 1898 in 2018, while urban areas had a stable trend from 1261 in 2003 to 1365 in 2018. Across geographic regions, the central region consistently had the highest prevalence, but the western region has an alarmingly increasing trend from 623/100,000 in 2003 to 1898/100,000 in 2018 (Ptrend<0.001), surpassing the eastern region in 2013. Advanced age, male sex, rural area, central region, hypertension, diabetes, depression, low education and income level, retirement or unemployment, excessive physical activity, and unimproved sanitation facilities were significantly associated with stroke. In conclusion, the increasing prevalence of stroke in China was primarily driven by economically underdeveloped regions. It is important to develop targeted prevention programs in underdeveloped regions. Besides traditional risk factors, more attention should be paid to nontraditional risk factors to improve the prevention of stroke.
The aim of this study was to determine the complete mitochondrial genome (mitogenome) and the phylogenetic location of the Palaemonetes sinensis. The mitogenome was 15,736 bp in length, containing 22 ...transfer RNA genes (tRNAs), 13 protein-coding genes (PCGs), 2 ribosome RNA genes (rRNAs), and a control region (CR). The overall nucleotide composition is as follows: A, 35.69%; C, 21.66%; G, 12.39%; T, 30.26%. Nine and four PCGs were encoded on the heavy and light strands, respectively. Phylogenetic analysis suggested that P. sinensis shares a close relationship with Palaemon serenus and Palaemon capensis. These findings are helpful for better understanding the phylogenetic relationship among Caridea, as well as investigating the population genetics of P. sinensis in the future.
Understanding maturation pathways of broadly neutralizing antibodies (bnAbs) against HIV‐1 can be highly informative for HIV‐1 vaccine development. A lineage of J038 bnAbs is now obtained from a ...long‐term SHIV‐infected macaque. J038 neutralizes 54% of global circulating HIV‐1 strains. Its binding induces a unique “up” conformation for one of the V2 loops in the trimeric envelope glycoprotein and is heavily dependent on glycan, which provides nearly half of the binding surface. Their unmutated common ancestor neutralizes the autologous virus. Continuous maturation enhances neutralization potency and breadth of J038 lineage antibodies via expanding antibody‐Env contact areas surrounding the core region contacted by germline‐encoded residues. Developmental details and recognition features of J038 lineage antibodies revealed here provide a new pathway for elicitation and maturation of V2‐targeting bnAbs.

The combined effects of acute temperature and salinity on osmolality, expressions of heat shock proteins mRNA (hsp70, hsp90a and hsp90b) and superoxide dismutase mRNA (sod) were investigated in the ...sea cucumber Apostichopus japonicus Selenka. There were 12 treatments (combinations of temperature at 16, 20, 24 and 28 °C and salinity at 22, 27 and 32 ppt). In low salinity environments, the osmolality of the sea cucumber’s coelomic fluid decreased immediately and reached osmotic balance within 6 h. The decline of osmolality after 2 h of hypo-osmotic stress was faster at high temperatures (28 °C) than that at low temperatures (16 and 20 °C). Cellular level stress was indicated by up-regulation of hsp70, hsp90s and sod mRNA, and the maximal expression of all genes occurred at 6 h after stresses. The up-regulation of hsps and sod mRNA indicated the emergence of protein denaturation and oxidative damage and also suggested an increase in energy consumption at high temperature and low salinity. These results indicated that high temperature and low salinity could change biochemical pathways and energy budgets and then potentially impair the osmoregulation of the sea cucumber. Therefore, effective ways should be taken (e.g., draining off the upper freshwater, exchanging water and adding man-made sea water) to prevent the damage to sea cucumber culture caused by low salinity induced by rainstorms, especially at high temperature.

The Omicron variants of SARS-CoV-2, primarily authenticated in November 2021 in South Africa, has initiated the 5th wave of global pandemics. Here, we systemically examined immunological and ...metabolic characteristics of Omicron variants infection. We found Omicron resisted to neutralizing antibody targeting receptor binding domain (RBD) of wildtype SARS-CoV-2. Omicron could hardly be neutralized by sera of Corona Virus Disease 2019 (COVID-19) convalescents infected with the Delta variant. Through mass spectrometry on MHC-bound peptidomes, we found that the spike protein of the Omicron variants could generate additional CD8 + T cell epitopes, compared with Delta. These epitopes could induce robust CD8 + T cell responses. Moreover, we found booster vaccination increased the cross-memory CD8 + T cell responses against Omicron. Metabolic regulome analysis of Omicron-specific T cell showed a metabolic profile that promoted the response of memory T cells. Consistently, a greater fraction of memory CD8 + T cells existed in Omicron stimulated peripheral blood mononuclear cells (PBMCs). In addition, CD147 was also a receptor for the Omicron variants, and CD147 antibody inhibited infection of Omicron. CD147-mediated Omicron infection in a human CD147 transgenic mouse model induced exudative alveolar pneumonia. Taken together, our data suggested that vaccination booster and receptor blocking antibody are two effective strategies against Omicron.
