Fiber addition enhances the composite action between the steel tube and concrete core, increasing the strength of the concrete core. To better understand how fiber-reinforced infilled steel-concrete ...composite thin-walled columns (SCTWCs) behave, multiple investigations have been conducted using both experimental and analytical methods. This article provides a comprehensive review of SCTWCs' confinement approaches using fiber-reinforced polymer (FRP) and carbon fiber-reinforced polymer (CFRP). In this research, the behavior and formation of FRP and CFRP wrappings of the SCTWCs are reviewed and discussed. The ability of the FRP to serve as a confining material and reinforcement for the columns has increased its use in columns applications. The FRP can be applied to reinforce the structures from the exterior. By applying the CFRP strips, the columns' load-carrying capacity is improved up to 30% when compared with their corresponding un-strengthened columns. External bonding of the CFRP strips efficiently creates external confinement pressure, prevents local buckling of the steel tubes, and enhances the load-carrying capacity of the SCTWCs. The primary goal is to facilitate a clear understanding of the SCTWCs. This article helps structural researchers and engineers better understand the behavior of the SCTWCs that include the FRP and CFRP composites as external reinforcement. Future research directions are also suggested, which utilize previous research works.

The symmetry breaking of protein distribution and cytoskeleton organization is an essential aspect for the development of apicobasal polarity. In embryonic cells this process is largely cell ...autonomous, while differentiated epithelial cells collectively polarize during epithelium formation. Here, we demonstrate that the de novo polarization of mature hepatocytes does not require the synchronized development of apical poles on neighbouring cells. De novo polarization at the single-cell level by mere contact with the extracellular matrix and immobilized cadherin defining a polarizing axis. The creation of these single-cell liver hemi-canaliculi allows unprecedented imaging resolution and control and over the lumenogenesis process. We show that the density and localization of cadherins along the initial cell-cell contact act as key triggers of the reorganization from lateral to apical actin cortex. The minimal cues necessary to trigger the polarization of hepatocytes enable them to develop asymmetric lumens with ectopic epithelial cells originating from the kidney, breast or colon.

Human induced pluripotent stem cells (iPSCs) represent a unique opportunity for regenerative medicine because they offer the prospect of generating unlimited quantities of cells for autologous ...transplantation, with potential application in treatments for a broad range of disorders. However, the use of human iPSCs in the context of genetically inherited human disease will require the correction of disease-causing mutations in a manner that is fully compatible with clinical applications. The methods currently available, such as homologous recombination, lack the necessary efficiency and also leave residual sequences in the targeted genome. Therefore, the development of new approaches to edit the mammalian genome is a prerequisite to delivering the clinical promise of human iPSCs. Here we show that a combination of zinc finger nucleases (ZFNs) and piggyBac technology in human iPSCs can achieve biallelic correction of a point mutation (Glu342Lys) in the α(1)-antitrypsin (A1AT, also known as SERPINA1) gene that is responsible for α(1)-antitrypsin deficiency. Genetic correction of human iPSCs restored the structure and function of A1AT in subsequently derived liver cells in vitro and in vivo. This approach is significantly more efficient than any other gene-targeting technology that is currently available and crucially prevents contamination of the host genome with residual non-human sequences. Our results provide the first proof of principle, to our knowledge, for the potential of combining human iPSCs with genetic correction to generate clinically relevant cells for autologous cell-based therapies.

One of the main challenges in screening of enhanced oil recovery (EOR) techniques is the class imbalance problem, where the number of different EOR techniques is not equal. This problem hinders the ...generalization of the data-driven methods used to predict suitable EOR techniques for candidate reservoirs. The main purpose of this paper is to propose a novel approach to overcome the above challenge by taking advantage of the Power-Law Committee Machine (PLCM) technique optimized by Particle Swam Optimization (PSO) to combine the output of five cutting-edge machine learning methods with different types of learning algorithms. The PLCM method has not been used in previous studies for EOR screening. The machine learning models include the Artificial Neural Network (ANN), CatBoost, Random Forest (RF), K-Nearest Neighbors (KNN), and Support Vector Machine (SVM). The CatBoost is used for the first time in this work for screening of EOR methods. The role of the PSO is to find the optimal values for the coefficients and exponents of the power-law model. In this study, a bigger dataset than those in previous studies, including 2563 successful worldwide EOR experiences, was gathered. A bigger dataset improves the generalization of the data-driven methods and prevents overfitting. The hyperparameters of the individual machine-learning models were tuned using the fivefold cross-validation technique. The results showed that all the individual methods could predict the suitable EOR method for unseen cases with an average score of 0.868. Among the machine learning models, the KNN and SVM had the highest scores with a value of 0.894 and 0.892, respectively. Nonetheless, after combining the output of the models using the PLCM method, the score of the predictions improved to 0.963, which was a substantial increase. Finally, a feature importance analysis was conducted to find out the most influential parameters on the output. The novelty of this work is having shown the ability of the PLCM technique to construct an accurate model to overcome the class-imbalance issue in EOR screening by utilizing different types of data-driven models. According to feature importance analysis, oil gravity and formation porosity were recognized as the most influential parameters on EOR screening.

Previous studies support the textbook model that shape and color are extracted by distinct neurons in primate primary visual cortex (V1). However, rigorous testing of this model requires sampling a ...larger stimulus space than previously possible. We used stable GCaMP6f expression and two-photon calcium imaging to probe a very large spatial and chromatic visual stimulus space and map functional microarchitecture of thousands of neurons with single-cell resolution. Notable proportions of V1 neurons strongly preferred equiluminant color over achromatic stimuli and were also orientation selective, indicating that orientation and color in V1 are mutually processed by overlapping circuits. Single neurons could precisely and unambiguously code for both color and orientation. Further analyses revealed systematic spatial relationships between color tuning, orientation selectivity, and cytochrome oxidase histology.

Bypass versus Angioplasty for Severe Ischaemia of the Leg (BASIL)-2 was an open-label, pragmatic, multicentre, phase 3, randomised trial done at 41 vascular surgery units in the UK (n=39), Sweden (n=1), and Denmark (n=1). Eligible patients were those who presented to hospital-based vascular surgery units with chronic limb-threatening ischaemia due to atherosclerotic disease and who required an infra-popliteal, with or without an additional more proximal infra-inguinal, revascularisation procedure to restore limb perfusion. Participants were randomly assigned (1:1) to receive either vein bypass (vein bypass group) or best endovascular treatment (best endovascular treatment group) as their first revascularisation procedure through a secure online randomisation system. Participants were excluded if they had ischaemic pain or tissue loss considered not to be primarily due to atherosclerotic peripheral artery disease. Most vein bypasses used the great saphenous vein and originated from the common or superficial femoral arteries. Most endovascular interventions comprised plain balloon angioplasty with selective use of plain or drug eluting stents. Participants were followed up for a minimum of 2 years. Data were collected locally at participating centres. In England, Wales, and Sweden, centralised databases were used to collect information on amputations and deaths. Data were analysed centrally at the Birmingham Clinical Trials Unit. The primary outcome was amputation-free survival defined as time to first major (above the ankle) amputation or death from any cause measured in the intention-to-treat population. Safety was assessed by monitoring serious adverse events up to 30-days after first revascularisation. The trial is registered with the ISRCTN registry, ISRCTN27728689.
Between July 22, 2014, and Nov 30, 2020, 345 participants (65 19% women and 280 81% men; median age 72·5 years 62·7–79·3) with chronic limb-threatening ischaemia were enrolled in the trial and randomly assigned: 172 (50%) to the vein bypass group and 173 (50%) to the best endovascular treatment group. Major amputation or death occurred in 108 (63%) of 172 patients in the vein bypass group and 92 (53%) of 173 patients in the best endovascular treatment group (adjusted hazard ratio HR 1·35 95% CI 1·02–1·80; p=0·037). 91 (53%) of 172 patients in the vein bypass group and 77 (45%) of 173 patients in the best endovascular treatment group died (adjusted HR 1·37 95% CI 1·00–1·87). In both groups the most common causes of morbidity and death, including that occurring within 30 days of their first revascularisation, were cardiovascular (61 deaths in the vein bypass group and 49 in the best endovascular treatment group) and respiratory events (25 deaths in the vein bypass group and 23 in the best endovascular treatment group; number of cardiovascular and respiratory deaths were not mutually exclusive).
In the BASIL-2 trial, a best endovascular treatment first revascularisation strategy was associated with a better amputation-free survival, which was largely driven by fewer deaths in the best endovascular treatment group. These data suggest that more patients with chronic limb-threatening ischaemia who required an infra-popliteal, with or without an additional more proximal infra-inguinal, revascularisation procedure to restore limb perfusion should be considered for a best endovascular treatment first revascularisation strategy.

The liver parenchyma is composed of hepatocytes and bile duct epithelial cells (BECs). Controversy exists regarding the cellular origin of human liver parenchymal tissue generation during embryonic ...development, homeostasis or repair. Here we report the existence of a hepatobiliary hybrid progenitor (HHyP) population in human foetal liver using single-cell RNA sequencing. HHyPs are anatomically restricted to the ductal plate of foetal liver and maintain a transcriptional profile distinct from foetal hepatocytes, mature hepatocytes and mature BECs. In addition, molecular heterogeneity within the EpCAM
population of freshly isolated foetal and adult human liver identifies diverse gene expression signatures of hepatic and biliary lineage potential. Finally, we FACS isolate foetal HHyPs and confirm their hybrid progenitor phenotype in vivo. Our study suggests that hepatobiliary progenitor cells previously identified in mice also exist in humans, and can be distinguished from other parenchymal populations, including mature BECs, by distinct gene expression profiles.
Exposure to fine particulate matter (PM2.5) is associated with cardiovascular disease risk. To date, there are few studies on short-term PM2.5 exposure in different microenvironments and its impact ...on immediate health effects, particularly in the Southeast Asia region. This study assessed PM2.5 concentrations in different microenvironments in a densely populated city in the tropics using low-cost personal PM2.5 sensors as well as their associations with acute cardiovascular health outcomes. A total of 49 adult participants affiliated with the National University of Singapore (NUS) community were recruited. Personal low-cost sensors were used to measure PM2.5 concentrations between September 2017 and March 2019. Demographic information and time-activity patterns were collected using questionnaires. Wilcoxon pairwise comparisons were used to determine statistical differences between PM2.5 exposures at 18 different microenvironments. Generalized Estimating Equations (GEE) models were used to assess the association between PM2.5 exposure and blood pressure as well as heart rate. All models were adjusted for age, sex, body mass index, physical activity, temperature, duration of exposure, and baseline cardiovascular parameters. Significant differences in PM2.5 concentrations were observed across different microenvironments. Air-conditioned offices and tertiary teaching spaces had the lowest (median = 13.1 μg/m3) and hawker centres had the highest (median = 32.0 μg/m3) PM2.5 concentrations. Significant positive associations between PM2.5 exposure and heart rate (β = 0.40, p = 4.6 × 10−5) as well as diastolic blood pressure (β = 0.16, p = 0.0077) were also observed. Short-term exposure to PM2.5 was significantly associated with higher heart rate and blood pressure. Further work is needed to investigate the variations within each type of microenvironment and expand the study to other sub-populations such as the elderly and children.

The modification of the surface wetting characteristics in fractured oil-wet carbonate reservoirs, by reversing wettability from oil-wet to water-wet, leads to improved oil recovery. However, in ...order to obtain a successful oil recovery process, it is crucial to understand the active mechanisms of wettability alteration. This study looks at the effect of sulfate ions as one of the most promising wettability influencing ions on the wetting properties of oil-wet calcite; the effect is studied both with and without the presence of cationic surfactant and possible mechanisms of wettability alteration are explored. A number of analytical techniques were utilized to analyze the mineral surface before and after treatment. The study presents a thorough discussion of the influence of sulfate ions in displacing adsorbed carboxylate from the oil-wet surface, both with and without the presence of cationic surfactant are discussed thoroughly. The interaction between sulfate ions and the calcium ions attached to carboxylate groups on the surface is believed to be the main active mechanism of wettability alteration at high concentration of sulfate ions. Ion-exchange between the hydroxide group and the adsorbed stearate ion on the calcite surface is shown to act as a supplementary mechanism that desorbs stearate ions from the surface. In the treatment of an aged calcite surface with sulfate ions, a combination of these two mechanisms resulted in a more water-wet surface. The copresence of sulfate ions and cationic surfactant resulted in a further reduction in the amount of adsorbed carboxylate on the surface. This can be attributed to the release of adsorbed carboxylate groups on the surface through ion pair formation with the cationic surfactant. The desorption of negatively charged carboxylate groups from the surface facilitates the approach of negatively charged sulfate ions to the aged calcite surface. It can therefore be concluded that sulfate ions accompanied by cationic surfactant molecules can alter the wetting properties toward water-wet state more effectively than sulfate ions alone.
