Entanglement of animals is one of the main environmental impacts of waste plastic. A 2015 review of entanglement records found that the proportion of affected seabirds increased from 16% of species ...to 25% over the last two decades. However, this was restricted to published records; Google Images and other web-based sources indicate that at least 147 seabird species (36%), as well as 69 freshwater birds (10%) and 49 landbirds (0.5%) from 53 families have been entangled in plastic or other synthetic materials. Fishing gear is responsible for entangling most species (83%), although it is often difficult to differentiate entanglement from bycatch on active gear. Mitigation measures include banning high-risk applications where there are alternatives (e.g. six-pack rings), discouraging the use of high-risk items (e.g. balloons on strings, ‘manja’ kites), and encouraging fishers to not discard waste fishing gear by providing specific receptacles and associated educational signage in fishing areas.

Floating persistent debris, primarily made from plastic, disperses long distances from source areas and accumulates in oceanic gyres. However, biofouling can increase the density of debris items to ...the point where they sink. Buoyancy is related to item volume, whereas fouling is related to surface area, so small items (which have high surface area to volume ratios) should start to sink sooner than large items. Empirical observations off South Africa support this prediction: moving offshore from coastal source areas there is an increase in the size of floating debris, an increase in the proportion of highly buoyant items (e.g. sealed bottles, floats and foamed plastics), and a decrease in the proportion of thin items such as plastic bags and flexible packaging which have high surface area to volume ratios. Size-specific sedimentation rates may be one reason for the apparent paucity of small plastic items floating in the world's oceans.
A striking change has happened in the field of immunology whereby specific metabolic processes have been shown to be a critical determinant of immune cell activation. Multiple immune receptor types ...rewire metabolic pathways as a key part of how they promote effector functions. Perhaps surprisingly for immunologists, the Krebs cycle has emerged as the central immunometabolic hub of the macrophage. During proinflammatory macrophage activation, there is an accumulation of the Krebs cycle intermediates succinate and citrate, and the Krebs cycle-derived metabolite itaconate. These metabolites have distinct nonmetabolic signaling roles that influence inflammatory gene expression. A key bioenergetic target for the Krebs cycle, the electron transport chain, also becomes altered, generating reactive oxygen species from Complexes I and III. Similarly, alternatively activated macrophages require α-ketoglutarate-dependent epigenetic reprogramming to elicit anti-inflammatory gene expression. In this review, we discuss these advances and speculate on the possibility of targeting these events therapeutically for inflammatory diseases.
Plastic pollution is ubiquitous throughout the marine environment, yet estimates of the global abundance and weight of floating plastics have lacked data, particularly from the Southern Hemisphere ...and remote regions. Here we report an estimate of the total number of plastic particles and their weight floating in the world's oceans from 24 expeditions (2007-2013) across all five sub-tropical gyres, costal Australia, Bay of Bengal and the Mediterranean Sea conducting surface net tows (N = 680) and visual survey transects of large plastic debris (N = 891). Using an oceanographic model of floating debris dispersal calibrated by our data, and correcting for wind-driven vertical mixing, we estimate a minimum of 5.25 trillion particles weighing 268,940 tons. When comparing between four size classes, two microplastic <4.75 mm and meso- and macroplastic >4.75 mm, a tremendous loss of microplastics is observed from the sea surface compared to expected rates of fragmentation, suggesting there are mechanisms at play that remove <4.75 mm plastic particles from the ocean surface.

The low-rank approximation is a complexity reduction technique to approximate a tensor or a matrix with a reduced rank, which has been applied to the simulation of high dimensional problems to reduce the memory required and computational cost. In this work, a dynamical low-rank approximation method is developed for the time-dependent radiation transport equation in 1-D and 2-D Cartesian geometries. Using a finite volume discretization in space and a spherical harmonics basis in angle, we construct a system that evolves on a low-rank manifold via an operator splitting approach. Numerical results on five test problems demonstrate that the low-rank solution requires less memory and computational time than solving the full rank equations with the same accuracy. It is furthermore shown that the low-rank algorithm can obtain high-fidelity results by increasing the number of basis functions while keeping the rank fixed.

The emergence of SARS-CoV-2 variants has raised concerns about altered sensitivity to antibody-mediated immunity. The relative resistance of SARS-CoV-2 variants B.1.1.7 and B.1.351 to antibody ...neutralization has been recently investigated. We report that another emergent variant from Brazil, P.1, is not only refractory to multiple neutralizing monoclonal antibodies but also more resistant to neutralization by convalescent plasma and vaccinee sera. The magnitude of resistance is greater for monoclonal antibodies than vaccinee sera and evident with both pseudovirus and authentic P.1 virus. The cryoelectron microscopy structure of a soluble prefusion-stabilized spike reveals that the P.1 trimer adopts exclusively a conformation in which one of the receptor-binding domains is in the “up” position, which is known to facilitate binding to entry receptor ACE2. The functional impact of P.1 mutations thus appears to arise from local changes instead of global conformational alterations. The P.1 variant threatens current antibody therapies but less so protective vaccine efficacy.

South Africa is thought to be one of the worst contributors of plastic into the sea globally. Although some plastic items derive from offshore sources (mainly fishing and other maritime activities, ...but also long-distance transport), the importance of local, land-based sources is indicated by the composition of beach debris and the concentration of macro-, meso- and microplastics close to urban source areas. Some 60–90% of plastic from land-based sources is expected to strand on beaches, but plastic standing stocks on beaches are much lower than global model predictions of land-based pollution. Burial in beaches and transport into backshore vegetation are significant sinks, although this plastic is likely to be released as the climate crisis leads to rising sea levels and more extreme storms. Most buried items are fairly small, while many larger items, which account for most of the mass of plastic, are removed from beaches by cleaning efforts. However, even daily accumulation rate estimates – which exclude the effects of cleaning – fall well short of model predictions of plastic leakage from land-based sources. Oceanographic models predict that plastics entering the sea from South Africa are exported to the South Atlantic and Indian Oceans, with the proportion depending on source location and item density. At sea, floating macroplastic is concentrated close to urban centres. Farther offshore, plastic items tend to be large and buoyant because biofouling causes small, low buoyancy items to sink. Size-selective removal of plastics by biota might also contribute to the paucity of floating microplastics (25 mm). • The seabed is a long-term sink for marine plastics, but densities of plastic on the seabed around South Africa are still modest. • The global model prediction of plastic leakage from South Africa into the sea probably is a gross overestimate.

Saturated fatty acids (FA) exert adverse health effects and are more likely to cause insulin resistance and type 2 diabetes than unsaturated FA, some of which exert protective and beneficial effects. ...Saturated FA, but not unsaturated FA, activate Jun N-terminal kinase (JNK), which has been linked to obesity and insulin resistance in mice and humans. However, it is unknown how saturated and unsaturated FA are discriminated. We now demonstrate that saturated FA activate JNK and inhibit insulin signaling through c-Src activation. FA alter the membrane distribution of c-Src, causing it to partition into intracellular membrane subdomains, where it likely becomes activated. Conversely, unsaturated FA with known beneficial effects on glucose metabolism prevent c-Src membrane partitioning and activation, which are dependent on its myristoylation, and block JNK activation. Consumption of a diabetogenic high-fat diet causes the partitioning and activation of c-Src within detergent insoluble membrane subdomains of murine adipocytes.

Dynamical low-rank (DLR) approximation methods have previously been developed for time-dependent radiation transport problems. One crucial drawback of DLR is that it does not conserve important quantities of the calculation, which limits the applicability of the method. Here we address this conservation issue by solving a low-order equation with closure terms computed via a high-order solution calculated with DLR. We observe that the high-order solution well approximates the closure term, and the low-order solution can be used to correct the conservation bias in the DLR evolution. We also apply the linear discontinuous Galerkin method for the spatial discretization. We then demonstrate with the numerical results that this so-called high-order/low-order (HOLO) algorithm is conservative without sacrificing computational efficiency and accuracy.

Post-traumatic stress disorder, panic disorder and phobia manifest in ways that are consistent with an uncontrollable state of fear. Their development involves heredity, previous sensitizing ...experiences, association of aversive events with previous neutral stimuli, and inability to inhibit or extinguish fear after it is chronic and disabling. We highlight recent progress in fear learning and memory, differential susceptibility to disorders of fear, and how these findings are being applied to the understanding, treatment and possible prevention of fear disorders. Promising advances are being translated from basic science to the clinic, including approaches to distinguish risk versus resilience before trauma exposure, methods to interfere with fear development during memory consolidation after a trauma, and techniques to inhibit fear reconsolidation and to enhance extinction of chronic fear. It is hoped that this new knowledge will translate to more successful, neuroscientifically informed and rationally designed approaches to disorders of fear regulation.
