Edward Jenner and his contemporaries believed that his variolae vaccinae originated in horses and molecular analyses show that modern vaccinia virus (VACV) strains share common ancestry with horsepox ...virus (HPXV). Given concerns relating to the toxicity of modern VACV vaccines, we asked whether an HPXV-based vaccine might provide a superior alternative. Since HPXV may be extinct and the only specimen of HPXV that has been identified is unavailable for investigation, we explored whether HPXV could be obtained by large-scale gene synthesis. Ten large (10-30 kb) fragments of DNA were synthesized based on the HPXV sequence along with two 157 nt VACV terminal sequences, and were recombined into a live synthetic chimeric HPXV (scHPXV) in cells infected with Shope fibroma virus (SFV). Sequencing of the 212 kbp scHPXV confirmed it encoded a faithful copy of the input DNA. We believe this is the first complete synthesis of a poxvirus using synthetic biology approaches. This scHPXV produced smaller plaques, produced less extracellular virus and exhibited less virulence in mice than VACV, but still provided vaccine protection against a lethal VACV challenge. Collectively, these findings support further development of scHPXV as a novel replication-proficient smallpox vaccine.

Despite allegations that foreign aid promotes corruption and patronage, little is known about how recipient governments' electoral incentives influence aid spending. This article proposes a ...distributional politics model of aid spending in which governments use their informational advantages over donors in order to allocate a disproportionate share of aid to electorally strategic supporters, allowing governments to translate aid into votes. To evaluate this argument, the author codes data on the spatial distribution of multilateral donor projects in Kenya from 1992 to 2010 and shows that Kenyan governments have consistently influenced the aid allocation process in favor of copartisan and coethnic voters, a bias that holds for each of Kenya's last three regimes. He confirms that aid distribution increases incumbent vote share. This evidence suggests that electoral motivations play a significant role in aid allocation and that distributional politics may help explain the gap between donor intentions and outcomes.

In this paper, we review algorithmic bias in education, discussing the causes of that bias and reviewing the empirical literature on the specific ways that algorithmic bias is known to have ...manifested in education. While other recent work has reviewed mathematical definitions of fairness and expanded algorithmic approaches to reducing bias, our review focuses instead on solidifying the current understanding of the concrete impacts of algorithmic bias in education—which groups are known to be impacted and which stages and agents in the development and deployment of educational algorithms are implicated. We discuss theoretical and formal perspectives on algorithmic bias, connect those perspectives to the machine learning pipeline, and review metrics for assessing bias. Next, we review the evidence around algorithmic bias in education, beginning with the most heavily-studied categories of race/ethnicity, gender, and nationality, and moving to the available evidence of bias for less-studied categories, such as socioeconomic status, disability, and military-connected status. Acknowledging the gaps in what has been studied, we propose a framework for moving from unknown bias to known bias and from fairness to equity. We discuss obstacles to addressing these challenges and propose four areas of effort for mitigating and resolving the problems of algorithmic bias in AIED systems and other educational technology.

and humans, and the first assessment of how changes over time affect the landscape of predicted orthologs between two species. Although we find that updates to the orthology-prediction methods significantly changed the landscape of
-human orthologs predicted by individual programs and-unexpectedly-reduced agreement among them, we also show that our meta-analysis approach "buffered" against changes in gene content. We show that adding results from more programs did not lead to many additions to the list and discuss reasons to avoid assigning "scores" based on support by individual orthology-prediction programs; the treatment of "legacy" genes no longer predicted by these programs; and the practical difficulties of updating due to encountering deprecated, changed, or retired gene identifiers. In addition, we consider what other criteria may support claims of orthology and alternative approaches to find potential orthologs that elude identification by these programs. Finally, we created a new web-based tool that allows for rapid searches of OL2 by gene identifiers, protein domains InterPro and SMART (Simple Modular Architecture Research Tool, or human disease associations (OMIM (Online Mendelian Inheritence in Man, and also includes available RNA-interference resources to facilitate potential translational cross-species studies.
Abstract
The planetary and lunar ephemerides called DE440 and DE441 have been generated by fitting numerically integrated orbits to ground-based and space-based observations. Compared to the previous ...general-purpose ephemerides DE430, seven years of new data have been added to compute DE440 and DE441, with improved dynamical models and data calibration. The orbit of Jupiter has improved substantially by fitting to the Juno radio range and Very Long Baseline Array (VLBA) data of the Juno spacecraft. The orbit of Saturn has been improved by radio range and VLBA data of the Cassini spacecraft, with improved estimation of the spacecraft orbit. The orbit of Pluto has been improved from use of stellar occultation data reduced against the Gaia star catalog. The ephemerides DE440 and DE441 are fit to the same data set, but DE441 assumes no damping between the lunar liquid core and the solid mantle, which avoids a divergence when integrated backward in time. Therefore, DE441 is less accurate than DE440 for the current century, but covers a much longer duration of years −13,200 to +17,191, compared to DE440 covering years 1550–2650.

Here we use a combination of transcriptomics, metabolic logic and pathway reconstitution to elucidate a near-complete biosynthetic pathway to colchicine without prior knowledge of biosynthetic genes, a sequenced genome or genetic tools in the native host. We uncovered eight genes from Gloriosa superba for the biosynthesis of N-formyldemecolcine, a colchicine precursor that contains the characteristic tropolone ring and pharmacophore of colchicine
. Notably, we identified a non-canonical cytochrome P450 that catalyses the remarkable ring expansion reaction that is required to produce the distinct carbon scaffold of colchicine. We further used the newly identified genes to engineer a biosynthetic pathway (comprising 16 enzymes in total) to N-formyldemecolcine in Nicotiana benthamiana starting from the amino acids phenylalanine and tyrosine. This study establishes a metabolic route to tropolone-containing colchicine alkaloids and provides insights into the unique chemistry that plants use to generate complex, bioactive metabolites from simple amino acids.

We propose that advertisement (ad) content for food products can affect taste perception by affecting sensory cognitions. Specifically, we show that multisensory ads result in higher taste ...perceptions than ads focusing on taste alone, with this result being mediated by the excess of positive over negative sensory thoughts. Since the ad effect is thoughts‐driven or cognitive, restricting cognitive resources (imposing cognitive load) attenuates the enhancing effect of the multiple‐sense ad. Our results are exhibited across three experiments and have many implications for cognition and sensory perception research within consumer behavior, as well as several practical implications.

This article draws upon existing literature to document and describe the rise of finance in food provisioning. It queries the role of financialization in the contemporary food crisis and analyzes its ...impacts upon the distribution of power and wealth within and along the generalized agro-food supply chain. A systematic treatment of key nodes in the supply chain reveals four key insights: (1) the line between finance and food provisioning has become increasingly blurred in recent decades, with financial actors taking a growing interest in food and agriculture and agro-food enterprises becoming increasingly involved in financial activities; (2) financialization has reinforced the position of food retailers as the dominant actors within the agro-food system, though they are largely subject to the dictates of finance capital and face renewed competition from financialized commodity traders; (3) financialization has intensified the exploitation of food workers, increasing their workload while pushing down their real wages and heightening the precarity of their positions, and (4) small-scale farmers have been especially hard hit by financialization, as their livelihoods have become even more uncertain due to increasing volatility in agricultural markets, they have become weaker vis-à-vis other actors in the agro-food supply chain, and they face growing competition for their farmland. The paper concludes by identifying themes for future research and asking readers to reimagine the role of finance in food provisioning.
