To control the COVID-19 pandemic and prevent its resurgence in areas preparing for a return of economic activities, a method for a rapid, simple, and inexpensive point-of-care diagnosis and mass ...screening is urgently needed. We developed and evaluated a one-step colorimetric reverse-transcriptional loop-mediated isothermal amplification assay (COVID-19-LAMP) for detection of SARS-CoV-2, using SARS-CoV-2 isolate and respiratory samples from patients with COVID-19 (
= 223) and other respiratory virus infections (
= 143). The assay involves simple equipment and techniques and low cost, without the need for expensive qPCR machines, and the result, indicated by color change, is easily interpreted by naked eyes. COVID-19-LAMP can detect SARS-CoV-2 RNA with detection limit of 42 copies/reaction. Of 223 respiratory samples positive for SARS-CoV-2 by qRT-PCR, 212 and 219 were positive by COVID-19-LAMP at 60 and 90 min (sensitivities of 95.07% and 98.21%) respectively, with the highest sensitivities among nasopharyngeal swabs (96.88% and 98.96%), compared to sputum/deep throat saliva samples (94.03% and 97.02%), and throat swab samples (93.33% and 98.33%). None of the 143 samples with other respiratory viruses were positive by COVID-19-LAMP, showing 100% specificity. Samples with higher viral load showed shorter detection time, some as early as 30 min. This inexpensive, highly sensitive and specific COVID-19-LAMP assay can be useful for rapid deployment as mobile diagnostic units to resource-limiting areas for point-of-care diagnosis, and for unlimited high-throughput mass screening at borders to reduce cross-regional transmission.

Through a survey of 296 female students in Singapore aged 12–18 years regarding their selfie-posting behaviors on Instagram, this study explored teenage girls’ perspectives on the importance of receiving peer feedback on selfies. The associations between peer feedback importance and self-esteem as well as depressed mood were also examined. Drawing on coping theory, this study further investigated how the importance of peer feedback and depressed mood influence girls’ coping responses to the stress from lacking feedback. The results of structural equation modeling demonstrate that self-esteem exerts a negative effect on the importance of peer feedback and depressed mood, but the importance of peer feedback is positively associated with depressed mood. In addition, both the importance of peer feedback and depressed mood positively impact problem-focused and emotion-focused coping strategies. The results suggest that peer feedback has become a “KPI” of selfie-posting behaviors for teenage girls, and these girls handle the involved stress by a concomitant adoption of both types of coping.

Systemic lupus erythematosus (SLE), an autoimmune disorder that damages various organ systems, is caused by a combination of genetic and environmental factors. Although germline mutations of several ...genes are known to cause juvenile SLE, most of the susceptibility genetic variants of adult SLE are common variants of the population, somatic mutations that cause or exacerbate SLE have not been reported. We hereby report a refractory SLE case with monocytosis accompanying somatic KRAS mutation that have been shown to cause lupus-like symptoms.
A 60-year-old female patient who had been diagnosed with SLE was admitted to our hospital. Although prednisolone and tacrolimus treatments had kept her thrombocytopenia and anti-DNA Ab level at bay for more than 4 years, a diagnosis of transverse myelitis was made when she became acutely ill with pleocytosis. Elevated cells (predominately monocytes), protein, IgG, and IL-6 levels were also found in the cerebrospinal fluid (CSF) of the patient. Standard pulse treatments of methylprednisolone, high-dose of prednisolone, and intravenous cyclophosphamide in combination with plasma exchange could not alleviate the refractory neural and autoimmune manifestation. Monocytosis of peripheral blood was also noted. Flow cytometric analysis revealed elevated ratio of CD14+CD16+ atypical monocytes, which excluded the possibility of chronic myelomonocytic leukemia. Lupus-like symptoms with monocytosis reminded us of Ras-associated autoimmune leukoproliferative disorder, and Sanger sequencing of KRAS and NRAS genes from the patients' peripheral blood mononuclear cells (PBMC), sorted CD3+ lymphocytes and CD14+ monocytes, and cerebrospinal fluid were performed. An activating KRAS somatic mutation was found in the patients' DNA at the time of encephalomyelitis diagnosis.
Somatic mutations of some genes including KRAS may cause the refractoriness of SLE.
Since its first discovery in 1967, human coronavirus OC43 (HCoV-OC43) has been associated with mild self-limiting upper respiratory infections worldwide. Fatal primary pneumonia due to HCoV-OC43 is ...not frequently described. This study describes a case of fatal primary pneumonia associated with HCoV-OC43 in a 75-year-old patient with good past health. The viral loads of the respiratory tract specimens (bronchoalveolar lavage and endotracheal aspirate) from diagnosis to death were persistently high (3.49 × 10

copies/ml). HCoV-OC43 at a 6.46 × 10
copies/ml level was also detected from his pleural fluid 2 days before his death. Complete genome sequencing and phylogenetic analysis showed that the present HCoV-OC43 forms a distinct cluster with three other HCoV-OC43 from United States, with a bootstrap value of 100% and sharing 99.9% nucleotide identities. Pairwise genetic distance between this cluster and other HCoV-OC43 genotypes ranged from 0.27 ± 0.02% to 1.25 ± 0.01%. In contrast, the lowest pairwise genetic distance between existing HCoV-OC43 genotypes was 0.26 ± 0.02%, suggesting that this cluster constitutes a novel HCoV-OC43 genotype, which we named genotype I. Unlike genotypes D, E, F, G, and H, no recombination event was observed for this novel genotype. Structural modeling revealed that the loop with the S1/S2 cleavage site was four amino acids longer than other HCoV-OC43, making it more exposed and accessible to protease, which may have resulted in its possible hypervirulence.
A composite of poly(3-hexylthiophene) (P3HT) grafted on TiO(2) nanotubes was synthesized. It was characterized using XRD, TEM, TGA, FTIR and XPS. Cyclic voltammetry (CV) was used to elucidate the ...electrochemical behavior and evaluate the HOMO and LUMO energy levels. Photoluminescence (PL) measurements show that the emission intensity of P3HT mixed with TiO(2) nanotubes was one third of that of random P3HT, while that of P3HT grafted onto TiO(2) nanotubes was 10% of random P3HT. The results show that the P3HT grafted onto TiO(2) nanotubes is more efficient in photoinduced charge transfer than a physical mixture of P3HT and TiO(2) nanotubes, indicating this composite has potential for the fabricating hybrid organic-inorganic solid state solar cells.

Although there is considerable interest in the advance bookings model as a forecasting method in the hotel industry, there has been little research analyzing the use of an advance booking curve in ...forecasting hotel reservations. The mainstream of advance booking models reviewed in the literature uses only the bookings-on-hand data on a certain day and ignores the previous booking data. This empirical study analyzes the entire booking data set for one year provided by the Hotel ICON in Hong Kong, and identifies the trends and patterns in the data. The analysis demonstrates the use of an advance booking curve in forecasting hotel reservations at property level.

While there have been many studies using machine learning (ML) algorithms to predict process outcomes and device performance in semiconductor manufacturing, the extensively developed technology ...computer-aided design (TCAD) physical models should play a more significant role in conjunction with ML. While TCAD models have been effective in predicting the trends of experiments, a machine learning statistical model is more capable of predicting the anomalous effects that can be dependent on the chambers, machines, fabrication environment, and specific layouts. In this paper, we use an analytics-statistics mixed training (ASMT) approach using TCAD. Under this method, the TCAD models are incorporated into the machine learning training procedure. The mixed dataset with the experimental and TCAD results improved the prediction in terms of accuracy. With the application of ASMT to the BOSCH process, we show that the mean square error (MSE) can be effectively decreased when the analytics-statistics mixed training (ASMT) scheme is used instead of the classic neural network (NN) used in the baseline study. In this method, statistical induction and analytical deduction can be combined to increase the prediction accuracy of future intelligent semiconductor manufacturing.

Compared with conventional solar cells, thermophotovoltaics (TPV) can be more efficient and, thus, exceeds the detailed balance efficiency limit. In particular, the emitter of TPV is a critical ...design since it determines whether the thermalization loss can be reduced and whether the sub-band-gap radiation can be suppressed, as far as the photovoltaic (PV) process is concerned. In this paper, we propose a selective planar emitter composed of alternating ultrathin metal and dielectric layers. An aperiodic dielectric stacking is designed to tailor the emission spectrum by shaping the emission peak and by adjusting the emission wavelength. We show that the peak emission wavelength (λ emission ) is adjustable from λ = 1500 nm to λ = 2500 nm. Furthermore, the long-wavelength cutoff is very sharp for our proposed emitter structure, and the absorption is suppressed to 0.1 beyond the cutoff. The preliminary experiment is also conducted to fulfill the concept of a fully planar ultrathin refractory metal emitter design, and the result is similar to the calculated ones. We believe that the planar thermal emitter based on ultrathin metals and aperiodic dielectric stacking is very promising for future thermal emission applications since it requires no lithography and etching, provides strong peak emission power, and possesses wide wavelength scalability.

Dielectric mirrors have recently emerged for solar cells due to the advantages of lower cost, lower temperature processing, higher throughput, and zero plasmonic absorption as compared to ...conventional metallic counterparts. Nonetheless, in the past, efforts for incorporating dielectric mirrors into photovoltaics were not successful due to limited bandwidth and insufficient light scattering that prevented their wide usage. In this work, it is shown that the key for ultra-broadband dielectric mirrors is aperiodicity, or randomization. In addition, it has been proven that dielectric mirrors can be widely applicable to thin-film and thick wafer-based solar cells to provide for light trapping comparable to conventional metallic back reflectors at their respective optimal geometries. Finally, the near-field angular emission plot of Poynting vectors is conducted, and it further confirms the superior light-scattering property of dielectric mirrors, especially for diffuse medium reflectors, despite the absence of surface plasmon excitation. The preliminary experimental results also confirm the high feasibility of dielectric mirrors for photovoltaics.
