AIM： To investigate the dynamic functional and ultrastructural changes of gastric parietal cells induced by water immersion-restraint stress （WRS） in rats.METHODS： WRS model of Sprague-Dawley （SD） ...rats was established. Fifty-six male SD rats were randomly divided into control group, stress group and post-stress group. The stress group was divided into 1, 2 and 4 h stress subgroups. The post-stress group was divided into 24, 48 and 72 h subgroups. The pH value of gastric juice, ulcer index （UI） of gastric mucosa and H^＋, K^＋- ATPase activity of gastric parietal cells were measured. Ultrastructural change of parietal cells was observed under transmission electron microscope （TEM）.RESULTS： The pH value of gastric juice decreased time-dependently in stress group and increased in post-stress group. The H^＋, K^＋-ATPase activity of gastric parietal cells and the UI of gastric mucosa increased time-dependently in stress group and decreased in poststress group. Compared to control group, the pH value decreased remarkably （P = 0.0001）, the UI and H^＋, K^＋- ATPase activity increased significantly （P = 0.0001, P = 0.0174） in 4 h stress subgroup. UI was positively related with stress time （r = 0.9876, P 〈 0.01） but negatively with pH value （r = -0.8724, P 〈 0.05）. The parietal cells became active in stress group, especially in 4 h stress subgroup, in which plenty of intracellular canalicular and mitochondria were observed under TEM. In post-stress group, the parietal cells recovered to resting state.CONCOUSION： The acid secretion of parietal cells is consistent with their ultrastructural changes during the development and healing of stress ulcer induced by WRS and the degree of gastric mucosal lesions, suggesting gastric acid play an important role in the development of stress ulcer and is closely related with the recovery of gastric mucosal lesions induced by WRS.
Elemene, isolated from the Chinese medicinal herb Rhizoma Zedoariae, was shown to exhibit antitumor activity. Our previous studies showed that elemene had a markedly antineoplastic activity on ...glioma. This study was designed to investigate the proliferation inhibitory effect and the apoptosis-inducing activity of elemene on glioma cells.
The effects of elemene on cell proliferation were studied in vitro by using (3)H-TdR incorporation. The morphological alterations were confirmed by Hoechst 33258/PI staining. The apoptosis was evaluated by flow cytometry analysis and agarose gel electrophoresis.
Elemene exhibited a marked antiproliferative effect on rat glioma cell C6 and human glioma cell SHG-44. The rolls in contemporary four-high mills cannot be maintained parallel during the rolling process. Therefore, four-high rolling mill vibrations take place in six degree of freedom (DOF) leading to ...spatial behaviors involving vertical, horizontal, axial, torsional, cross and swinging vibration modes resulting in complex relative motions between the rolls. Two numerical methods, modified Riccati-transfer matrix method (Riccati-TMM) and finite element method (FEM), are presented to analyze a spatial vibration characteristic of two four-high rolling mills with different stability. The natural frequency and mode shape of four-high rolling mills are obtained, and the clearance has a great effect on natural frequency and mode shape. In addition, field testing experiment is also conducted to measure natural frequency by power spectrum analysis of rolling mill vibration. Experimental results basically agree with those calculated by Riccati-TMM and FEM, which means that the Riccati-TMM and the FEM can be used for analysis of spatial vibration of four-high rolling mill. Meanwhile, the spatial vibration shows more compound vibration behaviors and the negative effect of horizontal, vertical, cross and swinging vibration modes are effectively controlled after redesign of rolling mill. These advantages have a great significance for the rolling mill to be operated with a much higher rolling speed and improved yield of products.

AIM: To investigate the association between serum alpha-fetoprotein(AfP) levels and fatty liver disease(fLD) in a Chinese population.METHODS: A cross-sectional study was performed among subjects who ...presented for a health examination at the First Affiliated Hospital, College of Medicine, Zhejiang University in 2013. fLD was diagnosed based onan ultrasonography examination. Serum AfP levels were measured with a chemiluminescence immunoassay.RESULTS: Of the 9800 subjects enrolled, 2601 were diagnosed with fLD. Subjects with fLD had higher serum AfP levels than those without the disease. Subjects with high serum AfP levels had a higher prevalence of fLD, metabolic syndrome, and its components. Univariate logistic analysis showed that elevated serum AfP levels were associated with an increased risk of fLD(OR = 1.057, 95%CI: 1.031-1.084). however, after adjusting for covariates, AFP no longer remained significantly associated with the risk factors for fLD. CONCLUSION: Our results suggest that serum AfP levels are significantly associated with fLD and that AfP acts as a cofactor, but not as an independent factor, for fLD.
This systematic review was performed to compare the efficacy and complications of transperineal （TP） vs. transrectal （TR） prostate biopsy. A systematic research of PUBM ED, EMBASE and the Cochrane ...Library was performed to identify all clinical controlled trials on prostate cancer （PCa） detection rate and complications achieved by TP and TR biopsies. Prostate biopsies included sextant, extensive and saturation biopsy procedures. All patients were assigned to a TR group and a TP group. Subgroup analysis was performed according to prostate-specific antigen （PSA） levels and digital rectal examination （DRE） findings. The Cochrane Collaboration＇s RevMan 5.1 software was used for the meta-analysis. A total of seven trials, including three randomized controlled trials （RCTs） and four casecontrol studies （CCS）, met our inclusion criteria. There was no significant difference in the cancer detection rate between the sextant TR and TP groups （risk difference （RD）, -0.02; 95% confidence interval （CI）, -0.08-0.03; P=0.34）. Meta-analysis for RCTs combined with CCS showed that there was no difference in the cancer detection rate between the extensive TR and TP group （RD, -0.01; 95% CI, -0.05-0.04; P=0.81）. There was no significant difference in PCa detection rate between the saturation TR and TP approaches （31.4% vs. 25.7%, respectively;P=0.3）. There were also no significant differences in cancer detection between the TR and TP groups in each subgroup. Although the data on complications were not pooled for the meta-analysis, no significant difference was found when comparing TR and TP studies. TR and TP biopsies were equivalent in terms of efficiency and related complications. TP prostate biopsy should be an available and alternative procedure for use by urologists.
Background Antithrombin-Ⅲ （AT-Ⅲ）, the major inhibitor of thrombin in plasma, also has anti-inflammation property and might have positive effect on sepsis. The present study aimed to investigate the ...effects of AT-Ⅲ on inflammatory reaction and pulmonary protection in endotoxin-induced acute lung injury （ALI） rat. Methods Sixty male Sprague-Dawley rats were randomly assigned equally to normal control group, ALl group, AT-Ⅲ treatment group, AT-Ⅲ＋heparin treatment group, and heparin treatment group. The pulmonary vascular permeability index （PVPI） was measured by single nuclide tracer technique. The activity of AT-Ⅲ in plasma was determined by the method of synthetic chromogenic substrata. Tumor necrosis factor-a （TNF-a） and interleukin-6 （IL-6） levels in serum were determined by enzyme-linked immunosorbent assay. The expressions of lung tissue mitogen-activated protein kinases （ERK1/2, P38 and JNK MAPK） were determined by Western blotting. Results Rats had significantly improved lung histopathology in the AT-Ⅲ treatment group and heparin treatment group compared with the ALl group, The PVPI of the ALl group was 0.38±0.04, significantly higher than that of the normal control group （0.20±0.02, P 〈0.01）, AT-Ⅲ treatment group （0.30±0.04, P 〈0.01） and heparin treatment group （0.28±0.04, P 〈0.01） respectively. There were no significant differences of PVPI in the ALl group and AT-Ⅲ＋heparin treatment group. The activity of AT-Ⅲ in plasma in the ALl group was （76±8）%, significantly lower than that of the normal control group （（96±11）%, P 〈0.05） and AT-Ⅲ treatment group （（105±17）%, P 〈0.05） respectively. The serum levels of TNF-α and I L-6 of the ALl group were （2.770±0.373） μg/L and （1.615±0.128） ng/ml respectively, significantly higher than those of the normal control group （0.506±0.093） μg/L and （0.233±0.047） ng/ml respectively, all P 〈0.01）, AT-Ⅲ treatment group （（1.774±0.218） pg/L and （1.140±0145） ng/ml respectively, all P 〈0.01） and heparin treatment group （（1.924±0.349） μg/L and （1.223±0.127） ng/ml respectively, all P 〈0.01）. The lung tissue levels of phospho-ERK1/2 and phospho-P38 MAPK expressions were markedly higher in the ALl group than in the normal control group, AT-Ⅲ treatment group and heparin treatment group respectively. Conclusions AT-Ⅲ without concomitant heparin inhibited the activation of ERK1/2 and P38 MAPK, down-regulated the levels of downstream cytokines TNF-a and IL-6, relieved endothelial permeability, and improved the ALl in endotoxin-induced rats. It might be helpful to administrate AT-Ⅲ alone, not with concomitant heparin, to those patients with ALl and sepsis.
To establish an enzyme-linked immunosorbent assay (ELISA) for detection of IgG against penicillins and then investigate this antibody response in children.

Penicillins reacted with bovine serum albumin (BSA) to generate conjugate which was used as coating antigen (100 mg.L-1) in ELISA. A good correlation was found between absorbance (A) and dilutions of sample with a linear coefficient of 0.9918. Thirteen subjects who were clinically suspicious of penicillin allergy were tested for specific IgG by ELISA, 4 positive for benzylpenicillin, 1 for benzylpenicillin and piperacillin, and 3 for benzylpenicillin, piperacillin and ampicillin.
An ELISA was successfully established and penicillin reactive IgG response in children was heterogeneous. IgG antibodies recognized benzylpenicillin molecule, degraded products and new antigenic determinant from conjugate. Cross reactions occurred among benzylpenicillin, ampicillin and piperacillin.

During d 1-d 50 after immunization, positive % of specific IgM, IgG, and IgA to benzylpenicillin were 100%, 50%-100%, and 17%-100%, respectively. IgM and IgG to benzylpenicillin also recognized ampicillin and piperacillin. The positive % of IgM and IgG to ampicillin were 23%-100% and 50%-100%, to piperacillin 43%-100% and 50%-100%, respectively. Aged benzylpenicillin showed an inhibitory effect on specific antibodies in a dose-dependent manner, and inhibitory % of specific IgM and IgG were 29%-87% and 29%-71%. However, freshly prepared benzylpenicillin had no effect.
Specific non-IgE antibody response was successfully induced by benzylpenicillin in mice, in which the isotypes were mainly IgM and then IgG and IgA. Antibodies recognized degraded products, not benzylpenicillin molecule itself. Antigenic cross reactions occurred between benzylpenicillin, ampicillin, and piperacillin. Isotypes of antibodies responsible for cross reactions were mainly IgG and then IgM.
AIM: To investigate the photodynamic effect of Cd Se/Zn S quantum dots(QDs) on pancreatic cancer cells and elucidate the probable mechanisms.METHODS: The pancreatic cancer cell line SW1990 was ...treated with different concentrations of Cd Se/Zn S QDs(0, 0.5, 1.0, 1.5, 2.0, 2.5 μmol/L), with or without illumination. The viability of SW1990 cells was tested using the Cell Counting Kit-8(CCK-8) assay. The ultrastructural changes of SW1990 cells were observed by transmission electron microscopy. Apoptosis was detected by nuclear staining and flow cytometry(FCM). Reactive oxygen species(ROS) were measured by dichlorofluorescein diacetate via fluorescence microscopy. Expression of Bax, Bcl-2 and caspase-3 was measured by real-time polymerase chain reaction(PCR) and protein immunoblotting 24 h after SW1990 cells were treated with Cd Se/Zn S QDs and illuminated.RESULTS: The CCK-8 assay results showed that both Cd Se/Zn S QDs with and without illumination suppressed SW1990 cell proliferation. Cell viability was significantly lower when illuminated or with a longer incubation time and a higher light dose. Cd Se/Zn S QDs with illumination caused ultrastructural changes in SW1990 cells, such as organelle degeneration and chromatin condensation and aggregation at the periphery of the nucleus. Fluorescence microscopy and FCM showed that Cd Se/Zn S QDs(1.5 μmol/L) with illumination increased SW1990 cell apoptosis(53.2%) and ROS generation compared with no illumination. Real-time PCR showed that expression of Bax and caspase-3 was upregulated and Bcl-2 was downregulated. Immunoblotting results were consistent with real-time PCR results. Inhibition of ROS and apoptosis both attenuated QD-photodynamictherapy-induced cell death.CONCLUSION: Cd Se/Zn S QDs can be used as a photosensitizer to inhibit SW1990 cell proliferation through ROS generation and apoptotic protein expression regulation.
