Abstract
A simple graph with edge‐connectivity and minimum degree is maximally edge‐connected if . In 1964, given a nonincreasing degree sequence , Jack Edmonds showed that there is a realization of ...that is ‐edge‐connected if and only if with when . We strengthen Edmonds's result by showing that given a realization of if is a spanning subgraph of with such that when , then there is a maximally edge‐connected realization of with as a subgraph. Our theorem tells us that there is a maximally edge‐connected realization of that differs from by at most edges. For , if has a spanning forest with components, then our theorem says there is a maximally edge‐connected realization that differs from by at most edges. As an application we combine our work with Kundu's ‐factor theorem to show there is a maximally edge‐connected realization with a ‐factor for and present a partial result to a conjecture that strengthens the regular case of Kundu's ‐factor theorem.

Phosphate remediation is important for preventing eutrophication in fresh waters and maintaining water quality. One approach for phosphate removal involves the utilization of molecular receptors. ...However, our understanding of anion recognition in aqueous solution and at aqueous interfaces is underdeveloped, and the rational design of surface-immobilized receptors is still largely unexplored. Herein, we evaluated the driving forces controlling phosphate binding to elementary amphiphilic receptors anchored at air–water interfaces. We designed biologically inspired receptors with neutral thiourea, positively charged guanidinium, and thiouronium units that all formed Langmuir monolayers. Phosphate binding was quantitatively examined using surface pressure–area isotherms and infrared reflection–absorption spectroscopy (IRRAS). The receptors within this homologous series differ in functional group, charge, and number of alkyl chains to help distinguish the fundamental components influencing anion recognition at aqueous interfaces. The two charged receptors bearing two alkyl chains each displayed strong phosphate affinities and 103- and 101-fold anti-Hofmeister selectivity over chloride, respectively. Neutral thiourea and the single-chain guanidinium receptor did not bind phosphate, revealing the importance of electrostatic interactions and supramolecular organization. Consistently, charge screening at high ionic strength weakens binding. Spectroscopic results confirmed phosphate binding to the double alkyl chain guanidinium receptor, whereas surface pressure isotherm results alone showed a minimal change, thus emphasizing the importance of interfacial spectroscopy. We found that the binding site identity, charged interface created by the electrical double layer, and supramolecular superstructure all affect interfacial binding. These detailed insights into phosphate recognition at aqueous interfaces provide a foundation to develop efficient receptors for phosphate capture.

The lowering genotyping cost is ushering in a wider interest and adoption of genomic prediction and selection in plant breeding programs worldwide. However, improper conflation of historical and ...recent linkage disequilibrium between markers and genes restricts high accuracy of genomic prediction (GP). Multiple ancestors may share a common haplotype surrounding a gene, without sharing the same allele of that gene. This prevents parsing out genetic effects associated with the underlying allele of that gene among the set of ancestral haplotypes. We present “Parental Allele Tracing, Recombination Identification, and Optimal predicTion” (i.e., PATRIOT) approach that utilizes marker data to allow for a rapid identification of lines carrying specific alleles, increases the accuracy of genomic relatedness and diversity estimates, and improves genomic prediction. Leveraging identity-by-descent relationships, PATRIOT showed an improvement in GP accuracy by 16.6% relative to the traditional rrBLUP method. This approach will help to increase the rate of genetic gain and allow available information to be more effectively utilized within breeding programs.
Recent aircraft measurements over the northwest Atlantic enable an investigation of how entrainment from the free troposphere (FT) impacts cloud condensation nucleus (CCN) concentrations in the ...marine boundary layer (MBL) during cold-air outbreaks (CAOs), motivated by the role of CCN in mediating transitions from closed to open-cell regimes. Observations compiled over eight flights indicate predominantly far lesser CCN concentrations in the FT than in the MBL. For one flight, a fetch-dependent MBL-mean CCN budget is compiled from estimates of sea-surface fluxes, entrainment of FT air, and hydrometeor collision-coalescence, based on in-situ and remote-sensing measurements. Results indicate a dominant role of FT entrainment in reducing MBL CCN concentrations, consistent with satellite-observed trends in droplet number concentration upwind of CAO cloud-regime transitions over the northwest Atlantic. Relatively scant CCN may widely be associated with FT dry intrusions, and should accelerate cloud-regime transitions where underlying MBL air is CCN-rich, thereby reducing regional albedo.

To meet this goal, we developed a multiview image-based yield estimation framework utilizing deep learning architectures. Plant images captured from different angles were fused to estimate the yield and subsequently to rank soybean genotypes for application in breeding decisions. We used data from controlled imaging environment in field, as well as from plant breeding test plots in field to demonstrate the efficacy of our framework via comparing performance with manual pod counting and yield estimation. Our results demonstrate the promise of ML models in making breeding decisions with significant reduction of time and human effort and opening new breeding method avenues to develop cultivars.
Sickle cell disease (SCD) is a group of related yet genetically complex hemoglobinopathies. Universal newborn screening (NBS) for SCD is performed in the United States and many other nations. ...Classical, protein-based laboratory methods are often adequate for the diagnosis of SCD but have specific limitations in the context of NBS. A particular challenge is the differentiation of sickle cell anemia (SCA) from the benign condition, compound heterozygosity for HbS and gene-deletion hereditary persistence of fetal hemoglobin (HbS/HPFH). We describe a sequential cohort of 44 newborns identified over 4.5 years who had molecular genetic testing incorporated into NBS for presumed SCA (an “FS” pattern). The final diagnosis was something other than SCA in six newborns (12%). Three (7%) had HbS/HPFH. All had a final, correct diagnosis at the time of their first scheduled clinic visit in our center (median 8 weeks of age). None received initial counseling for an incorrect diagnosis. In summary, genetic testing as a component of NBS for SCD is necessary to provide correct genetic counseling and education for all newborns' families at their first visit to a sickle cell center. Genetic testing also permits the use of early, pre-symptomatic hydroxyurea therapy by preventing infants with HbS/HPFH from receiving unnecessary therapy. We argue that genetic testing should be incorporated into contemporary NBS for SCD.
Abstract
Background
Sickle cell disease (SCD) is a severe and devastating hematological disorder that affects over 100,000 persons in the USA and millions worldwide. Hydroxyurea is the primary ...disease-modifying therapy for the SCD, with proven benefits to reduce both short-term and long-term complications. Despite the well-described inter-patient variability in pharmacokinetics (PK), pharmacodynamics, and optimal dose, hydroxyurea is traditionally initiated at a weight-based dose with a subsequent conservative dose escalation strategy to avoid myelosuppression. Because the dose escalation process is time consuming and requires frequent laboratory checks, many providers default to a fixed dose, resulting in inadequate hydroxyurea exposure and suboptimal benefits for many patients. Results from a single-center trial of individualized, PK-guided dosing of hydroxyurea for children with SCD suggest that individualized dosing achieves the optimal dose more rapidly and provides superior clinical and laboratory benefits than traditional dosing strategies. However, it is not clear whether these results were due to individualized dosing, the young age that hydroxyurea treatment was initiated in the study, or both. The Hydroxyurea Optimization through Precision Study (HOPS) aims to validate the feasibility and benefits of this PK-guided dosing approach in a multi-center trial.
Methods
HOPS is a randomized, multicenter trial comparing standard vs. PK-guided dosing for children with SCD as they initiate hydroxyurea therapy. Participants (ages 6 months through 21 years), recruited from 11 pediatric sickle cell centers across the USA, are randomized to receive hydroxyurea either using a starting dose of 20 mg/kg/day (Standard Arm) or a PK-guided dose (Alternative Arm). PK data will be collected using a novel sparse microsampling approach requiring only 10 μL of blood collected at 3 time-points over 3 h. A protocol-guided strategy more aggressive protocols is then used to guide dose escalations and reductions in both arms following initiation of hydroxyurea. The primary endpoint is the mean %HbF after 6 months of hydroxyurea.
Discussion
HOPS will answer important questions about the clinical feasibility, benefits, and safety of PK-guided dosing of hydroxyurea for children with SCD with potential to change the treatment paradigm from a standard weight-based approach to one that safely and effectively optimize the laboratory and clinical response.

Prostatic carcinoma (Pca) at cystoprostatectomy is usually an incidental finding with the majority thought to be clinically insignificant. Most studies have not specifically addressed the location of ...Pca or the incidence and location of in situ or invasive urothelial carcinoma (Uca) in prostates of cystoprostatectomy specimens. The frequency of involvement of the apex with these processes has clinical implications. Specifically urinary continence following orthotopic diversion may be enhanced by prostate apical sparing. In this study the pathological features of Pca and Uca, and the frequency of apical involvement were investigated in prostates from cystoprostatectomy specimens.
Whole mounted prostates from 121 consecutive cystoprostatectomy specimens were analyzed. Pca location, tumor volume, grade, stage, surgical margin and pelvic lymph node status of Pcas were assessed. Clinically insignificant Pcas had a volume of less than 0.5 cc without Gleason pattern 4, extracapsular extension, seminal vesicle invasion, lymph node involvement or positive surgical margins. Prostate involvement by Uca or urothelial carcinoma in situ (CIS)/severe dysplasia and its location were assessed.
Of 121 prostates 50 (41%) had unsuspected Pca, of which 24 (48%) were clinically significant. Of Pcas 30 of 50 (60%) involved the apex, including 19 of 24 (79%) that were significant and 11 of 26 (42%) that were insignificant. Of 121 prostates 58 (48%) had Uca involving the prostatic stroma, noninvasive Uca or urothelial CIS/severe dysplasia in the prostatic urethra or periurethral ducts, of which 19 (33%) had apical involvement. Overall only 32 of 121 patients (26%) had no Pca or prostate Uca/CIS and only 45 (37%) had no clinically significant Pca or Uca/CIS in the prostate. However, 74 of the 121 patients (61%) had no prostatic apical involvement by Pca or Uca/CIS and 85 (70%) had no apical involvement by clinically significant Pca or Uca/CIS. Patients with prostatic apical involvement by invasive or in situ Uca uniformly had involvement of more proximal (toward the base) portions of the prostate.
The majority of prostates from cystoprostatectomies had no involvement of the prostatic apex by Uca or clinically significant Pca. Hence, most patients may be candidates for prostate apical sparing. However, involvement of the apex by Uca in any patient raises concern about procedures that leave portions of the prostate urethra after cystectomy in an effort to improve continence. In candidates for orthotopic neobladder reconstruction removing all of the prostatic urethra and sparing the remainder of the prostatic apex may allow improved preservation of urinary continence with an acceptable low risk of clinical Pca progression. Whether future strategies for preoperative exclusion of apical Pca and intraoperative assessment of more proximal prostate to help exclude apical urothelial disease may identify patients suitable for prostatic apical sparing remains to be determined. The impact on functional outcomes and cancer control also require additional study.
