During endocytosis, clathrin and the clathrin adaptor protein AP-2 (ref. 1), assisted by a variety of accessory factors, help to generate an invaginated bud at the cell membrane,. One of these ...factors is Eps15, a clathrin-coat-associated protein that binds the α-adaptin subunit of AP-2 (refs 4-8). Here we investigate the function of Eps15 by characterizing an important binding partner for its region containing EH domains; this protein, epsin, is closely related to the Xenopus mitotic phosphoprotein MP90 (ref. 10) and has a ubiquitous tissue distribution. It is concentrated together with Eps15 in presynaptic nerve terminals, which are sites specialized for the clathrin-mediated endocytosis of synaptic vesicles. The central region of epsin binds AP-2 and its carboxy-terminal region binds Eps15. Epsin is associated with clathrin coats in situ, can be co-precipitated with AP-2 and Eps15 from brain extracts, but does not co-purify with clathrin coat components in a clathrin-coated vesicle fraction. When epsin function is disrupted, clathrin-mediated endocytosis is blocked. We propose that epsin may participate, together with Eps15, in the molecular rearrangement of the clathrin coats that are required for coated-pit invagination and vesicle fission.

The results of the Polarimeter upgrade at the Nuclotron internal target are presented. The measurements are performed in the frame of the DSS project. targets at incident neutron ...momenta 3.0–4.2 GeV/c by detecting one charged particle in forward direction. The polarized neutron measurements are the first of their kind. The experiment was performed using the Nuclotron accelerator in JINR Dubna, where polarized neutrons and protons were obtained from break-up of a polarized deuteron beam which has a maximum momentum of 13 GeV/c. The polarimeter ALPOM2 was used to obtain the analyzing power dependence on the transverse momentum of the final-state nucleon. These data have been used to estimate the figure of merit of a proposed experiment at Jefferson Laboratory to measure the recoiling neutron polarization in the quasi-elastic

Clathrin-mediated endocytosis involves cycles of assembly and disassembly of clathrin coat components and their accessory proteins. Dephosphorylation of rat brain extract was shown to promote the ...assembly of dynamin 1, synaptojanin 1, and amphiphysin into complexes that also included clathrin and AP-2. Phosphorylation of dynamin 1 and synaptojanin 1 inhibited their binding to amphiphysin, whereas phosphorylation of amphiphysin inhibited its binding to AP-2 and clathrin. Thus, phosphorylation regulates the association and dissociation cycle of the clathrin-based endocytic machinery, and calcium-dependent dephosphorylation of endocytic proteins could prepare nerve terminals for a burst of endocytosis.

The Multipurpose Detector (MPD) 1 is designed to study of hot and dense baryonic matter in collisions of heavy ions over the atomic mass range 1-197 at the centre of mass energy up to ?S sub()NN 11 ...GeV (for Au79+). The MPD experiment will be carried out at the JINR accelerator complex NICA 2 which is under construction. The barrel part of the MPD consists of various detectors surrounding the interaction point. It includes a precise tracking system (time projection chamber (TPC) and silicon inner tracker (IT)) and high-performance particle identification system based on time-of-flight (TOF) and calorimeter (ECal). The triple-stack multigap resistive plate chamber is chosen as an active element of the TOF. It provides good time resolution and long term stability. This article presents parameters of the MRPC obtained using the deuteron beam of JINR accelerator Nuclotron. The time resolution is 0~4 ps with efficiency of 99%. Rate capability studies resulted with a time resolution of 60 ps and efficiency higher than 90% on the beam with particle flux densities up to 2 kHz/cm super(2).
The lymphatic drainage system of the brain (LDSB) is the removal of metabolites and wastes from its tissues. A dysfunction of LDSB is an important sign of aging, brain oncology, the Alzheimer's and ...Parkinson's diseases. The development of new strategies for diagnosis of LDSB injuries can improve prevention of age-related cerebral amyloid angiopathy, neurodegenerative and cerebrovascular diseases. There are two conditions, such as deep sleep and opening of the blood-brain-barrier (OBBB) associated with the LDSB activation. A promising candidate for measurement of LDSB could be electroencephalography (EEG). In this pilot study on rats, we tested the hypothesis, whether deep sleep and OBBB can be an informative platform for an effective extracting of information about the LDSB functions. Using the nonlinear analysis of EEG dynamics and machine learning technology, we discovered that the LDSB activation during OBBB and sleep is associated with similar changes in the EEG θ-activity. The OBBB causes the higher LDSB activation vs. sleep that is accompanied by specific changes in the low frequency EEG activity extracted by the power spectra analysis of the EEG dynamics combined with the coherence function. Thus, our findings demonstrate a link between neural activity associated with the LDSB activation during sleep and OBBB that is an important informative platform for extraction of the EEG-biomarkers of the LDSB activity. These results open new perspectives for the development of technology for the LDSB diagnostics that would open a novel era in the prognosis of brain diseases caused by the LDSB disorders, including OBBB.

The reaction p + CH2→ forward charge particle + X is used for this aim traditionally. Analyzing power of this reaction falls off as 1 p, where p is the laboratory momentum. At the proton momenta of ...order 8 GeV c, which are expected at the JLab experiment, the low analyzing power creates problems for off-line analysis of data. On the other hand, it is well known that the reaction p+p → p+p has the much more analyzing power. So, the calorimeter is predestinated for suppression of inelastic events in this reaction. In the report it is shown that the problem is solved quite well.
Clathrin-mediated endocytosis is a special form of vesicle budding important for the internalization of receptors and extracellular ligands, for the recycling of plasma membrane components, and for ...the retrieval of surface proteins destined for degradation. In nerve terminals, clathrin-mediated endocytosis is crucial for synaptic vesicle recycling. Recent structural studies have provided molecular details of coat assembly. In addition, biochemical and genetic studies have identified numerous accessory proteins that assist the clathrin coat in its function at synapses and in other systems. This review summarizes these advances with a special focus on accessory factors and highlights new aspects of clathrin-mediated endocytosis revealed by the study of these factors.

The aim of the study is to develop a methodology for the comprehensive optimization of promising schemes of water supply systems, ensuring the selection of optimal water withdrawals taking into ...account water quality, structure and parameters of transporting facilities, as well as operating modes. An algorithm of theoretical, practical and methodological recommendations on optimizing promising schemes of water supply systems is presented. A new optimization technique for group water supply systems is proposed, which allows the complex to solve the following problems: to optimize the location and performance of water intake structures from surface and underground water sources; to optimize installation sites, productivity and composition of water treatment facilities; choose the route of group pipelines and their optimal diameters.
