ABSTRACT
Eggshell quality deterioration associated with heat stress is a well-known phenomenon. The involvement of the 28-kDa calcium-binding protein (calbindin, CaBP-D28k) localization in this ...failure is not clearly understood. To test a possible direct effect of ambient temperature on calbindin-D28k localization, 40 White Leghorn laying hens were housed in individual cages and exposed to high ambient temperature (30–33°C) and thermoneutral temperature (20–22°C) which served as a control. Eggshell quality characteristics and immunohistochemical localization of all intestinal segments and eggshell gland calbindin-D28k were performed under both environmental conditions. As expected, egg weight, eggshell thickness, eggshell percentage, and eggshell density were negatively affected by high ambient temperature (P ≤ 0.01). Immunohistochemistry showed that calbindin was localized in the intestinal enterocyte cytoplasm and glandular cell cytoplasm under thermoneutral conditions. However, the calbindin intensity was prominently decreased in ileum, cecum, colon, and eggshell gland under heat stress conditions. Therefore, it could be concluded that calbindin-D28k localization in intestinal segments and eggshell gland is negatively affected by high ambient temperature which might be related to the deterioration of eggshell quality characteristics under heat stress conditions.

We report an improved measurement of the cosmic microwave background B-mode polarization power spectrum with the Polarbear experiment at 150 GHz. By adding new data collected during the second season ...of observations (2013-2014) to re-analyzed data from the first season (2012-2013), we have reduced twofold the band-power uncertainties. The band powers are reported over angular multipoles , where the dominant B-mode signal is expected to be due to the gravitational lensing of E-modes. We reject the null hypothesis of no B-mode polarization at a confidence of 3.1 including both statistical and systematic uncertainties. We test the consistency of the measured B-modes with the Λ Cold Dark Matter (ΛCDM) framework by fitting for a single lensing amplitude parameter AL relative to the Planck 2015 best-fit model prediction. We obtain 0.14(foreground) 0.04(multi), where is the fiducial ΛCDM value.
We combine high-redshift Type Ia supernovae from the first three years of the Supernova Legacy Survey (SNLS) with other supernova (SN) samples, primarily at lower redshifts, to form a high-quality ...joint sample of 472 SNe (123 low-z, 93 SDSS, 242 SNLS, and 14 Hubble Space Telescope). SN data alone require cosmic acceleration at >99.999% confidence, including systematic effects. For the dark energy equation of state parameter (assumed constant out to at least z = 1.4) in a flat universe, we find w = --0.91+0.16 --0.20(stat)+0.07 --0.14(sys) from SNe only, consistent with a cosmological constant. Our fits include a correction for the recently discovered relationship between host-galaxy mass and SN absolute brightness. We pay particular attention to systematic uncertainties, characterizing them using a systematic covariance matrix that incorporates the redshift dependence of these effects, as well as the shape-luminosity and color-luminosity relationships. Unlike previous work, we include the effects of systematic terms on the empirical light-curve models. The total systematic uncertainty is dominated by calibration terms. We describe how the systematic uncertainties can be reduced with soon to be available improved nearby and intermediate-redshift samples, particularly those calibrated onto USNO/SDSS-like systems.
Mrp antiporters and their homologues in the cation/proton antiporter 3 family of the Membrane Transporter Database are widely distributed in bacteria. They have major roles in supporting cation and ...cytoplasmic pH homeostasis in many environmental, extremophilic, and pathogenic bacteria. These antiporters require six or seven hydrophobic proteins that form hetero-oligomeric complexes, while most other cation/proton antiporters require only one membrane protein for their activity. The resemblance of three Mrp subunits to membrane-embedded subunits of the NADH:quinone oxidoreductase of respiratory chains and to subunits of several hydrogenases has raised interest in the evolutionary path and commonalities of their proton-translocating domains. In order to move toward a greater mechanistic understanding of these unusual antiporters and to rigorously demonstrate that they function as secondary antiporters, powered by an imposed proton motive force, we established a method for purification and functional reconstitution of the seven-subunit Mrp antiporter from alkaliphilic Bacillus pseudofirmus OF4. Na+/H+ antiporter activity was demonstrated by a fluorescence-based assay with proteoliposomes in which the Mrp complex was coreconstituted with a bacterial FoF1-ATPase. Proton pumping by the ATPase upon addition of ATP generated a proton motive force across the membranes that powered antiporter activity upon subsequent addition of Na+.
Upper urinary tract urothelial carcinoma (UTUC) is one of the common urothelial cancers. Its molecular pathogenesis, however, is poorly understood, with no useful biomarkers available for accurate ...diagnosis and molecular classification. Through an integrated genetic study involving 199 UTUC samples, we delineate the landscape of genetic alterations in UTUC enabling genetic/molecular classification. According to the mutational status of TP53, MDM2, RAS, and FGFR3, UTUC is classified into five subtypes having discrete profiles of gene expression, tumor location/histology, and clinical outcome, which is largely recapitulated in an independent UTUC cohort. Sequencing of urine sediment-derived DNA has a high diagnostic value for UTUC with 82.2% sensitivity and 100% specificity. These results provide a solid basis for better diagnosis and management of UTUC.

Based on an integrated genomic analysis of 199 upper urinary tract urothelial carcinomas (UTUCs), Fujii et al. identify five genetic subtypes with discrete profile of gene mutation, expression, histology, and clinical outcome and also demonstrate a high diagnostic value of sequencing urinary sediment-derived DNA for non-invasive detection of UTUCs.
