The transcription regulatory network inside a eukaryotic cell is defined by the combinatorial actions of transcription factors (TFs). However, TF binding studies in plants are too few in number to ...produce a general picture of this complex network. In this study, we use large-scale ChIP-seq to reconstruct it in the maize leaf, and train machine-learning models to predict TF binding and co-localization. The resulting network covers 77% of the expressed genes, and shows a scale-free topology and functional modularity like a real-world network. TF binding sequence preferences are conserved within family, while co-binding could be key for their binding specificity. Cross-species comparison shows that core network nodes at the top of the transmission of information being more conserved than those at the bottom. This study reveals the complex and redundant nature of the plant transcription regulatory network, and sheds light on its architecture, organizing principle and evolutionary trajectory.
The main objective of this work is to perform and validate a series of CFD simulations of the cavitating flow through a ball check valve. Experimental tests are performed in order to obtain the mass ...flow rate through the valve under different operating conditions, inducing or preventing the appearance of cavitation by conveniently adjusting the pressure level on the valve outlet port. The measurements are compared with the results of numerical (CFD) simulations of the fluid flow through the valve, with and without the inclusion of a cavitation model. The characteristic flow coefficient of the valve and the hydraulic forces on the ball are analysed.

Although iron-mediated oxidative stress has been proposed as a potential pathomechanism in Parkinson's disease, the global distribution of iron accumulation in Parkinson's disease has not yet been ...elucidated. This study used a new magnetic resonance imaging contrast, quantitative susceptibility mapping, and state-of-the-art methods to map for the first time the whole-brain landscape of magnetostatic alterations as a surrogate for iron level changes in n = 25 patients with idiopathic Parkinson's disease versus n = 50 matched controls. In addition to whole-brain analysis, a regional study including sub-segmentation of the substantia nigra into dorsal and ventral regions and qualitative assessment of susceptibility maps in single subjects were also performed. The most remarkable basal ganglia effect was an apparent magnetic susceptibility increase-consistent with iron deposition-in the dorsal substantia nigra, though an effect was also observed in ventral regions. Increased bulk susceptibility, additionally, was detected in rostral pontine areas and in a cortical pattern tightly concordant with known Parkinson's disease distributions of α-synuclein pathology. In contrast, the normally iron-rich cerebellar dentate nucleus returned a susceptibility reduction suggesting decreased iron content. These results are in agreement with previous post-mortem studies in which iron content was evaluated in specific regions of interest; however, extensive neocortical and cerebellar changes constitute a far more complex pattern of iron dysregulation than was anticipated. Such findings also stand in stark contrast to the lack of statistically significant group change using conventional magnetic resonance imaging methods namely voxel-based morphometry, cortical thickness analysis, subcortical volumetry and tract-based diffusion tensor analysis; confirming the potential of whole-brain quantitative susceptibility mapping as an in vivo biomarker in Parkinson's disease.
Spin electronics (spintronics) exploits the magnetic nature of electrons, and this principle is commercially applied in, for example, the spin valves of disk-drive read heads. There is currently ...widespread interest in developing new types of spintronic devices based on industrially relevant semiconductors, in which a spin-polarized current flows through a lateral channel between a spin-polarized source and drain. However, the transformation of spin information into large electrical signals is limited by spin relaxation, so that the magnetoresistive signals are below 1% (ref. 2). Here we report large magnetoresistance effects (61% at 5 K), which correspond to large output signals (65 mV), in devices where the non-magnetic channel is a multiwall carbon nanotube that spans a 1.5 microm gap between epitaxial electrodes of the highly spin polarized manganite La(0.7)Sr(0.3)MnO3. This spintronic system combines a number of favourable properties that enable this performance; the long spin lifetime in nanotubes due to the small spin-orbit coupling of carbon; the high Fermi velocity in nanotubes that limits the carrier dwell time; the high spin polarization in the manganite electrodes, which remains high right up to the manganite-nanotube interface; and the resistance of the interfacial barrier for spin injection. We support these conclusions regarding the interface using density functional theory calculations. The success of our experiments with such chemically and geometrically different materials should inspire new avenues in materials selection for future spintronics applications.

The main objective of this work is to develop a methodology for the parametric modelling of the flow rate in hydraulic valve systems. This methodology is based on the derivation, from CFD simulations, of the flow coefficient of the critical restrictions as a function of the Reynolds number, using a generalized square root function with two parameters. The methodology is then demonstrated by applying it to two completely different hydraulic systems: a brake master cylinder and an ABS valve. This type of parametric valve models facilitates their implementation in dynamic simulation models of complex hydraulic systems.

Spatial memory, among many other brain processes, shows hemispheric lateralization. Most of the published evidence suggests that the right hippocampus plays a leading role in the manipulation of ...spatial information. Concurrently in the hippocampus, memory consolidation during sleep periods is one of the key steps in the formation of newly acquired spatial memory traces. One of the most characteristic oscillatory patterns in the hippocampus are sharp-wave ripple (SWR) complexes. Within this complex, fast-field oscillations or ripples have been demonstrated to be instrumental in the memory consolidation process. Since these ripples are relevant for the consolidation of memory traces associated with spatial navigation, and this process appears to be lateralized, we hypothesize that ripple events between both hippocampi would exhibit different temporal dynamics. We tested this idea by using a modified "split-hyperdrive" that allows us to record simultaneous LFPs from both right and left hippocampi of Sprague-Dawley rats during sleep. We detected individual events and found that during sleep periods these ripples exhibited a different occurrence patterns between hemispheres. Most ripple events were synchronous between intra- rather than inter-hemispherical recordings, suggesting that ripples in the hippocampus are independently generated and locally propagated within a specific hemisphere. In this study, we propose the ripples' lack of synchrony between left and right hippocampi as the putative physiological mechanism underlying lateralization of spatial memory.

Neurodegenerative diseases (NDs) affect the West due to the increase in life expectancy. Nervous cells accumulate oxidative damage, which is one of the factors that triggers and accelerates ...neurodegeneration. However, cells have mechanisms that scavenge reactive oxygen species (ROS) and alleviate oxidative stress (OS). Many of these endogenous antioxidant systems are regulated at the gene expression level by the transcription factor Nrf2 (nuclear factor erythroid 2-related factor 2). In the presence of prooxidant conditions, Nrf2 translocates to the nucleus and induces the transcription of genes containing ARE (antioxidant response element). In recent years, there has been an increase in the study of the Nrf2 pathway and the natural products that positively regulate it to reduce oxidative damage to the nervous system, both in in vitro models with neurons and microglia subjected to stress factors and in vivo models using mainly murine models. Quercetin, curcumin, anthocyanins, tea polyphenols, and other less studied phenolic compounds such as kaempferol, hesperetin, and icariin can also modulate Nrf2 by regulating several Nrf2 upstream activators. Another group of phytochemical compounds that upregulate this pathway are terpenoids, including monoterpenes (aucubin, catapol), diterpenes (ginkgolides), triterpenes (ginsenosides), and carotenoids (astaxanthin, lycopene). This review aims to update the knowledge on the influence of secondary metabolites of health interest on the activation of the Nrf2 pathway and their potential as treatments for NDs.

Understanding the pathology resulting from Staphylococcus aureus and Pseudomonas aeruginosa polymicrobial wound infections is of great importance due to their ubiquitous nature, increasing ...prevalence, growing resistance to antimicrobial agents, and ability to delay healing. Methicillin-resistant S. aureus USA300 is the leading cause of community-associated bacterial infections resulting in increased morbidity and mortality. We utilized a well-established porcine partial thickness wound healing model to study the synergistic effects of USA300 and P. aeruginosa on wound healing. Wound re-epithelialization was significantly delayed by mixed-species biofilms through suppression of keratinocyte growth factor 1. Pseudomonas showed an inhibitory effect on USA300 growth in vitro while both species co-existed in cutaneous wounds in vivo. Polymicrobial wound infection in the presence of P. aeruginosa resulted in induced expression of USA300 virulence factors Panton-Valentine leukocidin and α-hemolysin. These results provide evidence for the interaction of bacterial species within mixed-species biofilms in vivo and for the first time, the contribution of virulence factors to the severity of polymicrobial wound infections.

A group of European gas transportation companies within the European Gas Research Group launched in 2007 the ‘MOLAS’ Project to provide a software program for the analysis of the Liquefied Natural ...Gas (LNG) ageing process during ship transportation. This program contains two different modeling approaches: a physical algorithm and an ‘intelligent’ model. Both models are fed with the same input data, which is composed of the ship characteristics (BOR and capacity), voyage duration, LNG composition, temperature, pressure, and volume occupied by liquid phase at the port of origin, together with pressure at the port of destination. The results obtained are the LNG composition, temperature and liquid volume at the port of destination. Furthermore, the physical model obtains the evolution over time of such variables en route as it is based on unsteady mass balances over the system, while the i-model applies neural networks to obtain regression coefficients from historical data composed only of origin and destination measurements. This paper describes both models and validates them from previous published models and experimental data measured in ENAGAS LNG regasification plants.
