In the United States, federal agencies are required to work towards providing equal access to resources for minority and low-income populations. Access to quality public transportation is critical ...for mobility to many of these populations. Determining how transit service is distributed among vulnerable groups has the potential to significantly enhance policy analysis. While many measures of accessibility exist, due to the complexity of transit networks and the scale of the urban areas, limited research has been conducted on developing a tool to measure how equitable the distribution of transit access is in a region. This paper develops a comprehensive method to quantify the quality of service and accessibility at each transit node in a network, combined with an index to measure the inequity (concentration of quality service) at the micro scale. These measures are applied to the distribution of all residential housing units, a random sampling of units and U.S. Department of Housing and Urban Development subsidized units in Baltimore, Maryland; to determine if the subsidized housing programs are achieving major policy objectives of providing equitable transit access to vulnerable groups. The results show that transit connectivity and accessibility is distributed among some types of subsidized housing units more equitably than can be achieved by random sampling in the general population, but for other types, the distribution is less equitable; indicating some policies to enhance transit access among these units have not been effective. Evidence from this study suggests that developers of affordable housing and transportation planners should work together to find development locations that place more emphasis on transit locations with high connectivity rather than simply reducing distance to any transit.

An equitable transit system can cater to the needs of captive riders and maximize transit service coverage. Historically transit equity has not considered in planning or has been an afterthought in ...the process; leading to the underutilization of transit and encouraging travelers to be auto dependent. In this paper, the authors propose a methodology to estimate transit equity using a number of attributes such as frequency, speed, capacity, and built environment in a multi-modal transit network. We propose a methodology to measure transit equity from a graph theoretical approach for all levels of transit service coverage integrating routes, schedules, socio-economic, demographic and spatial activity patterns.

The collection of big data, as an alternative to traditional resource-intensive manual data collection approaches, has become significantly more feasible over the past decade. The availability of ...such data, coupled with more sophisticated predictive statistical techniques, has contributed to an increase in attention towards the application of these data, particularly for transportation analysis. Within the transportation literature, there is a growing emphasis on developing sources of commonly collected public transportation data into more powerful analytical tools. A commonly held belief is that application of big data to transportation problems will yield new insights previously unattainable through traditional transportation data sets. However, there exist many ambiguities related to what constitutes big data, the ethical implications of big data collection and application, and how to best utilize the emerging data sets. The existing literature exploring big data provides no clear and consistent definition. While the collection of big data has grown and its application in both research and practice continues to expand, there is a significant disparity between methods of analysis applied to such data. This paper summarizes the recent literature on sources of big data and commonly applied methods used in its application to public transportation problems. We assess predominant big data sources, most frequently studied topics, and methodologies employed. The literature suggests smart card and automated data are the two big data sources most frequently used by researchers to conduct public transit analyses. The studies reviewed indicate that big data has largely been used to understand transit users' travel behavior and to assess public transit service quality. The techniques reported in the literature largely mirror those used with smaller data sets. The application of more advanced statistical methods, commonly associated with big data, has been limited to a small number of studies. In order to fully capture the value of big data, new approaches to analysis will be necessary.

Numerous biosynthetic investigations offer a glimpse at the relative ease with which Nature is able to assemble this class of molecules, while providing synthetic chemists inspiration for the development of more efficient syntheses. Herein, we discuss the isolation and characterization, proposed fungal biogeneses, and total syntheses of epidithiodioxopiperazines.

A lytic bacteriophage (φNC10) specific to serotype O1 Yersinia ruckeri has been identified and evaluated as a model to assess the potential use of bacteriophages and their products for disease ...control in aquaculture. Electron microscopy of purified φNC10 revealed a virion particle with a small (70 nm) polyhedral head and short tail. φNC10 infected only serotype O1 strains of Y. ruckeri and failed to bind a defined Y. ruckeri mutant strain lacking O1 lipopolysaccharides (O1‐LPS), suggesting that φNC10 uses O1‐LPS as its receptor. In addition, spontaneous φNC10‐resistant mutants of Y. ruckeri exhibited defects in O1‐LPS production and were sensitive to rainbow trout serum. Purified φNC10 displayed a polysaccharide depolymerase activity capable of degrading Y. ruckeri O1‐LPS and thereby sensitizing Y. ruckeri to the bactericidal effects of rainbow trout serum. The φNC10‐associated polysaccharide depolymerase activity also reduced the ability of Y. ruckeri cells to cause mortality following intraperitoneal injection into rainbow trout. These data demonstrate a potential utility of φNC10 and its associated polysaccharide depolymerase activity for Y. ruckeri disease prevention.

Connectivity plays a crucial role as agencies at the federal and state level focus on expanding the public transit system to meet the demands of a multimodal transportation system. Transit agencies have a need to explore mechanisms to improve connectivity by improving transit service. This requires a systemic approach to develop measures that can prioritize the allocation of funding to locations that provide greater connectivity, or in some cases direct funding towards underperforming areas. The concept of connectivity is well documented in social network literature and to some extent, transportation engineering literature. However, connectivity measures have limited capability to analyze multi-modal public transportation systems which are much more complex in nature than highway networks.
In this paper, we propose measures to determine connectivity from a graph theoretical approach for all levels of transit service coverage integrating routes, schedules, socio-economic, demographic and spatial activity patterns. The objective of using connectivity as an indicator is to quantify and evaluate transit service in terms of prioritizing transit locations for funding; providing service delivery strategies, especially for areas with large multi-jurisdictional, multi-modal transit networks; providing an indicator of multi-level transit capacity for planning purposes; assessing the effectiveness and efficiency for node/stop prioritization; and making a user friendly tool to determine locations with highest connectivity while choosing transit as a mode of travel. An example problem shows how the graph theoretical approach can be used as a tool to incorporate transit specific variables in the indicator formulations and compares the advantage of the proposed approach compared to its previous counterparts. Then the proposed framework is applied to the comprehensive transit network in the Washington–Baltimore region. The proposed analysis offers reliable indicators that can be used as tools for determining the transit connectivity of a multimodal transportation network.

While both virulent and putatively avirulent Yersinia ruckeri strains exist in aquaculture environments, the relationship between the distribution of virulence-associated factors and de facto ...pathogenicity in fish remains poorly understood. Pan-genome analysis of 18 complete genomes, representing established virulent and putatively avirulent lineages of Y. ruckeri, revealed the presence of a number of accessory genetic determinants. Further investigation of 68 draft genome assemblies revealed that the distribution of certain putative virulence factors correlated well with virulence and host-specificity. The inverse-autotransporter invasin locus yrIlm was, however, the only gene present in all virulent strains, while absent in lineages regarded as avirulent. Strains known to be associated with significant mortalities in salmonid aquaculture display a combination of serotype O1-LPS and yrIlm, with the well-documented highly virulent lineages, represented by MLVA clonal complexes 1 and 2, displaying duplication of the yrIlm locus. Duplication of the yrIlm locus was further found to have evolved over time in clonal complex 1, where some modern, highly virulent isolates display up to three copies.
The LREX' prostate cancer model is resistant to the antiandrogen enzalutamide via activation of an alternative nuclear hormone receptor, glucocorticoid receptor (GR), which has similar DNA-binding ...specificity to the androgen receptor (AR). Small molecules that target DNA to interfere with protein-DNA interactions may retain activity against enzalutamide-resistant prostate cancers where ligand-binding domain antagonists are ineffective. We reported previously that a pyrrole-imidazole (Py-Im) polyamide designed to bind the consensus androgen response element half-site has antitumor activity against hormone-sensitive prostate cancer. In enzalutamide-resistant LREX' cells, Py-Im polyamide interfered with both AR- and GR-driven gene expression, whereas enzalutamide interfered with only that of AR. Genomic analyses indicated immediate interference with the AR transcriptional pathway. Long-term treatment with Py-Im polyamide demonstrated a global decrease in RNA levels consistent with inhibition of transcription. The polyamide was active against two enzalutamide-resistant xenografts with minimal toxicity. Overall, our results identify Py-Im polyamide as a promising therapeutic strategy in enzalutamide-resistant prostate cancer.

Selective breeding of animals for increased disease resistance is an effective strategy to reduce mortality in aquaculture. However, implementation of selective breeding programs is limited by an ...incomplete understanding of host resistance traits. We previously reported results of a rainbow trout selection program that demonstrated increased survival following challenge with Flavobacterium psychrophilum, the causative agent of bacterial cold water disease (BCWD). Mechanistic study of disease resistance identified a positive phenotypic correlation between post-challenge survival and spleen somatic-index (SI). Herein, we investigated the hypothesis of a genetic correlation between the two traits influenced by colocalizing QTL. We evaluated the inheritance and calculated the genetic correlation in five year-classes of odd- and even-year breeding lines. A total of 322 pedigreed families (n = 25,369 fish) were measured for disease resistance, and 251 families (n = 5,645 fish) were evaluated for SI. Spleen index was moderately heritable in both even-year (h(2) = 0.56±0.18) and odd-year (h(2) = 0.60±0.15) lines. A significant genetic correlation between SI and BCWD resistance was observed in the even-year line (rg = 0.45±0.20, P = 0.03) but not in the odd-year line (rg = 0.16±0.12, P = 0.19). Complex segregation analyses of the even-year line provided evidence of genes with major effect on SI, and a genome scan of a single family, 2008132, detected three significant QTL on chromosomes Omy19, 16 and 5, in addition to ten suggestive QTL. A separate chromosome scan for disease resistance in family 2008132 identified a significant BCWD QTL on Omy19 that was associated with time to death and percent survival. In family 2008132, Omy19 microsatellite alleles that associated with higher disease resistance also associated with increased spleen size raising the hypothesis that closely linked QTL contribute to the correlation between these traits. To our knowledge, this is the first estimation of spleen size heritability and evidence for genetic linkage with specific disease resistance in a teleost fish.

Antimicrobial resistance in Yersinia pestis is rare, yet constitutes a significant international public health and biodefense threat. In 1995, the first multidrug resistant (MDR) isolate of Y. pestis ...(strain IP275) was identified, and was shown to contain a self-transmissible plasmid (pIP1202) that conferred resistance to many of the antimicrobials recommended for plague treatment and prophylaxis. Comparative analysis of the DNA sequence of Y. pestis plasmid pIP1202 revealed a near identical IncA/C plasmid backbone that is shared by MDR plasmids isolated from Salmonella enterica serotype Newport SL254 and the fish pathogen Yersinia ruckeri YR71. The high degree of sequence identity and gene synteny between the plasmid backbones suggests recent acquisition of these plasmids from a common ancestor. In addition, the Y. pestis pIP1202-like plasmid backbone was detected in numerous MDR enterobacterial pathogens isolated from retail meat samples collected between 2002 and 2005 in the United States. Plasmid-positive strains were isolated from beef, chicken, turkey and pork, and were found in samples from the following states: California, Colorado, Connecticut, Georgia, Maryland, Minnesota, New Mexico, New York and Oregon. Our studies reveal that this common plasmid backbone is broadly disseminated among MDR zoonotic pathogens associated with agriculture. This reservoir of mobile resistance determinants has the potential to disseminate to Y. pestis and other human and zoonotic bacterial pathogens and therefore represents a significant public health concern.
