Increasingly complex metabolic pathways have been engineered by modifying natural pathways and establishing de novo pathways with enzymes from a variety of organisms. Here we apply retro-biosynthetic ...screening to a modular pathway design to identify a redox neutral, theoretically high yielding route to a branched C6 alcohol. Enzymes capable of converting natural E. coli metabolites into 4-methyl-pentanol (4MP) via coenzyme A (CoA)-dependent chemistry were taken from nine different organisms to form a ten-step de novo pathway. Selectivity for 4MP is enhanced through the use of key enzymes acting on acyl-CoA intermediates, a carboxylic acid reductase from Nocardia iowensis and an alcohol dehydrogenase from Leifsonia sp. strain S749. One implementation of the full pathway from glucose demonstrates selective carbon chain extension and acid reduction with 4MP constituting 81% (90±7 mg l(-1)) of the observed alcohol products. The highest observed 4MP titre is 192±23 mg l(-1). These results demonstrate the ability of modular pathway screening to facilitate de novo pathway engineering.
The analysis of a bimaterial medium with various notch opening angles has been carried out using boundary finite element method (BFEM) under arbitrary loading conditions. Introduced as novel method ...for stress concentration problems at geometrical discontinuities, cracks, bimaterial notches etc., the BFEM has been proved as numerically highly efficient. This has become more and more important because wedge type construction creates stress concentrations which may lead to crack initiation in many practical situations where multi-layered composite material is used, e.g. within aerospace, ship or automobile structures. So, the computational prediction of potential directions for crack initiation is essential for the knowledge of weak regions. All the analysis results are based on the hypothesis of Erdogan and Sih and have been verified by the well established finite element method. Results for potential crack initiation angles of both homogeneous and bimaterial media are presented with multiple examples of different wedge angles and different loading combinations.

We present a case of a previously well 24-year-old female patient who developed severe and life-threatening Irukandji syndrome that required ventilation and inotropic support. This case provides ...further evidence that there are jellyfish other than the Irukandji jellyfish that can cause cardiac decompensation, and there is a suggestion that application of a pressure immobilization bandage may worsen the envenomation. We include our recommended treatment for the Irukandji syndrome.
Three Indigenous Australian adults died of invasive pneumococcal disease, despite vaccination with the pneumococcal polysaccharide vaccine an average of 18 months previously.
Pneumococcal isolates ...from the patients had serotypes that are included in the vaccine. Two of the adults were alcoholics and the other had heavy recent alcohol intake. The public health implications, and possible optimal vaccination schedules to minimise vaccine failures, are discussed.

We describe an outbreak of hepatitis A that occurred in an Intensive Care Unit (ICU) in a regional hospital in North Queensland. Seven people were infected including two patients, two close contacts ...of the index patient and three ICU nursing staff. The index case was admitted with an overdose and multiple trauma; he was not suspected to be incubating hepatitis A. The outbreak was initiated as a result of inadequate precautions taken whilst handling the index patient's bile. Problems identified upon reviewing the outbreak were inadequate terminal cleaning of equipment, food consumption in the ICU and inadequate handwashing practices. Implementation and maintenance of standard infection control practices is vital if further outbreaks of hospital-acquired hepatitis A and other enteric infections are to be avoided. We also suggest that the inactivated hepatitis A vaccine be considered for ICU staff.
The recent explosion in capability of embedded and portable electronics has not been matched by battery technology. The slow growth of battery energy density has limited device lifetime and added ...weight and volume. Passive energy harvesting from solar radiation, thermal sources, or mechanical vibration has potentially wide application in wearable and embedded sensors to complement batteries. The amount of energy from harvesting is typically small and highly variable, requiring circuits and architectures which are low power and can scale their power consumption with user requirements and available energy. We describe several circuit techniques for achieving these goals in signal processing applications for wireless sensor network nodes such as using Distributed Arithmetic to implement energy scalable signal processing algorithms. In addition, we propose increasing vibration energy harvesting efficiency by eliminating AC/DC conversion electronics, and have investigated self-timed circuits, power-on-reset circuitry, and memory for energy harvesting AC power supplies. These techniques can also be applied to energy harvesting from other sources. A chip will be fabricated to test the proposed circuits.
Severe ovarian hyperstimulation syndrome as a result of assisted reproductive therapy occurs rarely. However, this iatrogenic condition can result in a life threatening illness with difficult ...management dilemmas for the attending physicians. A patient with severe adult respiratory distress syndrome and septicaemia after in vitro fertilization required prolonged intensive care treatment and subsequently had a probable ectopic pregnancy treated with systemic methotrexate as an alternative to surgical management. A satisfactory outcome was obtained, followed by a spontaneous successful pregnancy some months after these events.
