Animal studies suggest that prenatal exposure to diethylstilbestrol (DES) causes epigenetic alterations in primordial germ cells that affect the next generation, but human studies are sparse.
We assessed hormonally mediated outcomes in third generation women whose mothers were prenatally DES-exposed and unexposed.
Compared to the unexposed, DES-exposed third generation women had an increased risk of irregular menses and amenorrhea; the respective prevalence ratios and 95% confidence intervals (CI) in follow-up data were 1.32 (95% CI: 1.10, 1.60) and 1.26 (95% CI: 1.06, 1.49); associations were more apparent in third generation women whose prenatally DES-exposed mothers were affected by vaginal epithelial changes. The follow-up data also indicated an association with preterm delivery (relative risk (RR): 1.54; 95% CI: 1.35, 1.75).
DES third generation women may have an increased risk of irregular menstrual cycles, amenorrhea, and preterm delivery, consistent with inter-generational effects of endocrine disrupting chemical exposure in humans.

Sex-linkage is predicted to evolve in response to sex-specific or sexually antagonistic selection. In line with this prediction, most sex-linked genes are associated with reproduction in the ...respective sex. In addition to traits directly involved in fertility and fecundity, mediators of maternal effects may be predisposed to evolve sex-linkage, because they indirectly affect female fitness through their effect on offspring phenotype. Here, we test for sex-linked inheritance of a key mediator of prenatal maternal effects in oviparous species, the transfer of maternally derived testosterone to the eggs. Consistent with maternal inheritance, we found that in Japanese quail (Coturnix japonica) granddaughters resemble their maternal (but not their paternal) grandmother in yolk testosterone deposition. This pattern of resemblance was not due to non-genetic priming effects of testosterone exposure during prenatal development, as an experimental manipulation of yolk testosterone levels did not affect the females' testosterone transfer to their own eggs later in life. Instead, W chromosome and/or mitochondrial variation may underlie the observed matrilineal inheritance pattern. Ultimately, the inheritance of mediators of maternal effects along the maternal line will allow for a fast and direct response to female-specific selection, thereby affecting the dynamics of evolutionary processes mediated by maternal effects.

Amyloid fibrils commonly exhibit multiple distinct morphologies in electron microscope and atomic force microscope images, often within a single image field. By using electron microscopy and ...solid-state nuclear magnetic resonance measurements on fibrils formed by the 40-residue$\beta-amyloid$peptide of Alzheimer's disease (Aβ1-40), we show that different fibril morphologies have different underlying molecular structures, that the predominant structure can be controlled by subtle variations in fibril growth conditions, and that both morphology and molecular structure are self-propagating when fibrils grow from preformed seeds. Different Aβ1-40fibril morphologies also have significantly different toxicities in neuronal cell cultures. These results have implications for the mechanism of amyloid formation, the phenomenon of strains in prion diseases, the role of amyloid fibrils in amyloid diseases, and the development of amyloid-based nanomaterials.

To date, vaginal/cervical clear cell adenocarcinoma (CCAC) has not been reported in the granddaughters of women treated with diethylstilbestrol (DES) during pregnancy. We present an 8-year-old girl ...with a history of severe vaginal bleeding who was diagnosed with cervical CCAC. She underwent fertility-sparing surgery and radiotherapy. No sign of recurrence was detected throughout a 10-year follow-up. Her grandmother had received DES therapy during pregnancy with the patient's mother. Although no direct causal link is demonstrated, this case raises for the first time, the hypothesis of multigenerational effects of DES in girls and strongly suggests the need to follow the granddaughters of DES-treated women.
The results of studies of the exterior characteristics of first-calf cows and their descendants of different generations are presented. The studies were conducted at high-producing herds (average ...yields per a cow about 10000 kg) LLC “Veleten” of the Glukhiv district of Sumy region and PJSC “Plemzavod “Stepnoy” of the Kamyansko-Dniprovskyy district of Zaporizhzhya region created out of the absorption crossing of the Ukrainian Black-and-White dairy breed females with Holstein breeders, first-calf cows and their descendants of the first, second, and at PJSC “Plemzavod “Stepnoy” – also of third generations. The sample of animals includes 62.5–100% share of Holstein inheritance. There was some differentiation between the cows of the controlled herds according to the investigated body measurements: the first-calf cows of PJSC “Plemzavod “Stepnoy” were highly dominated the herdmates of LLC “Veleten” by height at the withers by 3.2, chest depth by 1.4, chest girth by the shoulder by 1.2 and oblique body length – by 1.7 cm, but were inferior to them in chest width by 0.8 (P < 0.05), width of oysters – by 0.4 and girth of wrist – by 0.3 cm (P < 0.001). The cows of both farms, by all studied exterior characteristics, outweighed their descendants of different generations, which is explained by the lower age of first calving for daughters, grandchildren and great-grandchildren than the cows from which they came. However, such a slight, although in most cases, probable differentiation between the exterior features of the cows and their descendants of the above generations appears to be the result of a better level of animal production of each successive generation, as indicated by their milk yields for the first lactation. However, according to all the investigated measurements (exception – width of oysters of cows of all generations and girth of wrist in the descendants of the second and third generations) animals of both controlled herds outperformed the target parameters of exterior characteristics for the first-calf cows of the desired type of Ukrainian Black-and-White dairy breed. The analysis of body structure indices shows that the first-calf cows under control and their descendants of different generations were characterized by the type of body structure characteristic of dairy cattle. Between the body measurements of animals and their descendants of different generations, different strengths and directions of communication are established. The correlation coefficients between the exterior characteristics of cows and their daughters in LLC “Veleten”, depending on the size, were in the range -0.05 – +0.15, in cows and their granddaughters, within -0.15 – +0.06, and the regression coefficients are within -0.05 – +0.19 and -0.15 – +0.05, respectively. In PJSC “Plemzavod “Stepnoy” the relative variability of body measurements in cows and their descendants of the first, second and third generations was -0.07 – +0.23, respectively; -0.19 – +0.14 and -0.33 – +0.26, and regression coefficients ranged from -0.04 to +0.14; from -0.17 to +0.07 and from -0.47 to +0.16. It should be noted that in both farms the correlation coefficients between the exterior features of animals of different generations were by no means probable, and the regression coefficients were only reliable between the chest depth behind the shoulders of cows and their daughters in LLC “Velyten” and between heights in the cows' hedge and their great-grandchildren at PJSC “Plemzavod “Stepnoy”.

The research was carried out on cows of Ukrainian Black-and-White Dairy breed and their descendants of the first and second generations on the conditions of LLC “Veletenj” in the Glukhiv district of ...the Sumy region. In cows, based on a retrospective analysis of data and the results of their own searches, was studied the age dynamics of reproductive capacity of cows for the first-seventh and higher lactation, and their daughters and granddaughters for the first and higher lactation. It has been established that reproductive ability of cows to some extent is due to their age. Depending on the lactation, the duration of the parturition of the cows of LLC “Veletenj” varied from 278.2 to 280.0, the duration of the indifference period – from 64.4 to 69.0 days, service period – from 123.5 to 136.4, inter-calving period – from 402.2 to 414.8 and in the dry stable period – from 61.3 to 75.8 days, the reproduction rate was 0.91–0.93, the calves output for 100 cows was 90.9–93.0 the index of insemination is 2.41–2.56 and the fertility index is 42.2–44.8. The results of our research indicate that each subsequent generation of cows was characterized by a better reproductive ability, which, in our opinion, is due to the timely abandonment of fetuses and stimulation of infertile cows, as well as an improved level of feeding. The difference in the duration of the service period for the first lactation between the mothers and their daughters was 24.5, for the best lactation – 24.8, for the duration of the interotional period – 24.8 and 23.7 days, respectively, at P < 0.001 in all cases. A more significant difference in reproductive performance was observed between cows and their granddaughters. In terms of the duration of the service period for the first and best lactation, it was 15.5 (P < 0.05) and 33.3 (P < 0,001) and interotional period – 17.3 (P < 0.05) and 33,2 days (P < 0.001) respectively. Between the majority of the studied reproductive abilities of mothers and daughters for the first and higher lactation, the relative variability was positive and reliable, and mothers and granddaughters – low uncertainty and mostly negative (exception – the duration of calving). The highest positive correlation coefficients were observed between the duration of cervix and their daughters (r = 0.113 ± 0.039 at P < 0.001) and granddaughters (r = 0.211 ± 0.088 at P < 0.05) for the first lactation, as well as mothers and daughters for higher lactation (r = 0.114 ± 0.039 at P < 0.01). Between indicators of the indifference period (r = -0.036–0.051) and the index of fertility of cows (r = -0.080–0.014) and their descendants for the lactation under investigation have low, multidirectional, unreliable connections. The influence of mothers on indicators of reproductive function of descendants is established. They made the most significant impact on the duration of service and inter calving periods, reproduction rate, calving out of 100 cows and the index of daughter insemination (ηx2 = 27.57–52.22%). For higher lactation, this effect was significantly lower (exception – the index of insemination) – ηx2 = 15,39–28,10%.
