Lipoprotein peroxidation, especially the modification of apolipoprotein B-100, has been implicated to play an important role in the pathogenesis of atherosclerosis. However, there have been few ...detailed insights into the chemical mechanism of derivatization of apolipoproteins during oxidation. In the present study, we provide evidence that the formation of the toxic pollutant acrolein (CH2=CH-CHO) and its conjugate with lysine residues is involved in the oxidative modification of human low density lipoprotein (LDL). Upon incubation with LDL, acrolein preferentially reacted with lysine residues. To determine the structure of acrolein-lysine adduct in protein, the reaction of acrolein with a lysine derivative was carried out. Employing Nalpha-acetyllysine, we detected a single product, which was identified to be a novel acrolein-lysine adduct, Nalpha-acetyl-Nepsilon-(3-formyl-3,4-dehydropiperidino )lysine. The acid hydrolysis of the adduct led to the derivative that was detectable with amino acid analysis. It was revealed that, upon in vitro incubation of LDL with acrolein, the lysine residues that had disappeared were partially recovered by Nepsilon-(3-formyl-3, 4-dehydropiperidino)lysine. In addition, we found that the same derivative was detected in the oxidatively modified LDL with Cu2+ and that the adduct formation was correlated with LDL peroxidation assessed by the consumption of alpha-tocopherol and cholesteryl ester and the concomitant formation of cholesteryl ester hydroperoxide. Enzyme-linked immunosorbent assay that measures free acrolein revealed that a considerable amount of acrolein was released from the Cu2+-oxidized LDL. Furthermore, metal-catalyzed oxidation of arachidonate was associated with the formation of acrolein, indicating that polyunsaturated fatty acids including arachidonate represent potential sources of acrolein generated during the peroxidation of LDL. These results indicate that acrolein is not just a pollutant but also a lipid peroxidation product that could be ubiquitously generated in biological systems.

Lysine requirements for gain in maternal body reserves and piglet birth weight, during pregnancy, in contemporary prolific genotypes, are not well established. This study aimed to evaluate the effect ...of dietary lysine in late pregnancy on piglet birth weight, and on the gestational and lactational performance of gilts. Pregnant gilts were uniformly distributed into two groups and received, from 85 to 110 days of gestation, either of two lysine levels in their diet: Control group - 28g lysine/day (n=136), and Lysine group - 35g lysine/day (n=141). There were no effects (P>0.10) of supplemental lysine on body weight and backfat (BF) gain of females or on piglet birth weight. Gilts supplemented with lysine tended to have a lower percentage of stillbirths (P=0.077), reduced within-litter birth weight variation (P=0.094) and a lower percentage of piglets weighing less than 1100g (P=0.082) than in the Control group. During lactation, the performance of sows and litters was also evaluated in a subgroup of sows (n=26/group). There were no differences between the Control and Lysine groups (P>0.10) in voluntary feed intake, body reserve losses (weight and BF), weaning-to-estrus interval of the sows, and litter weaning weight. In conclusion, an increase in lysine (from 28 to 35g/day) in late gestation of gilts (85 to 110 days) tends to reduce the rate of stillbirths and to improve the uniformity of litter weight at birth, but does not affect the performance of females until farrowing or during subsequent lactation.
As exigências de lisina durante a gestação, para o ganho de reservas corporais maternas e peso ao nascer dos leitões, não estão bem estabelecidas nos genótipos contemporâneos prolíficos. Este estudo objetivou avaliar o efeito da suplementação de lisina no terço final da gestação, sobre o peso ao nascer dos leitões e o desempenho gestacional e lactacional de leitoas. Leitoas prenhes foram uniformemente distribuídas em dois grupos que receberam, dos 85 aos 110 dias de gestação, diferentes níveis de lisina na dieta: Grupo Controle - 28g lisina/dia (n=136), e Grupo Lisina - 35g lisina/dia (n=141). Não houve efeito (P>0,10) da suplementação de lisina no ganho de peso e espessura de toucinho (ET) das fêmeas e no peso dos leitões ao nascer. Leitoas suplementadas com lisina tenderam a ter menor percentagem de natimortos (P=0,077), menor variação de peso dentro da leitegada (P= 0,094) e menor percentagem de leitões pesando menos que 1100g (P=0,082), em comparação ao grupo controle. Durante a lactação, o desempenho das fêmeas e das leitegadas foi avaliado em um subgrupo de fêmeas (n=26/grupo). Não houve diferença entre os grupos Controle e Lisina (P>0,10) no consumo voluntário de ração, perdas de reservas corporais (peso e ET), intervalo desmame-estro das fêmeas e peso da leitegada ao desmame. Em conclusão, um incremento de lisina (de 28 para 35g dia-1) no terço final da gestação de leitoas (85 até 110 dias) tende a reduzir o número de natimortos e aumentar a uniformidade do peso dos leitões ao nascer, mas não afeta o desempenho das fêmeas até o parto ou na fase lactacional subsequente.

Objetivou-se por meio deste estudo determinar a necessidade nutricional de lisina digestível em rações para juvenis de tilápia-do-nilo (Oreochromis niloticus). Setecentos e vinte peixes ...masculinizados (7,30±0,11g) foram alimentados durante 30 dias com oito rações (26,81% de proteína digestível e 3090kcal/kg de energia digestível da ração) contendo teores crescentes de lisina digestível (1,24; 1,36; 1,48; 1,60; 1,72; 1,84; 1,96 e 2,08%). As tilápias foram distribuídas em delineamento inteiramente ao acaso, com oito tratamentos, seis repetições e 15 peixes por unidade experimental. Foram avaliadas variáveis de desempenho (ganho de peso, taxa de crescimento específico, taxa de sobrevivência, consumo de ração, consumo de lisina digestível, conversão alimentar aparente, eficiência proteica para ganho, eficiência de lisina para ganho e eficiência de retenção de nitrogênio) e de composição corporal (teores de umidade, gordura, proteína, matéria mineral corporal e as taxas de deposição diária de proteína e gordura corporais). A elevação do teor de lisina digestível na ração não influenciou (P>0,05) o consumo de ração, a taxa de sobrevivência e os teores de umidade e de matéria mineral corporal, mas melhorou de forma quadrática (P<0,05) os demais parâmetros avaliados, com exceção do consumo de lisina e da eficiência de lisina para ganho, que aumentou e reduziu, respectivamente, de forma linear (P<0,05). Recomenda-se que rações para juvenis de tilápia-do-nilo devam conter 1,84% de lisina digestível para máximo ganho de peso.
Determination of animal profile from production scenarios by modeling is essential to improve production. The objective of this study was to model and evaluate the performance, the supply and ...nutrients requirements for pigs, during the growing and finishing phases, in real production scenarios. Six scenarios with 2,200 animals, which consumed six ad libitum diets, were selected. The collected data from the production scenarios were modeled based on the average animal. Animals were housed at 65±7 days old and weighing 22.11±1.41kg and were slaughtered at 159±10 days old and weighing 121.18±7kg. Average of the scenarios was greater than 0.27kg for consumption and 0.12kg for weight gain; feed conversion was equivalent to the standard animal profile (SAP). Scenarios were 1.60g higher for the requirement and 2.67g higher for daily digestible lysine per animal when compared to the SAP. Production scenarios showed differences between performance, supply and nutrient requirements for pigs during the growing and finishing phases. Modeling is a tool that can be used to describe and compare the characteristics of each production scenario.
A determinação do perfil animal de cenários de produção, por modelagem, é imprescindível para melhorar a produção. O objetivo deste estudo foi modelar e avaliar o desempenho zootécnico, o fornecimento e as exigências de nutrientes para suínos nas fases de crescimento e terminação em cenários de produção reais. Foram selecionados seis cenários com total de 2.200 animais que consumiram seis dietas ad libitum. Os dados coletados nos cenários de produção foram modelados com base na média animal. Os animais foram alojados com 65±7 dias de idade e 22,11±1,41kg de peso e foram abatidos com 159±10 dias e 121,18±7kg. A média dos cenários foi superior a 0,27kg em consumo e 0,12kg em ganho de peso, a conversão alimentar foi equivalente ao perfil animal padrão (PAP). Os cenários foram superiores em 1,60g para exigência e 2,67g para fornecimento de lisina digestível diária por animal em relação ao PAP. Os cenários de produção apresentam diferenças para desempenho zootécnico, fornecimento e exigências de nutrientes para suínos nas fases de crescimento e terminação. A modelagem é uma ferramenta que pode ser utilizada para descrever e comparar as especificidades em cada cenário de produção.

Bovine β-lactoglobulin (β-Lg) has been studied extensively in both the isolated and the naturally occurring states. It is a commercially important whey protein of obvious nutritional value but, so ...far, one that has no clearly identified biological function. In common with many of the other members of the lipocalin family to which it belongs, β-Lg binds hydrophobic ligands, and it appears possible that there are at least two distinct binding sites per monomer for a variety of ligands. By comparison with other members of the family, there is a probable binding site in the central cavity of the molecule that is formed by the eight antiparallel β-strands that are typical of the lipocalins. We have now cocrystallized β-Lg with palmitic acid, and the refined structure (R = 0.204, Rfree = 0.240 for 6,888 reflections to 2.5-Å resolution) reveals that the ligand binds in the central cavity in a manner similar to the binding of retinol to the related lipocalin, serum retinol-binding protein. The carboxyl group binds to both Lys-60 and Lys-69 at the entrance to the cavity. The hydrophobic tail stretches in an almost fully extended conformation into the center of the protein. This is the first direct observation of a ligand binding to β-Lg.

Background: Ractopamine (RAC) supplementation in the feed has been evaluated as a strategy to increaseproductive efficiency in finishing pigs. Objective: To evaluate the effects of different RAC ...dietary levels on performance, carcass traits, efficiency of lysine (ELU) and energy (EEU) utilization, and economic viability in finishing pig. Methods: A total of 40 barrows (74.75 ± 5.22 kg) were fed four RAC levels (0, 5, 10 and 5-10 mg/kg step-up program) from 0-14, 15-31 and 0-31 days. Performance, carcass characteristics, ELU, EEU, cost per unit of weight gain (CWG), payment and profit parameters were measured. The animals were distributed in a completely randomized design in four treatments, with ten replicates per treatment. The experimental unit was each animal. Results: Pigs fed RAC diets showed increased body weights at 14 and 31 days, average daily gain (ADG) at 0–14 and 0–31 days, ELU at 0-14 days, and hot carcass weight as compared with those fed the control diet. The step-up program as compared to the 10 mg/kg RAC concentration resulted in increased body weight, feed/gain ratio (FGR), ADG, ELU, EEU and CWG at 0–14 days. Payment by weight and bonus payment were better for treatments with RAC as compared to control. Conclusions: Pigs fed RAC improved performance, carcass weight, ELU, EEU and economic viability. The results were better for the step-up program compared with the intermittent use of 10 mg/kg RAC.

El objetivo del trabajo fue evaluar dos niveles de lisina y treonina digestibles en cerdos en crecimiento, para analizar el comportamiento productivo, características de la canal y la concentración ...de urea en plasma. En el experimento 1 se utilizaron 36 cerdos con peso promedio inicial de 27.65 ± 5.65 kg durante cuatro semanas, distribuidos en un diseño completamente al azar con arreglo factorial 2x2, dos concentraciones de lisina: 0.83 y 0.93 % y treonina: 05.52 y 0.62 %, respectivamente. Con los datos se realizó un ANOVA y las medias de los tratamientos se compararon con la prueba de Tukey. No hubo efecto por la concentración de lisina y treonina en las variables (p > 0.05). En el experimento 2 se utilizaron 35 cerdos con peso promedio inicial de 16.71 ± 7.85 kg durante seis semanas para evaluar cinco niveles de treonina (0.52, 0.62, 0.72, 0.82 y 0.92 %), en un diseño completamente al azar. Se utilizaron contrastes ortogonales para detectar efectos lineales, cuadráticos y cúbicos para determinar el nivel óptimo biológico (NOB) para cada variable. El NOB para ganancia de peso y consumo de alimento fue 0.90 % de treonina digestible. En el experimento 1 y 2 la concentración de urea en plasma se modificó por la cantidad de lisina y treonina presentes en la dieta (p < 0.05). Se concluye que el nivel más adecuado de lisina digestible para dietas es de 0.83 % y de treonina digestible de 0.90 %.

Este estudo foi realizado com o objetivo de determinar a exigência de lisina digestível para juvenis de tilápia-do-nilo. Noventa e seis peixes revertidos sexualmente e com peso vivo inicial de 5,72 ...± 0,10 g foram alimentados por 42 dias com quatro rações (29.51% de PB e 3.235 kcal de energia digestível/kg) contendo diferentes níveis de L-lisina HCl (0,1; 0,3; 0,5 e 0,7%, correspondendo a rações com 1,04; 1,27; 1,51 e 1,741% de lisina digestível, respectivamente). Os peixes foram distribuídos em 16 tanques com capacidade de 1.000 L cada um, em um delineamento inteiramente casualizado, com quatro tratamentos, três repetições e seis peixes por unidade experimental. Não foi observado efeito dos níveis de lisina sobre o índice hepato-somático, a gordura visceral, a sobrevivência e os teores de água e extrato etéreo na carcaça. A análise de Linear Response Plateau do ganho de peso e da conversão alimentar versus níveis de lisina digestível demonstrou estimativa de exigência de 1,56 e 1,44% de lisina digestível, respectivamente. Foi observado efeito quadrático sobre o rendimento de carcaça, que aumentou até o nível de 1,61% de lisina digestível. Considerando o desempenho, recomenda-se que dietas para juvenis de tilápia-do-nilo devem conter 1,44% de lisina digestível (5,23% da proteína da dieta).
This trial was carried out to determine the digestible lysine requirements of Nile tilapia juveniles. Ninety- six reverted fishes averaging initial weight of 5.72 ± 0.10 g were fed four diets (29.51% of crude protein and 3,235 kcal/kg of digestible energy) containing increasing levels of L-lysine HCl (0.1, 0.3, 0.5, and 0.7%), during 42 days. The following treatments were evaluated: dietary lysine levels of 1.04; 1.27; 1.51 and 1.741%. Fishes were allotted to 1000-L 16 tanks, as a completely randomized design with four treatments, three replicates and eight fishes per experimental unit. No treatment effects on hepatosomatic index, visceral fat, survival rate and water ether extract in the carcass were observed. The Linear Response Plateau analysis of weight gain and feed:gain ratio versus digestible lysine level yielded an estimated digestible lysine requirement of 1.56 and 1.44% for Nile tilapia juveniles, respectively. Quadratic effect of treatments on carcass yield that increased up to 1.61% of digestible lysine was observed. Considering the performance, it is recommended diets containing up to 1.44% of digestible lysine (5.23% of protein) for Nile tilapia juveniles.
PURPOSE: To determine the effects of green propolis extracted in L-lysine (WSDP) and of L- lysine for 40 weeks on induced rat bladder carcinogenesis. METHODS: The animals (groups I, II, III, IV, V ...and VI) received BBN during 14 weeks. Group I was treated with propolis 30 days prior received BBN, and then these animals were treated daily with propolis; Groups II and III was treated with subcutaneous and oral propolis (respectively) concurrently with BBN. The animals of Group IV were treated L-lysine; Group V received water subcutaneous; and Group VI received only to BBN. Among the animals not submitted to carcinogenesis induction, Group VII received propolis, Group VIII received L-lysine and Group IX received water. RESULTS: The carcinoma incidence in Group I was lower than that of control (Group VI). The carcinoma multiplicity in Group IV was greater than in Group VI. All animals treated with L-lysine developed carcinomas, and they were also more invasive in Group IV than in controls. On the other hand, Group VIII showed no bladder lesions. CONCLUSION: The WSDP is chemopreventive against rat bladder carcinogenesis, if administered 30 days prior to BBN , and that L-lysine causes promotion of bladder carcinogenesis.OBJETIVO: Determinar os efeitos da própolis verde extraída em L - Lisina (WSDP) e da L-Lisina por 40 semanas em ratos induzidos a carcinogênese de bexiga. MÉTODOS: Os animais (grupos I, II, III, IV, V e VI) receberam BBN por 14 semanas. O grupo I foi tratado com própolis 30 dias antes de receber BBN e em seguida estes animais foram tratados diariamente com própolis; Os grupos II e III foram tratados com própolis subcutânea e oral (respectivamente) e concorretemente com BBN. Os animais do grupo IV foram tratados com L- Lisina; o grupo V recebeu água subcutânea; o grupo VI recebeu apenas BBN. Entre os animais não submetidos a indução de carcinogênese, Grupo VII, receberam própolis, Grupo VIII, receberam L-Lisina e Grupo IX receberam água. RESULTADOS: A incidência de carcinoma no grupo I foi menor que no grupo controle (grupo IV) A multiplicidade de carcinoma no grupo IV foi maior que no grupo VI. Todos os animais tratados com L - Lisina desenvolveram carcinomas e estes foram mais invasivos no grupo IV que no grupo controle. Por outro lado o grupo VIII não apresentou lesões. CONCLUSÃO: WSDP é quimiopreventiva contra a carcinogese de bexiga se administrada 30 dias antes do início do BBN, e a L - Lisina causa promoção da carcinogênese de bexiga.
