In the latest trend of anticancer chemotherapy research, there were many macromolecular anticancer drugs developed based on enhanced permeability and retention (EPR) effect, such as albumin bound ...paclitaxel nanoparticle (nab- PTX, also called Abraxane®). However, cancers with low vascular permeability posed a challenge for these EPR based therapeutic systems. Augmenting the intrinsic EPR effect with an intrinsic vascular modulator such as nitric oxide (NO) could be a promising strategy. S-nitrosated human serum albumin dimer (SNO-HSA Dimer) shown promising activity previously was evaluated for the synergistic effect when used as a pretreatment agent in nab-PTX therapy against various tumor models. In the high vascular permeability C26 murine colon cancer subcutaneous inoculation model, SNO-HSA Dimer enhanced tumor selectivity of nab-PTX, and attenuated myelosuppression. SNO-HSA Dimer also augmented the tumor growth inhibition of nab-PTX in low vascular permeability B16 murine melanoma subcutaneous inoculation model. Furthermore, nab-PTX therapy combined with SNO-HSA Dimer showed higher antitumor activity and improved survival rate of SUIT2 human pancreatic cancer orthotopic model. In conclusion, SNO-HSA Dimer could enhance the therapeutic effect of nab-PTX even in low vascular permeability or intractable pancreatic cancers. The possible underlying mechanisms of action of SNO-HSA Dimer were discussed.

Hypoalbuminemia is associated with inflammation. Despite being addressed repeatedly in the literature, there is still confusion regarding its pathogenesis and clinical significance. Inflammation ...increases capillary permeability and escape of serum albumin, leading to expansion of interstitial space and increasing the distribution volume of albumin. The half‐life of albumin has been shown to shorten, decreasing total albumin mass. These 2 factors lead to hypoalbuminemia despite increased fractional synthesis rates in plasma. Hypoalbuminemia, therefore, results from and reflects the inflammatory state, which interferes with adequate responses to events like surgery or chemotherapy, and is associated with poor quality of life and reduced longevity. Increasing or decreasing serum albumin levels are adequate indicators, respectively, of improvement or deterioration of the clinical state. In the interstitium, albumin acts as the main extracellular scavenger, antioxidative agent, and as supplier of amino acids for cell and matrix synthesis. Albumin infusion has not been shown to diminish fluid requirements, infection rates, and mortality in the intensive care unit, which may imply that there is no body deficit or that the quality of albumin “from the shelf” is unsuitable to play scavenging and antioxidative roles. Management of hypoalbuminaemia should be based on correcting the causes of ongoing inflammation rather than infusion of albumin. After the age of 30 years, muscle mass and function slowly decrease, but this loss is accelerated by comorbidity and associated with decreasing serum albumin levels. Nutrition support cannot fully prevent, but slows down, this chain of events, especially when combined with physical exercise.

Human serum albumin (HSA), the most abundant protein in plasma, is a monomeric multi-domain macromolecule, representing the main determinant of plasma oncotic pressure and the main modulator of fluid ...distribution between body compartments. HSA displays an extraordinary ligand binding capacity, providing a depot and carrier for many endogenous and exogenous compounds. Indeed, HSA represents the main carrier for fatty acids, affects pharmacokinetics of many drugs, provides the metabolic modification of some ligands, renders potential toxins harmless, accounts for most of the anti-oxidant capacity of human plasma, and displays (pseudo-)enzymatic properties. HSA is a valuable biomarker of many diseases, including cancer, rheumatoid arthritis, ischemia, post-menopausal obesity, severe acute graft-versus-host disease, and diseases that need monitoring of the glycemic control. Moreover, HSA is widely used clinically to treat several diseases, including hypovolemia, shock, burns, surgical blood loss, trauma, hemorrhage, cardiopulmonary bypass, acute respiratory distress syndrome, hemodialysis, acute liver failure, chronic liver disease, nutrition support, resuscitation, and hypoalbuminemia. Recently, biotechnological applications of HSA, including implantable biomaterials, surgical adhesives and sealants, biochromatography, ligand trapping, and fusion proteins, have been reported. Here, genetic, biochemical, biomedical, and biotechnological aspects of HSA are reviewed.

There is a considerable current interest regarding specificity of the commonly used dyes that are used in a clinic biochemistry laboratory to estimate human serum albumin. It is in this context we ...have separated human serum albumin from left over plasma samples collected from human volunteers on Native PAGE followed by staining of albumin with various dyes with and without incubation of ester substrates prior to staining. We have observed that dyes like bromophenol blue is not specific for albumin and the observation with various Fast dyes are dye specific. Based on our result we feel that specific staining of albumin on gel is a candidate method for checking the specificity of the dyes for albumin.

Adsorption of human serum albumin (HSA), recombinant HSA (rHSA) and the albumin dimer (dHSA) at solid/electrolyte interfaces is ...reviewed with the emphasis put on quantitative analysis of this process. Initially, various physicochemical data characterizing bulk properties of albumin molecules are discussed such as electrophoretic mobility, electrokinetic charge, zeta potential and diffusion coefficient. Adsorption kinetics of HSA, rHSA and dHSA at mica derived from AFM, streaming potential and XPS measurements is analyzed. Maximum coverages of irreversibly adsorbed molecules under various ionic strengths and pHs are quantitatively interpreted in terms of the random sequential adsorption model. Thorough acid-basic characteristic of albumin monolayers of well-controlled coverage are also presented. The results derived from the colloid deposition method that unveil albumin molecule orientation and charge distribution are discussed and interpreted in terms of the random site theory. Subsequently, adsorption of albumins at negatively and positively charged polymeric microparticles studied by the electrokinetic and the AFM aided concentration depletion methods is analyzed. These results are theoretically interpreted by applying the bead model of HSA and dHSA molecules. Orientation of adsorbed molecules and the stability of albumin monolayers in respect to pH cyclic changes are discussed. A universal, electrostatic interaction driven, mechanism of albumin adsorption at macroscopic surfaces and polymer microparticles is confirmed.

Human serum albumin (HSA) is characterized by 17 disulfides and by only one unpaired cysteine (Cys34), which can be free in the reduced albumin or linked as a mixed disulfide with cysteine, or in ...minor amount with other natural thiols, in the oxidized albumin. In healthy subjects, the level of the oxidized form is about 35%, but it rises up to 70% after oxidative insults or in patients with kidney diseases. Oxidized albumin is therefore considered a short‐term biomarker of oxidative stress as its level may increase or decrease under appropriate redox inputs in discrete temporal spans. This paper defines, for the first time, the kinetic properties of reduced and oxidized Cys34 of HSA in their reactions with natural disulfides and thiols. Kinetic constants support the evidence that the Cys34 redox oscillations observed in vivo are mainly due to the interaction with cysteine and cystine without the involvement of any enzymatic support. This study gives also a plausible explanation for the absence of involvement of the 17 disulfides naturally present in HSA in these redox transitions. This inert behavior toward cysteine is marginally due to solvent accessibility or flexibility factors of these bonds but mainly to their strong thermodynamic stability, which is caused essentially by a proximity effect. A similar mechanism is likely at play in the many proteins that maintain disulfide bridges in a reducing medium like the cytosol.

Serum albumin (SA) is the most abundant plasma protein in mammals. SA is a multifunctional protein with extraordinary ligand binding capacity, making it a transporter molecule for a diverse range of metabolites, drugs, nutrients, metals and other molecules. Due to its ligand binding properties, albumins have wide clinical, pharmaceutical, and biochemical applications. Albumins are also allergenic, and exhibit a high degree of cross-reactivity due to significant sequence and structure similarity of SAs from different organisms. Here we present crystal structures of albumins from cattle (BSA), horse (ESA) and rabbit (RSA) sera. The structural data are correlated with the results of immunological studies of SAs. We also analyze the conservation or divergence of structures and sequences of SAs in the context of their potential allergenicity and cross-reactivity. In addition, we identified a previously uncharacterized ligand binding site in the structure of RSA, and calcium binding sites in the structure of BSA, which is the first serum albumin structure to contain metal ions.

Intrapulmonary arteriovenous anastomoses (IPAVs) are large diameter connections that allow blood to bypass the lung capillaries and may provide a route for right-to-left embolus transmission. These ...anastomoses are recruited by exercise and catecholamines and hypoxia. Yet, whether IPAVs are recruited via direct, oxygen sensitive regulatory mechanisms or indirect effects secondary to redistribution pulmonary blood flow is unknown. Here, we hypothesized that the addition of exercise to hypoxic gas breathing, which increases cardiac output, would augment IPAVs recruitment in healthy humans. To test this hypothesis, we measured the transpulmonary passage of .sup.99m Tc-macroaggregated albumin particles (.sup.99m Tc-MAA) in seven healthy volunteers, at rest and with exercise at 85% of volitional max, with normoxic (FIO.sub.2 = 0.21) and hypoxic (FIO.sub.2 = 0.10) gas breathing. We found increased .sup.99m Tc-MAA passage in both exercise conditions and resting hypoxia. However, contrary to our hypothesis, we found the greatest .sup.99m Tc-MAA passage with resting hypoxia. As an additional, secondary endpoint, we also noted that the transpulmonary passage of .sup.99m Tc-MAA was well-correlated with the alveolar-arterial oxygen difference (A-aDO.sub.2) during exercise. While increased cardiac output has been proposed as an important modulator of IPAVs recruitment, we provide evidence that the modulation of blood flow through these pathways is more complex and that increasing cardiac output does not necessarily increase IPAVs recruitment. As we discuss, our data suggest that the resistance downstream of IPAVs is an important determinant of their perfusion.

As vital important bioactive species, human serum albumin (HSA) and sulfur dioxide (SO2) are essential molecules in the organisms and act a pivotal part in many biological events. Although studies ...have shown that SO2-induced HSA radicals can cause oxidative damage, the underlying mechanism of the synergistic effect of HSA and SO2 in various diseases is obscure, mainly because of the lack of powerful tools that can simultaneously detect HSA and SO2 in living systems. In this work, we report a novel single-site, double-sensing fluorescent probe 1 for the simultaneous detection of HSA and SO2. The probe is based on our finding that HSA can catalyze a Michael addition reaction between the probe and SO2, which induces a change in fluorescence. Probe 1 can effectively entered the endoplasmic reticulum and can be used to image exogenously introduced and de novo synthesis of HSA in endoplasmic reticulum. Furthermore, the simultaneous detection of HSA and SO2 was realized for the first time with probe 1. More important, we observed that HSA still retains its activity to catalyze the Michael addition reaction of 1 and SO2 in living cells, which may provide a significant boost in the study of the role of HSA in medicine and pharmacy.
