Saccade adaptation is the gradual adjustment of saccade end point to maintain spatial accuracy. Contextual adaptation refers to a situation in which the adaptation-related change in saccade end point ...is contingent on the behavioral context in which the saccade is made. For example, in some situations, the same saccade to the same retinotopic location can be simultaneously adapted in opposite directions depending on the context in which it is made. Saccade adaptation has traditionally been studied in isolated movements, but in everyday life, saccades are often planned and executed in sequences. The oculomotor system may therefore have adaptive mechanisms specific to sequential saccades. Here, in five experiments, we investigated contextual saccade adaptation in sequences of saccades. In the first experiment, we demonstrate that saccades to a given retinotopic location can be simultaneously adapted in opposite directions depending on whether they occur in isolation or in a sequence. In the other experiments, we measured the extent to which properties of the previous and following saccades in a sequence can induce contextual saccade adaptation. Overall, we find that the existence, direction, and amplitude of previous and subsequent saccades, as well as the order of the current saccade within a movement sequence, can all induce contextual adaptation. These novel findings demonstrate the surprising flexibility of the system in maintaining end point accuracy, and support the idea that saccades made in a movement sequence are planned concurrently rather than independently.
This study reveals a new type of contextual saccade adaptation: sequential saccades are able to induce contextual saccade adaptation when direction, amplitude, or the existence of preceding and following saccades are used as contexts. These novel findings are also consistent with the idea that saccades made in a sequence are planned concurrently rather than independently.
Objective
Although multiple step saccades (MSS) is occasionally observed in healthy subjects, it is more pronounced in patients with aging-related neurodegenerative diseases, particularly Parkinson’s ...disease (PD). Thus, MSS has been treated as a complementary biomarker for diagnosing PD. Despite the aforementioned knowledge, several questions remain unexplored: (1) How does aging affect MSS? (2) Is there a sex difference in MSS? (3) Are there differences in MSS between vertical and horizontal saccades? (4) Are MSS and corrective saccade (CS) the same behavior? (5) How do age and sex affect CS? The objectives of the present study are to address these questions.
Method
Four hundred eighty healthy participants were recruited to perform a visually guided reactive saccade task. Participants were divided into six groups according to their ages. Each group consisted of 40 male and 40 female participants. Eye movements were recorded with infrared eye trackers.
Results
The incidence of MSS increased as a function of age, whereas the incidence of CS first increased with age 20–49 and then decreased with age 50–79. The incidences of both MSS and CS did not show sex differences. The incidence of MSS in vertical saccades was significantly higher than that in horizontal saccades, and their difference increased with increasing age, whereas the incidence of CS showed a reversed pattern.
Conclusion
Age and saccadic direction affect the occurrences of MSS and CS differently, indicating that MSS and CS are different saccadic behaviors. In addition, measuring saccades could reliably reflect the function of human’s brain which is affected by aging.
The influence of internal brain state on behavioral performance is well illustrated by the gap saccade task, in which saccades might be initiated with short latency (express saccade) or with long ...latency (regular saccade) even though the external visual condition is identical. Accumulated evidence has demonstrated that the internal brain state is different before the initiation of an express saccade than a regular saccade. However, the reported origin of the fluctuation of internal brain state is disputed among previous studies, e.g., the fixation disengagement theory versus the oculomotor preparation theory. The present study examined these two theories by analyzing the rate and direction of fixational saccades, i.e., small amplitude saccades during fixation, because they could be modulated by the internal brain state. Since fixation disengagement is not spatially tuned, it might affect the rate but not the direction of fixational saccades. In contrast, oculomotor preparation can contain spatial information for the upcoming saccade and thus affect fixational saccade direction. We found that the different spatiotemporal characteristics of fixational saccades among tasks with different gap durations reveal diverse driving force to change the internal brain state. Under short gap duration (100 ms), fixation disengagement plays a primary role in switching internal brain states. Conversely, oculomotor preparation plays a primary role under medium (200 ms) and long (400 ms) gap durations. These results suggest that both fixation disengagement and oculomotor preparation can change the internal brain state, but their relative contributions are gap-duration dependent.
While performing the gap saccade task, the role of fixation disengagement and oculomotor preparation in modulating the internal brain state is gap-duration dependent. Fixation disengagement plays a primary role when gap duration is shorter (100 ms), whereas oculomotor preparation plays a primary role when gap duration is longer (200 ms and 400 ms).
•In closed-loop free-viewing visual search there is currently little information available to temporally isolate neural activity analogous to the P300 pattern observed in traditional go/no-go ...studies.•Using deep convolutional networks to decode EEG record around the moment of fixation we observed a probability distribution of a P300 analogue response.•We investigated whether fixation duration, salience, or saccade size led to the greatest reduction in temporal uncertainty in the onset of this response, as measured using the decoded profile.•Results show that only saccade size reliably determines the onset time of the P300 analogue during the closed-loop visual search task and, for this data, the reduction in uncertainty was sufficient to enable traditional ensemble analysis of the waveform as present in the EEG record.
To date the vast majority of research in the visual neurosciences have been forced to adopt a highly constrained perspective of the vision system in which stimuli are processed in an open-loop reactive fashion (i.e., abrupt stimulus presentation followed by an evoked neural response). While such constraints enable high construct validity for neuroscientific investigation, the primary outcomes have been a reductionistic approach to isolate the component processes of visual perception. In electrophysiology, of the many neural processes studied under this rubric, the most well-known is, arguably, the P300 evoked response. There is, however, relatively little known about the real-world corollary of this component in free-viewing paradigms where visual stimuli are connected to neural function in a closed-loop. While growing evidence suggests that neural activity analogous to the P300 does occur in such paradigms, it is an open question when this response occurs and what behavioral or environmental factors could be used to isolate this component.
The current work uses convolutional networks to decode neural signals during a free-viewing visual search task in a closed-loop paradigm within an open-world virtual environment. From the decoded activity we construct fixation-locked response profiles that enable estimations of the variable latency of any P300 analogue around the moment of fixation. We then use these estimates to investigate which factors best reduce variable latency and, thus, predict the onset time of the response. We consider measurable, search-related factors encompassing top-down (i.e., goal driven) and bottom-up (i.e., stimulus driven) processes, such as fixation duration and salience. We also consider saccade size as an intermediate factor reflecting the integration of these two systems.
The results show that of these factors only saccade size reliably determines the onset time of P300 analogous activity for this task. Specifically, we find that for large saccades the variability in response onset is small enough to enable analysis using traditional ensemble averaging methods.
The results show that P300 analogous activity does occur during closed-loop, free-viewing visual search while highlighting distinct differences between the open-loop version of this response and its real-world analogue. The results also further establish saccades, and saccade size, as a key factor in real-world visual processing.

Objective
Differences in saccadic eye movements are widely reported in mental illnesses, and can indirectly inform our understanding of neurobiological and cognitive underpinnings of psychiatric ...conditions, including anorexia nervosa (AN). Preliminary research has suggested that individuals with AN may show specific eye movement abnormalities; whether these deficits are representative of state or trait effects is, however, unclear. The aim of this study was to identify whether there are demonstrable differences in performance on saccadic eye movement tasks in individuals with current AN (c‐AN), those who are weight‐restored from AN (wr‐AN), biological sisters of individuals with AN (AN‐sis), and healthy controls (HC).
Methods
Eighty participants took part in the study (n = 20/group). A set of saccadic eye movement tasks was administered, including prosaccade, antisaccade, memory‐guided saccade, and visual scanpath tasks.
Results
The c‐AN group showed an increased rate of inhibitory errors to 10° targets on the memory‐guided saccade task.
Discussion
The results are discussed in terms of the potential role of the superior colliculus in AN, and that the findings may reflect a state measure of AN.

Saccades are rapid eye movements that are used to move the high acuity fovea in a serial manner in the exploration of the visual scene. Stimulus contrast is known to modulate saccade latency and metrics possibly via changing visual activity in the superior colliculus (SC), a midbrain structure causally involved in saccade generation. However, the quality of visual signals should also be modulated by the amount of lights projected onto the retina, which is gated by the size of the pupil. Although absolute pupil size should modulate visual signals and in turn affect saccade responses, research examining this relationship is very limited. Besides, pupil size is associated with motor preparation. However, the role of pupil dilation in saccade metrics remains unexplored. Through varying peripheral background luminance level and target visual contrast in the saccade task, we investigated the role of absolute pupil size and baseline-corrected pupil dilation in saccade latency and metrics. Higher target detection accuracy was obtained with lower background luminance level, and larger absolute pupil diameter correlated with smaller saccade amplitude and higher saccade peak velocities. More interestingly, the comparable modulation between pupil dilation and stimulus contrast was obtained, showing larger pupil dilation (or higher contrast stimuli) correlating with faster saccade latencies, larger amplitude, higher peak velocities, and smaller endpoint deviation. Together, our results demonstrated the influence of absolute pupil size induced by global luminance level and baseline-corrected pupil dilation associated with motor preparation on saccade latency and metrics, implicating the role of the SC in this behavior.

Tracking of eye movements is an established measurement for many types of experimental paradigms. More complex and more prolonged visual stimuli have made algorithmic approaches to eye-movement event ...classification the most pragmatic option. A recent analysis revealed that many current algorithms are lackluster when it comes to data from viewing dynamic stimuli such as video sequences. Here we present an event classification algorithm—built on an existing velocity-based approach—that is suitable for both static and dynamic stimulation, and is capable of classifying saccades, post-saccadic oscillations, fixations, and smooth pursuit events. We validated classification performance and robustness on three public datasets: 1) manually annotated, trial-based gaze trajectories for viewing static images, moving dots, and short video sequences, 2) lab-quality gaze recordings for a feature-length movie, and 3) gaze recordings acquired under suboptimal lighting conditions inside the bore of a magnetic resonance imaging (MRI) scanner for the same full-length movie. We found that the proposed algorithm performs on par or better compared to state-of-the-art alternatives for static stimulation. Moreover, it yields eye-movement events with biologically plausible characteristics on prolonged dynamic recordings. Lastly, algorithm performance is robust on data acquired under suboptimal conditions that exhibit a temporally varying noise level. These results indicate that the proposed algorithm is a robust tool with improved classification accuracy across a range of use cases. The algorithm is cross-platform compatible, implemented using the Python programming language, and readily available as free and open-source software from public sources.

To accurately foveate a moving target, the oculomotor system needs to estimate the position of the target at the saccade end, based on information about its position and ongoing movement, while ...accounting for neuronal delays and execution time of the saccade. We investigated human interceptive saccades and pursuit responses to moving targets defined by high and low luminance contrast or by chromatic contrast only (isoluminance). We used step-ramps with perpendicular directions between vertical target steps of 10 deg/s and horizontal ramps of 2.5 to 20 deg/s to separate errors with respect to the position step of the target in the vertical dimension, and errors related to target motion in the horizontal dimension. Interceptive saccades to targets of high and low luminance contrast landed close to the actual target positions, suggesting relatively accurate estimates of the amount of target displacement. Interceptive saccades to isoluminant targets were less accurate. They landed at positions the target had on average 100 ms before saccade onset. One account of this finding is that the integration of target motion is compromised for isoluminant targets moving in the periphery. In this case, the oculomotor system can use an accurate, but delayed position component, but cannot account for target movement. This deficit was also present for the postsaccadic pursuit speed. For the two luminance conditions, pursuit direction and speed were adjusted depending on the saccadic landing position. The rapid postsaccadic pursuit adjustments suggest shared position- and motion-related signals of target and eye for saccade and pursuit control.

Till date researchers have elucidated the neurobiological substrates in OCD using methods like neuroimaging. However, a potential biomarker is still elusive. The present study is an attempt to identify a potential biomarker in pediatric OCD using eye tracking. The present study measured pro-saccade and anti-saccade parameters in 36 cases of pediatric OCD and 31 healthy controls. There was no significant difference between cases and controls in the error rate, peak velocity, position gain and latency measures in both pro-saccade and anti-saccade eye tracking tasks. With age, anti-saccades become slower in velocity, faster in response and more accurate irrespective of disorder status of the child. Pro-saccades also show a similar effect that is less prominent than anti-saccades. Gain measures more significantly vary with age in children with OCD than the controls, whereas latency measures positively correlated with age in children with OCD as opposed to being negatively correlated in the controls.
Findings of this study do not support any of the eye tracking measures as putative diagnostic bio-markers in OCD. However, latency and gain parameters across different age groups in anti-saccade tasks need to be explored in future studies.

The planning and execution of sequential saccades can overlap in time, and abrupt changes in neural activity in the oculomotor system can alter the normal trajectory of saccades. In this study, we ...analyzed saccade trajectories to assess the combined programming of sequential saccades. In two separate psychophysical experiments, subjects were instructed to make a sequence of two saccades. The results showed modulation of saccade curvature by the direction and amplitude of both the preceding and following saccade: saccade curvature is modulated in the direction of preceding saccades and away from the direction of following saccades. Moreover, larger preceding and following saccades have stronger effects on curvature. These results support the idea that sequential saccades are programmed concurrently. Finally, the amount of saccade curvature is correlated with the deviation of saccade start and end points, and the time of maximum deviation of saccade trajectories is highly consistent in both experiments. Based on this, we propose a novel benefit for the modulation of saccade trajectories by the oculomotor system: minimizing the saccadic error in sequential saccades.
We show that in saccade sequences, saccade trajectory is modulated in the direction of the preceding saccade and away from the following saccade. The magnitude of this effect is correlated with preceding and following saccade amplitude. This confirms that programming of sequential saccades overlaps. Curvature is also correlated with the deviation of saccade start and end points. Thus, we propose a novel benefit for the modulation of saccade trajectories: minimizing end point error in sequential saccades.
