  • Molecular basis of size and antigenic variation of Mycoplasma synoviae hemagglutinin [Elektronski vir]
    Benčina, Dušan ...
Mycoplasma synoviae (MS) hemagglutinins (LHA) are encoded by a large family of vlhA genes. Sequence analyses of the vlhA 5'ends of 20 MS starains isolated from poultry in Slovenia or USA revealed a ... considerable diversity, particularly within the part endcoding the proline-rich region (PRR). The vlhA gene of the most pathogenic MS strain, K 1968, contained an insertion of seven amino acids (DNPQNPN) between the two nearly identical repeats (of 19 amino acids) within the PRR. In contrast, three other MS isolates, including the less pathogenic strain F 10 2AS, lacked 57 nucleotides encoding the second repeat of the PRR. A polypeptide (&#61566, 200 amino acids) from the vlhA 5' end of the hemagglutination-positive (HA+) clone of ULB 925 strain was expressed in E. coli. The recombinant protein (rpMSB)reacted with antibodies from poultry infected with MS demonstrating that this part of the VLHA proteinrepresents an immunodominant antigen. Moreover, antibodies raised against rpMSB inhibited growth of several MS strains in vitro. Sequence analyses raised of isogenic lineagens from different MS strains revealed polymorphisms that were associated with distinct antigenic variants of the VLHA proteins and eventually with changes in the HA-phenotype.
