  • Proučevanje imunskega odziva pri eksperimentalni okužbi miši z bakterijami Borrelia burgdorferi sensu stricto, Borrelia afzelii in Borrelia garinii : doktorsko delo

Lyme disease is the most common tick transmitted disease in Slovenia. The disease is caused by one of the three borrelia speciesČ Borrelia afzelii, Borrelia garinii or Borrelia burgdorferi sensu ... stricto. Human Lyme disease is manifested by a wide variety of clinical symptoms. Usually the diagnosis is based on clinical signs. The diagnosis of the atypical forms of the disease must be supported by laboratory tests. Since, on many occasions-in spite of the clear clinical picture of the disease-we cannot prove borrelia infection by laboratory tests, it may be suspected that the human immune system reacts insufficiently to borrelia infection, or that the laboratory tests are not specific and sensitive enough. To elucidate these questions we infected experimentally C57BLž6 mice with Borrelia afzelii, Borrelia garinii and Borrelia burgdorferi sensu stricto. We measured the development of immunity toborrelia infection by different laboratory methods. With the ELISPOT method we observed on a single cell level the production of IFN-gamma and IL-4 to thestimulation with borrelia antigens. Our results showed that in infected mice we counted significantly higher numbers of IFN-y producing T helper cellsin comparison to the control mice. The number of IL-4 producing cells after stimulation with borrelia antigens did not differ significantly among the groups of mice. We measured the development of cell-mediated immunity by the lymphocyte blast transformation test. We found that both the lymphocytes of the control and the infected mice respond to borrelia antigens. The strength of the cell mediated response was much stronger in the group of infected mice than in the control group. The response of lymphocytes was most intensive when the antigen used for stimulation was matched to the one used for infection of the mice. (Abstract truncated at 2000 characters)
