Vloga rastlinskega rastnega regulatorja jasmonske kisline pri regeneraciji celične stene protoplastov = [The role of the plant growth regulator jasmonic acid in protoplast cell wall regeneration]Vilhar, BarbaraDuring the last several years jasmonic acid (JA) and other jasmonates have been discovered as new plant growth regulators. They are widely distributed inhigher plants and posses different ... physiological activities. Our previous investigations with JA in tissue culture showed stimulatory effects on shoot elongation and lateral root branching of potato stem cuttings. But JA inhibited callus formation of potato meristems. Our aim was to establish how JA influences cell wall resynthesis and cell division in protoplast culture. Leaf protoplasts were isolated from potato plants (Solanum tuberosum cv. Vesna) propagated from stem nodes in vitro. The regeneration of cell walls wasdelayed in media with lower concentrations of JA (0.01 and 0.1 M microM JA)as compared to the control medium and that containing 1 microM JA after 4-5days of culture. After 10-11 days of culture all viable cells in all media formed cell walls, however the highest number of cell divisions and microcallus formation was observed in cell cultures growing on media containing 0.01 and 0.1 microM JA. In our experiments the effect of JA on microcallus formation was observed in combination with other growth regulators(2.2 microM BA and 4.5 microM 2,4-D). However very low concentrations of JA (0.01 and 0.1 microM) influenced cell divisions. Considering previous experiments these observations suggest that JA plays an important role in the regulation of plant development.Type of material - specialist thesisPublication and manufacture - Ljubljana : [B.
