  • Pomen TNFR-P55 pri progresiji malignega melanoma

    The importance of cytokines is well determined in connection with malignant diseases. On the other hand, the importance of neutralizing factors of the cytokines are not well known. Tumor necrosis ... factor (TNFa) is a member of a cytokine family and is produced by stimulated mononuclear cells and some fibroblasts. TNFa has been discovered because of its ability to provoke the necrosis of tumors. TNFa is cytotoxic and can in some circumstances activate proinflammatory factors. In accordance to the production of TNFa the two TNF receptors, p55 and p75 are produced. Both proteins can be found in the human sera or urine in the soluble form and are supposed to be the natural inhibitors of the cytokine. Both receptors are expressed on nearly every cell membrane with the exception of erythrocytes and resting T lymphocytes. We determined the relative concentrations of mRNA from 11 tumors of metastatic melanoma, 7 primary tumors of the skin melanoma and compared them to the relative mRNA concentrations from healthy tissue. According to our study, we were able to support the hypothesis that more aggresive malignant cells of metastasis expressed significantly more p55 mRNA and therefore soluble p55 than primary tumors or/and healthy tissue cells. We presume that this findingsmight suggest that the soluble p55 contributes to tumor formation and survival by inhibiting immunologic mechanisms in vivo.
