    Organophosphate diisopropyl fluorophosphate (DFP) is structural analog to sarin, a potent nervous agent used in chemical warfare. DFP strongly inhibits acetylcholinesterase, which is located at ... cholinergic synapses and breaks downthe acetylcholine molecule into choline and acetate. As a result, acetylcholine increases in quantity with successive nerve impulses so that large amounts of acetylcholine can accumulate and repetitively stimulate receptors. We presume, that this causes a change in pattern of gene expressionin affected neurons. Differential display (DD) is used worldwide as a method to identify changes in gene expression and to discover novel genes that are involved in important biological pathways. The principle of differential display is the systematic amplification of the 3ć termini of messenger RNAs by using anchored oligo-dT primers in combination with upstreamarbitrary primers. The separation of the polymerase chain reaction products by gel electrophoresis and their direct comparison allows the identification of differentially regulated genes. Because of its simplicity, sensitivity, reproducibility and automation, which increase the throughput andaccuracy, differential display has become one of the most widely used gene-screening methods in biomedical research involving mammals. Rat was injected subcutaneously with a single dose of DFP (10 mgžkg) and the control animal received an equivalent volume of physiologic saline. The toxic signs, tremor and fasciculations, were observed for 10 minutes after the administration of DFP. Rat was sacrified 4 hours after the onset of toxic simptoms, the brain removed and immediately frozen on dry ice and stored at -70°C. Differential display was made on RNA isolated from hippocampus of treated and control rat brain and charachteristic DD gel with pattern of bandswas obtained. Five differentially displayed bands of interest were excised from gel and sent for further analysis.
