    Background: Equinatoxin II (EqT II) is a cytolytic basic peptide isolated fromthe sea anemone Actinia equina (L.). After oligomerization this actinoporin forms cation selective pores in phospholipid ... membranes. Lethal effects of this toxin are due to the cardiorespiratory arrest. Despite intensive investigation the direct cardiotoxic effects and mechanism of a drastic decrease of the coronary perfusion remain unclear. The role of endothelium in the mechanism of the toxins lethal effects has been discussed, but is insufficiently explained. Therefore it seems reasonable to study the possible role of endothelial cells in the decreased perfusion. An increase of the volume of those cells may increase the resistance in the microvasculature resulting in a decreased perfusion rate. Aims: The aim of the study is to assess the effects of EqT II on 1) the dynamic changes of intracellular Ca2+ activity 2) volume and morphology changes in the endothelial cell culture, and3) the role of the composition of the extracellular fluid on those changes.Hypothesis: The main hypothesis is that the application of EqT II causes a significant increase of the endothelial cell volume. It has also beenassumed that the increase of the cell volume is linked to an increased intracellular Ca2+activity which is due to the Ca2+ entry and not due to its release from the intracellular stores.
