    Periodontal disease is an inflammatory disease of periodontal tissues, leadingto degradation of periodontal tissues and alveolar bone loss. Inflammatory reaction is characterised by release of some ... enrymes, inflammatory mediators and cytokines from structure and inflammatory cells. Proinflammatory cytokine tumor necrosis factor alpha (TNFa) was found in cervicular fluid and periodontal tissues of patients with periodontal disease and several in vitro experiments have demonstrated itćs potential osteoresorptive action. The aim of the first experiment was to demonstrate theosteoresorptive capacity of TNFa in vivo. Our hypothesis was that TNFa activates catabolic bone metabolism reactions in mice. Our second hypothesis was, that chronic infusion of TNFa accelerates the inflammatory reaction and promotes the progression of experimental periodontitis in rats. In our first experiment, 20 male C57-Black mice were used. Using microosmotic pumps ALZET, mouse recombinant TNFa was delivered subcutaneously (3pg per day) (n=5) or in tissues above the parietal bone (lpg per day) (n=5). Animals infused similarlywith PBS served as a control group. Serum ionisied calcium and anorganic phosphates were evaluated colorimetricaly. Parietal bone osteoclastswere marked histochemicaly, and surface density of these cells wereevaluated morphometricaly. In the second experiment, male Wistar rats wereused. Inflammation of periodontal tissues was initiated by placing silk ligatures around the cervices of the right second maxillary molars. After two days, the microosmotic pumps ALZET were implanted subsutaneously for two weeks, containing human recombinant TNFalpha (3 micro gžday (n=5) of PBS. Serum ionised calcium and anorganic phosphate levels were evaluated colorimetrically, as in the first experiment. (Abstract truncated at 2000 characters).
