    The aim of our study was to assess the role of telomerase re-activation in thecarcinogenesis of squamous cell carcinoma of the oral cavity, pharynx and larynx, and to validate whether the relative ... copy number of telomerase catalytic subunit (hTERT) mRNA could be used as a molecular biomarker of malignant transformation. The relative copy number of hTERT mRNA was determined in 89 invasive squamous cell carcinomas and 94 tissue samples adjacent to invasive tumour of the oral cavity, pharynx and larynx, by the commercially available ćLightCycler TeIoTAGGGhTERT Quantification Kitć using aLightCycler instrument. The presence and relative copy number of hTERT mRNA increased progressively with the degree of epithelial abnormalities, classified according to the Ljubljana classification. In atypical hyperplasia (also called risky epithelium), the relative copy numbers of hTERT mRNA were significantly higher than in reactive epithelial hyperplastic lesions (p<0,001), and significantly lower than in intraepithelial carcinomas (p<0,01). The presence and relative copy number of hTERT mRNA in invasive squamous cell carcinoma of the oral cavity, pharynx and larynx correlated withthe tumour grade. Poorly differentiated carcinomas had significantly higher relative copy numbers of hTERT mRNA than well differentiated carcinomas(p=0,028). No correlation was present between the relative levels ofhTERT mRNA in intraepithelial and invasive squamous cell carcinomas (p=0,59). The size of the primary tumour and lymph-node status was not associated with the hTERT mRNA relative copy number in the tumour. (Abstract truncated at 2000 characters).
