    In 1952 Alan Turing published his seminal paper called >>The Chemical Basis of Morphogenesis<< in which he presents a reaction-diffusion model of biological pattern formation. Based on the ... interactions of two morphogens, a slowly diffusing activator and a fast diffusing inhibitor, this model enables the formation of spatial patterns which closely resemble those on the leopard spots, zebra stripes, and many other patterns found in nature. Turing patterns have been observed in different chemical systems, whilst their engineering in synthetic biology still presents a big challenge. Synthetic engineering of Turing patterns would present a big step forward in synthetic biology and would provide a better explanation for how patterning occurs in biology. It can also provide practical information for synthetic tissue production, the use of which could contribute to future therapeutic purposes. Previously, different types of genetic circuits capable of Turing patterns production were analysed computationally, and the '3954' Turing circuit proved most promising. This genetic circuit was previously implemented in E. coli chassis with protocatechuic acid (3,4-dihydroxybenxoic acid; PCA) and salicylic acid (2-hydroxybenzoic acid; SA) as node A diffusers. Most of the circuit components, such as promoters, synthesis enzymes, degradation enzymes and transcription factors, were previously optimised. The focus of this study was node A optimisation, which was performed by creating three different variants of node A, obtained by molecular cloning. These were then tested to verify if node A is functioning as expected. Based on the results obtained in this study, expression levels from node A are believed to be optimized, and all of its components are functioning correctly.
