  • Development of HPLC method using liquid-liquid extraction for determination of antidepressant drugs in human plasma

    This study is focused on the development, optimization and validation of an analytical method for determination plasma concentrations of five commonly prescribed antidepressants as are mirtazapine, ... duloxetine, venlafaxine, desvenlafaxine and sertraline. Paroxetine was used as internal standard. The analytes were extracted from plasma using liquid-liquid sample preparation method and for their quantification high-performance liquid chromatography was selected. The optimal conditions for analytes% extraction were obtained by using 10 % of ethyl acetate in cyclohexane followed by their separation on C18 reversed - phase column using a gradient of acetonitrile in 25 mM phosphate buffer pH 3,5. The flow rate of the mobile phase was 1,2 mL/min and analytes were detected with photodiode-array UV and fluorescence detector. Calibration curves were linear from 10 ng/mL to 200 ng/ mL. The accuracy of quality controls samples at all concentrations were within 15 % of deviation except for venlafaxine and sertraline. Moreover, the within-run and between-run precision expressed with coefficients of variation was less than 15 %. Extraction recoveries were ranging from 87,2 to 110 %. This new method will be used for therapeutic drug monitoring of selected antidepressant drugs.
