  • Histofotometrična ocena aktivnosti presnovnih encimov v skeletnih mišičnih vlaknih pri prašiču = Histophotometric evaluation of metabolic enzyme activity in pig skeletal muscle fibres

    Using computerized image analysing system the activities of mitochondrial succinate dehydrogenase (SDH), mitochondrial alpha-glycerophosphate dehydrigenase (alpha-GPD) and alpha-glucan phosphorylase ... in muscle fibre typesI, IIa and IIb were determined. M. longissimus dorsi and m. biceps femoris of nine Duroc boars were analysed. The results of the analysis were compared to the muscle pH1 as an marker of the early post-mortem glycolysis. The positive correlation between pH1 and SDH activity was statistically significant (p < 0.05), while the correlation between the pH1 and alpha-GPD was not confirmed (p < 0,1). The negative correlation was determined between the SDH and a-GPD activity (p < 0,01). The alpha-glucan phosphorylase activitywas very unstable. There were no significant differences in the enzymeactivity between muscle fibre types, neither any correlation between thepH1 and the enzyme activity. The metabolic profile of the muscle fibres could be evaluated sufficiently only by mitochondrial enzymes activity. The SDH and alpha-GPD activity is stable also on the samples affected with intensive glycolysis. The best results were obtained by the histophotometric analysis of the SDH activity. This enzyme allows clear metabolic differentiation of the muscle fibres and is in correlation with the pH1 of the muscle.
