Exposure to methylmercury (MeHg) can result in detrimental health effects in wildlife. With advances in ecological indicators and analytical techniques for measurement of MeHg in a variety of tissues, numerous processes have been identified that can influence MeHg concentrations in wildlife. This review presents a synthesis of theoretical principals and applied information for measuring MeHg exposure and interpreting MeHg concentrations in wildlife. Mercury concentrations in wildlife are the net result of ecological processes influencing dietary exposure combined with physiological processes that regulate assimilation, transformation, and elimination. Therefore, consideration of both physiological and ecological processes should be integrated when formulating biomonitoring strategies. Ecological indicators, particularly stable isotopes of carbon, nitrogen, and sulfur, compound-specific stable isotopes, and fatty acids, can be effective tools to evaluate dietary MeHg exposure. Animal species differ in their physiological capacity for MeHg elimination, and animal tissues can be inert or physiologically active, act as sites of storage, transformation, or excretion of MeHg, and vary in the timing of MeHg exposure they represent. Biological influences such as age, sex, maternal transfer, and growth or fasting are also relevant for interpretation of tissue MeHg concentrations. Wildlife tissues that represent current or near-term bioaccumulation and in which MeHg is the predominant mercury species (such as blood and eggs) are most effective for biomonitoring ecosystems and understanding landscape drivers of MeHg exposure. Further research is suggested to critically evaluate the use of keratinized external tissues to measure MeHg bioaccumulation, particularly for less-well studied wildlife such as reptiles and terrestrial mammals. Suggested methods are provided to effectively use wildlife for quantifying patterns and drivers of MeHg bioaccumulation over time and space, as well as for assessing the potential risk and toxicological effects of MeHg on wildlife.
Methlymercury is a significant risk to environmental health globally. We examined the ecological drivers of methylmercury bioaccumulation in songbirds and its effect on body condition while ...experimentally removing the potentially confounding and predominant effects of site and habitat. We measured blood and feather mercury concentrations and body condition in nearly 1200 individuals representing resident or migrant songbirds of 52 species and 5 foraging guilds. Songbird mercury concentrations differed among species, foraging guilds, residency status, dates, and ages, but not sexes. Blood mercury concentrations 1) ranged from 0.003 in house finch to 0.85 μg/g ww in American robin, 2) were 125 times greater in insectivores than granivores and 3.6 times greater in insectivores than omnivores, 3) were 3.3 times greater in summer residents than in migrating songbirds, 4) increased by 25% throughout spring and summer, and 5) were 45% higher in adults than juveniles. Songbird mercury concentrations were negatively correlated with body condition, with blood mercury concentrations decreasing by 44% and 34% over the range of standardized body masses and fat scores, respectively. Our results highlight the importance of foraging and migration ecology in determining methylmercury contamination in birds, and the potential for reduced body condition with methylmercury exposure in songbirds.

Neuronal cell types are the nodes of neural circuits that determine the flow of information within the brain. Neuronal morphology, especially the shape of the axonal arbor, provides an essential ...descriptor of cell type and reveals how individual neurons route their output across the brain. Despite the importance of morphology, few projection neurons in the mouse brain have been reconstructed in their entirety. Here we present a robust and efficient platform for imaging and reconstructing complete neuronal morphologies, including axonal arbors that span substantial portions of the brain. We used this platform to reconstruct more than 1,000 projection neurons in the motor cortex, thalamus, subiculum, and hypothalamus. Together, the reconstructed neurons constitute more than 85 meters of axonal length and are available in a searchable online database. Axonal shapes revealed previously unknown subtypes of projection neurons and suggest organizational principles of long-range connectivity.

Toxicity of methylmercury (MeHg) to wildlife and humans results from its binding to cysteine residues of proteins, forming MeHg-cysteinate (MeHgCys) complexes that hinder biological functions. ...MeHgCys complexes can be detoxified in vivo, yet how this occurs is unknown. We report that MeHgCys complexes are transformed into selenocysteinate Hg(Sec)4 complexes in multiple animals from two phyla (a waterbird, freshwater fish, and earthworms) sampled in different geographical areas and contaminated by different Hg sources. In addition, high energy-resolution X-ray absorption spectroscopy (HR-XANES) and chromatography-inductively coupled plasma mass spectrometry of the waterbird liver support the binding of Hg(Sec)4 to selenoprotein P and biomineralization of Hg(Sec)4 to chemically inert nanoparticulate mercury selenide (HgSe). The results provide a foundation for understanding mercury detoxification in higher organisms and suggest that the identified MeHgCys to Hg(Sec)4 demethylation pathway is common in nature.
Feathers are widely used to represent mercury contamination in birds. Yet, few recommendations exist that provide guidance for using bird feathers in mercury monitoring programs. We conducted a ...literature review and 5 experiments to show that mercury concentrations vary substantially within (vane >100% higher than calamus) and among (>1000%) individual feathers from the same bird. We developed a research tool and guidelines for using bird feathers for mercury studies based on 3 components: 1) variability of feather mercury concentrations within an individual bird (coefficient of variation), 2) desired accuracy of the measured mercury concentration, and 3) feather and bird mass. Our results suggest a general rule that if the goal is to limit analytical and processing costs by using whole feathers in only one sample boat, then to achieve an accuracy within 10% of a bird's overall average feather mercury concentration a bird with a coefficient of variation ≤10% must be <190 g (size of a large shorebird). To achieve an accuracy within 20%, a bird with a coefficient of variation ≤10% must be <920 g (a large duck). When more than one sample boat is needed to fit the required number of feathers to achieve the desired accuracy, the results suggest homogenizing feathers and analyzing an aliquot of ≥20 mg for mercury. Methylmercury contamination of the environment is an important issue globally, and birds are useful bioindicators for mercury monitoring programs. The available data on mercury contamination of birds ...in western North America were synthesized. Original data from multiple databases were obtained and a literature review was conducted to obtain additional mercury concentrations. In total, 29219 original bird mercury concentrations from 225 species were compiled, and an additional 1712 mean mercury concentrations, representing 19998 individuals and 176 species, from 200 publications were obtained. To make mercury data comparable across bird tissues, published equations of tissue mercury correlations were used to convert all mercury concentrations into blood-equivalent mercury concentrations. Blood-equivalent mercury concentrations differed among species, foraging guilds, habitat types, locations, and ecoregions. Piscivores and carnivores exhibited the greatest mercury concentrations, whereas herbivores and granivores exhibited the lowest mercury concentrations. Bird mercury concentrations were greatest in ocean and salt marsh habitats and lowest in terrestrial habitats. Bird mercury concentrations were above toxicity benchmarks in many areas throughout western North America, and multiple hotspots were identified. Additionally, published toxicity benchmarks established in multiple tissues were summarized and translated into a common blood-equivalent mercury concentration. Overall, 66% of birds sampled in western North American exceeded a blood-equivalent mercury concentration of 0.2 μg/g wet weight (ww; above background levels), which is the lowest-observed effect level, 28% exceeded 1.0 μg/g ww (moderate risk), 8% exceeded 3.0 μg/g ww (high risk), and 4% exceeded 4.0 μg/g ww (severe risk). Mercury monitoring programs should sample bird tissues, such as adult blood and eggs, that are most-easily translated into tissues with well-developed toxicity benchmarks and that are directly relevant to bird reproduction. Results indicate that mercury contamination of birds is prevalent in many areas throughout western North America, and large-scale ecological attributes are important factors influencing bird mercury concentrations.

In birds, parents must provide their eggs with a safe thermal environment suitable for embryonic development. Species with uniparental incubation must balance time spent incubating eggs with time ...spent away from the nest to satisfy self-maintenance needs. Patterns of nest attendance, therefore, influence embryonic development and the time it takes for eggs to hatch. We studied nest attendance (time on the nest), incubation constancy (time nests were at incubation temperatures), and variation in nest temperature of 1,414 dabbling duck nests of three species in northern California. Daily nest attendance increased from only 1-3% on the day the first egg was laid to 51-57% on the day of clutch completion, and 80-83% after clutch completion through hatch. Variation in nest temperature also decreased gradually during egg-laying, and then dropped sharply (33-38%) between the day of and the day after clutch completion because increased nest attendance, particularly at night, resulted in more consistent nest temperatures. During the egg-laying stage, nocturnal nest attendance was low (13-25%), whereas after clutch completion, nest attendance was greater at night (≥87%) than during the day (70-77%) because most incubation recesses occurred during the day. Moreover, during egg-laying, nest attendance and incubation constancy increased more slowly among nests with larger final clutch sizes, suggesting that the number of eggs remaining to be laid is a major driver of incubation effort during egg-laying. Although overall nest attendance after clutch completion was similar among species, the average length of individual incubation bouts was greatest among gadwall (Mareca strepera; 779 minutes), followed by mallard (Anas platyrhynchos; 636 minutes) and then cinnamon teal (Spatula cyanoptera; 347 minutes). These results demonstrate that dabbling ducks moderate their incubation behavior according to nest stage, nest age, time of day, and clutch size and this moderation likely has important implications for egg development and overall nest success.

Abstract In birds, mercury embryotoxicity can occur through the transfer of mercury from the female to her eggs. Maternal transfer of mercury can vary by egg position in the laying sequence, with ...first‐laid eggs often exhibiting greater mercury concentrations than subsequently laid eggs. We studied egg mercury concentration, mercury burden (total amount of mercury in the egg), and egg morphometrics by egg position in the laying sequence for two songbirds: tree swallows ( Tachycineta bicolor ) and house wrens ( Troglodytes aedon ). Egg mercury concentration in the second egg laid was 14% lower for tree swallows and 6% lower for house wrens in comparison with the first egg laid. These results indicate that in both species, after an initial relatively high transfer of mercury into the first egg laid, a smaller amount of mercury was transferred to the second egg laid. This lower mercury concentration persisted among all subsequently laid eggs (eggs three to eight) in tree swallows (all were 14%–16% lower than egg 1), but mercury concentrations in subsequently laid house wren eggs (eggs three to seven) returned to levels observed in the first egg laid (all were 1% lower to 3% greater than egg 1). Egg size increased with position in the laying sequence in both species; the predicted volume of egg 7 was 5% and 6% greater than that of egg 1 in tree swallows and house wrens, respectively. This change was caused by a significant increase in egg width, but not egg length, with position in the laying sequence. The percentage of decline in mercury concentration with position in the laying sequence was considerably lower in tree swallows and house wrens compared with other bird taxonomic groups, suggesting that there are key differences in the maternal transfer of mercury into songbird eggs compared with other birds. Finally, we performed simulations to evaluate how within‐clutch variation in egg mercury concentrations affected estimates of mean mercury concentrations in each clutch and the overall sampled population, which has direct implications for sampling designs. Estuaries are globally important areas for methylmercury bioaccumulation because of high methylmercury production rates and use by fish and wildlife. We measured total mercury (THg) concentrations in ...ten fish species from 32 wetland and open bay sites in San Francisco Bay Estuary (2005–2008). Fish THg concentrations (μg/g dry weight ± standard error) differed by up to 7.4× among estuary habitats. Concentrations were lowest in open bay (0.17 ± 0.02) and tidal wetlands (0.42 ± 0.02), and highest in managed seasonal saline wetlands (1.27 ± 0.05) and decommissioned high salinity salt ponds (1.14 ± 0.07). Mercury also differed among fishes, with Mississippi silversides (0.87 ± 0.03) having the highest and longjaw mudsuckers (0.37 ± 0.01) the lowest concentrations. Overall, 26% and 12% of fish exceeded toxicity benchmarks for fish (0.20 μg/g wet weight) and piscivorous bird (0.30 μg/g wet weight) health, respectively. Our results suggest that despite managed wetlands' limited abundance within estuaries, they may be disproportionately important habitats of Hg risk to coastal wildlife.

Mercury contamination of oceans is prevalent worldwide and methylmercury concentrations in the mesopelagic zone (200–1000 m) are increasing more rapidly than in surface waters. Yet mercury ...bioaccumulation in mesopelagic predators has been understudied. Northern elephant seals (Mirounga angustirostris) biannually travel thousands of kilometres to forage within coastal and open-ocean regions of the northeast Pacific Ocean. We coupled satellite telemetry, diving behaviour and stable isotopes (carbon and nitrogen) from 77 adult females, and showed that variability among individuals in foraging location, diving depth and δ13C values were correlated with mercury concentrations in blood and muscle. We identified three clusters of foraging strategies, and these resulted in substantially different mercury concentrations: (i) deeper-diving and offshore-foraging seals had the greatest mercury concentrations, (ii) shallower-diving and offshore-foraging seals had intermediate levels, and (iii) coastal and more northerly foraging seals had the lowest mercury concentrations. Additionally, mercury concentrations were lower at the end of the seven-month-long foraging trip (n = 31) than after the two-month- long post-breeding trip (n = 46). Our results indicate that foraging behaviour influences mercury exposure and mesopelagic predators foraging in the northeast Pacific Ocean may be at high risk for mercury bioaccumulation.
