The research is devoted to the architectural identity of historical blocks in Kazan, the capital city of the Republic of Tatarstan. The paper raises the issue of national identity regarding the city ...blocks in the Old Tatar settlement and explores the history of the architectural environment and gradual decay of its identity, including the national one, by the end of the 20th century. The system of national color identifiers of Tatar architecture is described. The possibilities of their use on different types of buildings and structures are discussed. The experiences in developing and implementing various color solutions in the reconstruction of buildings in the Old Tatar settlement are reviewed. The color environment of the area during reconstruction is analyzed. Productive methods for professional interpretation of national color traditions in the urban environment are revealed. The study has enabled recommendations to be formulated for the development of color solutions for the historical part of the Old Tatar settlement in Kazan with the aim of recreating the national identity of the architectural environment.
The control of wheat diseases using bioagents is not well studied under field conditions. The present study was aimed at investigating, during four consecutive growing seasons (2017–2020), the ...efficacy of two integrated crop protection (ICP) systems to control the common wheat diseases for enhancing the productivity and profitability of winter wheat crops and ensuring nutritional and food security. Two environmental-friendly treatments were tested, biological (T1), which contained bioagents and fertilizers, and combined (T2), which included fertilizers and bioagents coupled with lower doses of fungicides. The chemical treatment (T3) was used for comparison with (T1) and (T2). Furthermore, two Russian winter wheat varieties (Nemchinovskaya 17 (V1) and Moscovskaya 40 (V2)) were studied. A randomized complete block design was used with four replicates. Diseases infestation rates for snow mold (SM), root rot (RR), powdery mildew (PM), and Fusarium (Fus), yield performances, and grain quality (measured through protein content) were determined according to the tested treatments, and the economic efficiency was calculated for each treatment. The combined treatment (T2) was the most effective against fungal diseases with 1.8% (SM), 1.2% (RR), 0.9% (PM), and 0.9% (Fus). The highest grain yield (6.8 t·ha−1), protein content (15.2%), and 1000-grain weight (43.7%) were observed for winter wheat variety Moscovskaya 40 with the combined treatment (T2). The highest number of productive stems (N.P.S) (556 stems/m2) was attained for combined treatment (T2), followed by biological treatment (T1) (552 stems/m2) with the variety Nemchinovskaya 17. The profitability (cost–benefit ratio) of the combined treatment (T2) was 2.38 with the Moscovskaya 40 variety (V2), while 2.03 was recorded for the biological treatment. Applying environmentally friendly combined and biological treatments resulted in high wheat yield and net income, as well as healthy products.
In the light of Vavilov's Law, grain traumatization in the standing crop of wheat and other crops due to the enzyme stage of enzyme-mycotic seed depletion (EMSD) was confirmed, the parameters of open ...and hidden harmfulness were detected, and a scale of plant resistance to such traumatization was developed. The current study demonstrates that pathogen contamination in grains occurs before harvesting and its degree is determined by favorable humidity and temperature conditions and by the open and hidden grain traumatization due to the enzyme stage of EMSD, i.e., the grain's hydrolytic enzymes providing a growth substrate for a fungal spread that is later substituted by pathogen enzymes leading to grain spoiling and self-warming. The most common technique to preserve grain quality is to support a moisture level that prevents further spreading of the fungi. The grains that are contaminated with very low temperature and humidity levels facilitate the germinability and high quality of the grain. The new ways to withstand EMSD should, first of all, include a selection of activities. Using biological, biochemical and physical (X-ray) methods, genetic sources of resistance towards EMSD were found in the VIR world collection that is recommended for further selection. These sources have become a basis for the varieties, such as Moskovskaya 39, Ilot (winter wheat), Gremme and Gremme 2U (hulless spelt), Alcoran (winter spelt) and Kanysh (spring wheat).

MicroRNAs (miRNAs) are a novel class of gene expression regulators implicated in cancer biology. Neuroblastoma (NB) is an embryonal tumour consisting of neural crest-derived undifferentiated cells ...and is characterised by variable clinical courses ranging from spontaneous regression to therapy-resistant progression. Recent advances identified a subset of miRNAs with putative function in NB biology. However, the full repertoire of miRNAs expressed in NBs is not available.
We describe miRNA profiles of 13 NB specimens and 2 NB cell lines as determined by miRNA cloning. A total of 3153 sequences were sequenced and analysed by a miRNA prediction tool (miRpredict). Our library covered 27% miRNAs known to date. 39 reads corresponding to 25 individual sequences were classified as novel miRNAs, including miRNA* species of 10 known miRNAs. Expression of 5 new miRNA* forms and 8 individual sequences was supported by Northern blotting. Most of the novel miRNA genes are not related to each other and do not share homology with the annotated sequences in the public miRNA database, but they are conserved within mammals or have close homologues in primates genomes.
We provide evidence for 29 new miRNA and miRNA-like sequences (24 novel sequences and 5 miRNAs discovered initially in other species). Some of these newly identified sequences reside within frequently altered chromosomal regions in NB tumours and may play a role in NB biology.

The antioxidant defense system can be stimulated by growth regulators in plants when they are under stress, such as exposure to pathogens. There are a lot of natural growth regulators on the market, ...but no research has been carried out yet to determine how effective they are. This field and laboratory study examines the impact of two commonly used Russian growth regulators, Crezacin and Zircon, along with artificial infection with Fusarium culmorum on the antioxidant system of naked oat. The results show that, compared to the control, Crezacin-treated plants had higher contents of low molecular weight fructose and nonenzymatic antioxidants like proline, phenolic compounds, and flavonoids. Zircon-treated plants had a lower content of proline, carbohydrates, and lower total antioxidant activity than the control plants. The obtained data show that Crezacin treatment mainly affected nonenzymatic systems of the antioxidant defense. This treatment was more successful than the Zircon application, which did not show any appreciable effectiveness and was typically associated with an improvement in oat productivity. The treatment with growth regulators and a fungal suspension performed at the flowering phase provided the best effect on the biochemical parameters and productivity of naked oats. Moreover, oat treatment with the pathogen promoted the reproductive capabilities of the plants, while growth regulators helped in avoiding infectious stress.
The paper presents the results of a 50-year research of the genepool of the winter wheat from the world’s largest wheat collection of N.I. Vavilov Institute of Plant Industry (VIR) to investigate its ...resistance to the abiotic stress factors of the Moscow region and see how closely it matches the attributes of a wheat ideotype as postulated by N.I. Vavilov in 1935. The critical years in studying the wheat’s winter resistance were 10 years out of 50: excessive water saturation during the year 2013; soil drought in 1988; and atmospheric drought in 1972 and 2010. During the investigation, the following gene pool features were analyzed: frost characterized by the cultivar Sojuz 50 (Russia), rapid temperature change, thawing, ice, and rotting resistance characterized by the cultivars Zarya 2 (Russia), Sv 75268, (Sweden), Caristerm and Tukan (Germany), PP 114-74 and Liwilla (Poland), Maris Ploughman and Granta (Great Britain), Titan (USA), Zdar (Czech), and Zenta (Switzerland); regeneration capacity in spring after poor wintering expressed by the cultivars Pamyati Fedina (Russia), TAW 3668.71 (Germany) and Rmo (Poland); resistance to excessive soil and air saturation exhibited by the cultivars Moskovskaya 39 (Russia), Tukan, Compal, Obelisk, Orestis, and Bussard (Germany); solid standing culm that is resistant to lodging characterized by the cultivars Tukan, Kronjuwel, Compal (Germany), Zenta (Switzerland), Moskovskaya 56 (Russia), and Hvede Sarah (Denmark); resistance to enzyme-mycotic depletion of seeds characterized by the cultivars Tukan, Compal, Obelisk, Orestis, Bussard (Germany), Sv 75268, Helge, VG 73394, Salut, Sv 75355 (Sweden), Zenta (Switzerland), Moskovskaya 39, and Ferrugineum 737.76 (Russia); and resistance to soil and atmospheric drought demonstrated by the cultivars Liessau, Heine Stamm, Severin, Neuzucht 14/4, Haynes, Rus 991, Halle 1020 (Germany), Gama (Poland), Sv 71536 (Sweden), and Moskovskaya 39 (Russia). Moreover, the cultivar Mironovskaya 808 (Ukraine) showed resistance to almost all abiotic stress factors studied. The performed study contributes towards the provision of potential sources of resistance to abiotic stress factors prevalent in the Moscow region that can be incorporated in advanced breeding programs.
The migrational propensity of neuroblastoma is affected by cell identity, but the mechanisms behind the divergence remain unknown. Using RNAi and time-lapse imaging, we show that ADRN-type NB cells ...exhibit RAC1- and kalirin-dependent nucleokinetic (NUC) migration that relies on several integral components of neuronal migration. Inhibition of NUC migration by RAC1 and kalirin-GEF1 inhibitors occurs without hampering cell proliferation and ADRN identity. Using three clinically relevant expression dichotomies, we reveal that most of up-regulated mRNAs in RAC1- and kalirin-GEF1-suppressed ADRN-type NB cells are associated with low-risk characteristics. The computational analysis shows that, in a context of overall gene set poverty, the upregulomes in RAC1- and kalirin-GEF1-suppressed ADRN-type cells are a batch of AU-rich element-containing mRNAs, which suggests a link between NUC migration and mRNA stability. Gene set enrichment analysis-based search for vulnerabilities reveals prospective weak points in RAC1- and kalirin-GEF1-suppressed ADRN-type NB cells, including activities of H3K27- and DNA methyltransferases. Altogether, these data support the introduction of NUC inhibitors into cancer treatment research.
To examine the cytotoxic activity of congeners of 3‐amino‐isoquinoline, we performed the phenotypic screening using panel of 60 cell lines and found that ...(N‐(6,7‐dimethoxy‐1‐methyl‐isoquinolin‐3‐yl)‐4‐{(1‐ethyl‐4‐methyl‐1H‐pyrazol‐3‐yl)methylamino}benzamide (4d)) exhibited the significant effect against different tumor cell lines while showing the high activity toward human colorectal cancer HCT‐116 cells (IC50 = 18 μm) and human breast cancer T‐47D cells (GI50 = 1.9 μm). Virtual screening indicated that these compounds target protein kinases and phosphodiesterases (PDE). However, wet screening among panel of protein kinases did not show any significant activity. By contrast, 50 μm of 4c and 4d inhibited the growth of HKe3‐mtKRAS spheroids in the 3D floating (3DF) culture suggesting that 4c and 4d target PDE4B which is selectively upregulated by mtKRAS in 3DF culture.
It was found by the phenotypic screening that congeners of 3‐amino‐isoquinoline possessed cytotoxic activity in the HCT116, HKe3, K562 cell lines. Primary hit (N‐(6,7‐dimethoxy‐1‐methylisoquinolin‐3‐yl)‐4‐{(1‐ethyl‐4‐methyl‐1H‐pyrazol‐3‐yl)methylamino}benzamide (4d)) from small combinatorial library had IC50 18 μm for HCT116 and GI50 1.9 μm for T‐47D cell line. Compounds 4c and 4d decreased the area of HKe3‐mtKRAS spheroids in the 3D floating (3DF) culture at 50 μm; thus, 4c and 4d selectively inhibited the growth of spheroids dependent on the mtKRAS in the 3DF culture and targeted PDE4B in HKe3‐mt‐KRAS spheroids.
Introduction
. Pneumococcal diseases remain relevant for the whole world. On the one hand, this is due to the high prevalence of pneumococcus and the other hand, the growth of antibiotic-resistant ...strains and the constant change of clinically significant serotypes of the pathogen.
The aim of the research
was to study of the protective activity of a mixture of pneumococcal antigens.
Material and methods
. we used preparations of a capsular polysaccharide (CPS) obtained from
Streptococcus pneumoniae
serotype 3; protein-containing fraction (PCF) obtained from an aqueous extract of cells of
S. pneumoniae
serotype 6B; recombinant pneumolysin (rPly). Mice were immunized intraperitoneally twice with an interval of 14 days with mixtures of bacterial antigens: CPS + PCF; CPS + rPly; PCF + rPly. To assess the protective activity of the studied drugs after double immunization animals were infected intraperitoneally with
S. pneumoniae
serotype 3. To study the effect of mixtures of bacterial preparations on the infectious process in the lungs immunized mice were infected with
S. pneumoniae
serotype 3. The humoral immune response was studied with IgG using the method of ELISA.
Results
. The CPS + rPly mixture protected mice from intraperitoneal infection with
S. pneumoniae
serotype 3 regardless of the infecting dose. Immunization with CPS + PCF or CPS + rPly mixtures influenced a significant decrease the number of seeded bacterial cells from lungs during the entire observation period (72 h) compared to the control. Administration of mixtures of bacterial antigens of CPS + PCF, CPS + rPly or PCF + rPly to animals led to a significant increase of the level of antibodies to all antigens, however, the highest levels of IgG were determined to PCF and rPly.
Conclusion
. The results obtained suggest that different antigenic drugs in mixtures affect different mechanisms of immunity activation.
