Little is understood about the socioeconomic predictors of tooth loss, a condition that can negatively impact individual's quality of life. The goal of this study is to develop a machine-learning ...algorithm to predict complete and incremental tooth loss among adults and to compare the predictive performance of these models. We used data from the National Health and Nutrition Examination Survey from 2011 to 2014. We developed multiple machine-learning algorithms and assessed their predictive performances by examining the area under the receiver operating characteristic curve (AUC), accuracy, sensitivity, specificity, and positive and negative predictive values. The extreme gradient boosting trees presented the highest performance in the prediction of edentulism (AUC = 88.7%; 95%CI: 87.1, 90.2), the absence of a functional dentition (AUC = 88.3% 95%CI: 87.3,89.3) and for predicting missing any tooth (AUC = 83.2%; 95%CI, 82.0, 84.4). Although, as expected, age and routine dental care emerged as strong predictors of tooth loss, the machine learning approach identified additional predictors, including socioeconomic conditions. Indeed, the performance of models incorporating socioeconomic characteristics was better at predicting tooth loss than those relying on clinical dental indicators alone. Future application of machine-learning algorithm, with longitudinal cohorts, for identification of individuals at risk for tooth loss could assist clinicians to prioritize interventions directed toward the prevention of tooth loss.

To provide an overview of the literature regarding the use of machine learning algorithms to predict hypertension. A systematic review was performed to select recent articles on the ...subject.
Recent Findings
The screening of the articles was conducted using a machine learning algorithm (ASReview). A total of 21 articles published between January 2018 and May 2021 were identified and compared according to variable selection, train-test split, data balancing, outcome definition, final algorithm, and performance metrics. Overall, the articles achieved an area under the ROC curve (AUROC) between 0.766 and 1.00. The algorithms most frequently identified as having the best performance were support vector machines (SVM), extreme gradient boosting (XGBoost), and random forest.
Summary
Machine learning algorithms are a promising tool to improve preventive clinical decisions and targeted public health policies for hypertension. However, technical factors such as outcome definition, availability of the final code, predictive performance, explainability, and data leakage need to be consistently and critically evaluated.
A high number of genetic mutations associated with cortical malformations are found in genes coding for microtubule-related factors. This has stimulated research to understand how the various ...microtubule-based processes are regulated to build a functional cerebral cortex. Here, we focus our review on the radial glial progenitor cells, the stem cells of the developing neocortex, summarizing research mostly performed in rodents and humans. We highlight how the centrosomal and acentrosomal microtubule networks are organized during interphase to support polarized transport and proper attachment of the apical and basal processes. We describe the molecular mechanism for interkinetic nuclear migration (INM), a microtubule-dependent oscillation of the nucleus. Finally, we describe how the mitotic spindle is built to ensure proper chromosome segregation, with a strong focus on factors mutated in microcephaly.
Introduction Le 26RFa est un neuropeptide initialement découvert dans l'hypothalamus mais qui est également présent en abondance dans le tractus gastrointestinal et les îlots pancréatiques. Nous ...avons récemment montré que le 26RFa régule l'homéostasie glucidique en agissant comme une incrétine et en augmentant l'insulino-sensibilité. L'objectif de la présente étude était d'évaluer l'impact du 26RFa sur la régulation glycémique au cours de l'obésité associée au diabète de type 2. Matériels et Méthodes Nous avons utilisé un modèle de souris obèses et diabétiques, en soumettant les animaux à un régime hypercalorique high fat (HF) pendant 3 mois. La glycémie et l'insulinémie ont été mesurées au cours de différents tests fonctionnels in vivo. L'expression du récepteur du 26RFa dans les tissus cibles de l'insuline a été mesurée par PCR quantitative. Nous avons également évalué l'effet du 26RFa sur la sécrétion d'insuline par les cellules β pancréatiques humaines EndoC-βH1, en condition native et dans un modèle de lipotoxicité induit par le palmitate. Résultats Chez les souris obèses et diabétiques, nous avons observé une perte de l'effet antihyperglycémiant du 26RFa et une diminution de son effet insulinosensibilisateur. Le récepteur du 26RFa, le GPR103, est exprimé dans le muscle et le tissu adipeux et son expression est diminuée chez les souris HF. Le 26RFa stimule significativement la sécrétion d'insuline gluco-induite par les cellules β pancréatiques humaines EndoC-βH1. L'exposition des cellules au palmitate pendant 4 jours diminue significativement la sécrétion d'insuline par les EndoC-βH1, et abolit totalement l'effet insulino-sécrétant du 26RFa. De plus, le traitement au palmitate induit une baisse d'expression du GPR103. Conclusions Chez les animaux obèses et diabétiques, le 26RFa perd ses propriétés antihyperglycémiantes. Les effets insulino-senbilisateurs et insulino-sécrétants du 26RFa sont altérés, ce qui pourrait s'expliquer en partie par une baisse d'expression de son récepteur dans les tissus cibles de l'insuline et dans les cellules β pancréatiques.
Dynein recruitment to the nuclear envelope is required for pre-mitotic nucleus-centrosome interactions in nonneuronal cells and for apical nuclear migration in neural stem cells. In each case, dynein ...is recruited to the nuclear envelope (NE) specifically during G2 via two nuclear pore-mediated mechanisms involving RanBP2-BicD2 and Nup133-CENP-F. The mechanisms responsible for cell-cycle control of this behavior are unknown. We now find that Cdk1 serves as a direct master controller for NE dynein recruitment in neural stem cells and HeLa cells. Cdk1 phosphorylates conserved sites within RanBP2 and activates BicD2 binding and early dynein recruitment. Late recruitment is triggered by a Cdk1-induced export of CENP-F from the nucleus. Forced NE targeting of BicD2 overrides Cdk1 inhibition, fully rescuing dynein recruitment and nuclear migration in neural stem cells. These results reveal how NE dynein recruitment is cell-cycle regulated and identify the trigger mechanism for apical nuclear migration in the brain.

Neural stem cells undergo motor protein-dependent nuclear oscillations between mitotic divisions, but how nuclear migration regulation is linked to the cell cycle has been unclear. Baffet et al. report that Cdk1 activates dynein recruitment to the nuclear pores during G2 phase through an effect on the dynein recruitment factors BicD2 and CENP-F.

Strike patterns during running have received attention in the recent ...literature due to their potential mechanical differences and associated injury risks.
Electronic databases (MEDLINE, Embase, LILACS, SciELO, and SPORTDiscus) were searched through July 2014. Studies (cross-sectional, case-control, prospective, and retrospective) comparing the biomechanical characteristics of foot-strike patterns during running in distance runners at least 18 years of age were included in this review. Two independent reviewers evaluated the risk of bias. A meta-analysis with a random-effects model was used to combine the data from the included studies.
Sixteen studies were included in the final analysis. In the meta-analyses of kinematic variables, significant differences between forefoot and rearfoot strikers were found for foot and knee angle at initial contact and knee flexion range of motion. A forefoot-strike pattern resulted in a plantar-flexed ankle position and a more flexed knee position, compared to a dorsiflexed ankle position and a more extended knee position for the rearfoot strikers, at initial contact with the ground. In the comparison of rearfoot and midfoot strikers, midfoot strikers demonstrated greater ankle dorsiflexion range of motion and decreased knee flexion range of motion compared to rearfoot strikers. For kinetic variables, the meta-analysis revealed that rearfoot strikers had higher vertical loading rates compared to forefoot strikers.
There are differences in kinematic and kinetic characteristics between foot-strike patterns when running. Clinicians should be aware of these characteristics to help in the management of running injuries and advice on training.
Electrically active field-effect transistors (FET) based biosensors are of paramount importance in life science applications, as they offer direct, fast, and highly sensitive label-free detection ...capabilities of several biomolecules of specific interest. In this work, we report a detailed investigation on surface functionalization and covalent immobilization of biomarkers using biocompatible ethanolamine and poly(ethylene glycol) derivate coatings, as compared to the conventional approaches using silica monoliths, in order to substantially increase both the sensitivity and molecular selectivity of nanowire-based FET biosensor platforms. Quantitative fluorescence, atomic and Kelvin probe force microscopy allowed detailed investigation of the homogeneity and density of immobilized biomarkers on different biofunctionalized surfaces. Significantly enhanced binding specificity, biomarker density, and target biomolecule capture efficiency were thus achieved for DNA as well as for proteins from pathogens. This optimized functionalization methodology was applied to InP nanowires that due to their low surface recombination rates were used as new active transducers for biosensors. The developed devices provide ultrahigh label-free detection sensitivities ∼1 fM for specific DNA sequences, measured via the net change in device electrical resistance. Similar levels of ultrasensitive detection of ∼6 fM were achieved for a Chagas Disease protein marker (IBMP8-1). The developed InP nanowire biosensor provides thus a qualified tool for detection of the chronic infection stage of this disease, leading to improved diagnosis and control of spread. These methodological developments are expected to substantially enhance the chemical robustness, diagnostic reliability, detection sensitivity, and biomarker selectivity for current and future biosensing devices.
Introduction Le 26RFa est un neuropeptide orexigène, ligand endogène du récepteur GPR103. Des travaux réalisés chez l’animal suggèrent sa participation dans la physiopathologie de l’obésité. Notre ...travail vise à évaluer les concentrations plasmatiques du 26RFa chez des patients obèses et/ou diabétiques et l’implication du peptide dans la régulation glycémique. Patients et méthodes Un dosage plasmatique du 26RFa a été réalisé chez 161 sujets répartis en 4 groupes : contrôles normopondéraux normoglycémiques (n = 37), obèses non diabétiques (n = 52), patients DT1 (n = 19), patients DT2 (n = 53). Le 26RFa plasmatique a également été évalué au décours d’une hyperglycémie par voie orale (HGPO) chez des sujets contrôles. La localisation du 26RFa et du GPR103 a été recherchée sur des coupes pancréatiques humaines. Enfin, des tests de tolérance au glucose avec administration du 26RFa ont été réalisés chez la souris. Résultats La concentration plasmatique à jeun du 26RFa est significativement plus élevée chez les patients obèses comparativement aux contrôles (352 ± 47 vs 224 ± 32 pg/ml ; p < 0,05), et plus encore chez les patients DT1 et DT2 respectivement 605 ± 115 pg/ml, et 677 ± 99 pg/ml, p < 0,001 vs contrôles. L’analyse multivariée révèle une corrélation positive entre le 26RFa plasmatique et la glycémie à jeûn après ajustement avec l’âge, le sexe et l’IMC. La concentration plasmatique du 26RFa s’élève significativement (+ 57,3 % ; p < 0,05) à T120 min de l’HGPO chez les contrôles. Les études immunohistochimiques montrent la présence du 26RFa et de son récepteur dans les cellules béta des îlots de Langerhans. Enfin, l’injection ip de 26RFa au décours d’un test de tolérance au glucose chez la souris réduit l’hyperglycémie en augmentant significativement l’insulinémie. Conclusion Nous rapportons pour la première fois que le 26RFa est élevé chez les patients obèses et diabétiques en lien avec la glycémie à jeun. De plus, nous montrons que le peptide est impliqué dans l’homéostasie glucidique en régulant la sécrétion d’insuline.

Introduction Le neuropeptide 26RFa exerce une activité orexigène, via l’activation de son récepteur le GPR103, en régulant le système NPY/POMC de l’hypothalamus. L’expression hypothalamique du 26RFa ...est augmentée chez le rongeur soumis à un régime High Fat et nous avons déjà rapporté une augmentation des concentrations plasmatiques de ce peptide chez les patients obèses et diabétiques. L’objectif du présent travail est de déterminer l’origine du 26RFa plasmatique et son implication dans l’homéostasie du glucose. Matériels et méthodes un immunomarquage sur différents segments du tractus digestif de souris a été réalisé avec un anticorps dirigé contre le 26RFa. Les concentrations plasmatiques du 26RFa ont été déterminées par dosage radio-immunologique chez la souris après l’administration orale de glucose. La sécrétion intestinale du 26RFa lors d’une exposition au glucose a été évaluée in vitro à partir des différents segments de tractus digestif montés en chambre de Ussing. Enfin, les profils glycémique et insulinémique après administration de 26RFa (500 µg/kg) ont été déterminés au décours d’une charge intrapéritonéale en glucose (2 g/kg). Un test de tolérance à l’insuline avec le 26RFa a également été réalisé Résultats Des cellules à 26RFA présentant un phénotype neuroendocrine sont observées dans l’ensemble du tractus digestif. Les concentrations plasmatiques de 26RFa chez la souris sont significativement augmentées 30 minutes après l’administration orale de glucose. Parallèlement, le glucose induit in vitro une libération significative de 26RFa par l’intestin proximale. Enfin, le 26RFa diminue significativement l’hyperglycémie induite par une charge en glucose en augmentant la sensibilité à l’insuline et la sécrétion de l’insuline Conclusion L’ensemble de ces données suggère pour la première fois que le 26RFa d’origine digestive est impliqué dans la régulation glycémique via une activité de type incrétine. Attention deficit/hyperactivity disorder (ADHD) is a common neurodevelopmental psychiatric disorder. Genome-wide association studies (GWAS) have identified several loci associated with ADHD. However, ...understanding the biological relevance of these genetic loci has proven to be difficult. Here, we conduct an ADHD transcriptome-wide association study (TWAS) consisting of 19,099 cases and 34,194 controls and identify 9 transcriptome-wide significant hits, of which 6 genes were not implicated in the original GWAS. We demonstrate that two of the previous GWAS hits can be largely explained by expression regulation. Probabilistic causal fine-mapping of TWAS signals prioritizes KAT2B with a posterior probability of 0.467 in the dorsolateral prefrontal cortex and TMEM161B with a posterior probability of 0.838 in the amygdala. Furthermore, pathway enrichment identifies dopaminergic and norepinephrine pathways, which are highly relevant for ADHD. Overall, our findings highlight the power of TWAS to identify and prioritize putatively causal genes.
