Quercetin has been studied extensively for its anti-Alzheimer's disease (AD) and anti-aging effects. Our previous studies have found that quercetin and in its glycoside form, rutin, can modulate the ...proteasome function in neuroblastoma cells. We aimed to explore the effects of quercetin and rutin on intracellular redox homeostasis of the brain (reduced glutathione/oxidized glutathione, GSH/GSSG), its correlation with β-site APP cleaving enzyme 1 (BACE1) activity, and amyloid precursor protein (APP) expression in transgenic TgAPP mice (bearing human Swedish mutation APP transgene, APPswe). On the basis that BACE1 protein and APP processing are regulated by the ubiquitin-proteasome pathway and that supplementation with GSH protects neurons from proteasome inhibition, we investigated whether a diet containing quercetin or rutin (30 mg/kg/day, 4 weeks) diminishes several early signs of AD. Genotyping analyses of animals were carried out by PCR. In order to determine intracellular redox homeostasis, spectrofluorometric methods were adopted to quantify GSH and GSSG levels using o-phthalaldehyde and the GSH/GSSG ratio was ascertained. Levels of TBARS were determined as a marker of lipid peroxidation. Enzyme activities of SOD, CAT, GR, and GPx were determined in the cortex and hippocampus. ΒACE1 activity was measured by a secretase-specific substrate conjugated to two reporter molecules (EDANS and DABCYL). Gene expression of the main antioxidant enzymes: APP, BACE1, a Disintegrin and metalloproteinase domain-containing protein 10 (ADAM10), caspase-3, caspase-6, and inflammatory cytokines were determined by RT-PCR. First, overexpression of APPswe in TgAPP mice decreased GSH/GSSG ratio, increased malonaldehyde (MDA) levels, and, overall, decreased the main antioxidant enzyme activities in comparison to wild-type (WT) mice. Treatment of TgAPP mice with quercetin or rutin increased GSH/GSSG, diminished MDA levels, and favored the enzyme antioxidant capacity, particularly with rutin. Secondly, both APP expression and BACE1 activity were diminished with quercetin or rutin in TgAPP mice. Regarding ADAM10, it tended to increase in TgAPP mice with rutin treatment. As for caspase-3 expression, TgAPP displayed an increase which was the opposite with rutin. Finally, the increase in expression of the inflammatory markers IL-1β and IFN-γ in TgAPP mice was lowered by both quercetin and rutin. Collectively, these findings suggest that, of the two flavonoids, rutin may be included in a day-to-day diet as a form of adjuvant therapy in AD.
Knowledge of the neutrino flux produced by the Neutrinos at the Main Injector (NuMI) beamline is essential to the neutrino oscillation and neutrino interaction measurements of the MINERvA, MINOS+, ...NOvA and MicroBooNE experiments at Fermi National Accelerator Laboratory. We have produced a flux prediction which uses all available and relevant hadron production data, incorporating measurements of particle production off of thin targets as well as measurements of particle yields from a spare NuMI target exposed to a 120 GeV proton beam. The result is the most precise flux prediction achieved for a neutrino beam in the one to tens of GeV energy region. We have also compared the prediction to in situ measurements of the neutrino flux and find good agreement.
Muon-neutrino elastic scattering on electrons is an observable neutrino process whose cross section is precisely known. Consequently a measurement of this process in an accelerator-based nu sub(mu) ...beam can improve the knowledge of the absolute neutrino flux impinging upon the detector; typically this knowledge is limited to ~10% due to uncertainties in hadron production and focusing. We have isolated a sample of 135+ or -17 neutrino-electron elastic scattering candidates in the segmented scintillator detector of MINERvA, after subtracting backgrounds and correcting for efficiency. We show how this sample can be used to reduce the total uncertainty on the NuMI flux nu sub(mu) from 9% to 6%. Our measurement provides a flux constraint that is useful to other experiments using the NuMI beam, and this technique is applicable to future neutrino beams operating at multi-GeV energies.
Neutrino-induced coherent charged pion production on nuclei νμA→μ(±)π(∓)A is a rare, inelastic interaction in which a small squared four-momentum |t| is transferred to the recoil nucleus, leaving it ...intact in the reaction. In the scintillator tracker of MINERvA, we remove events with evidence of particles from nuclear breakup and reconstruct |t| from the final-state pion and muon. We select low |t| events to isolate a sample rich in coherent candidates. By selecting low |t| events, we produce a model-independent measurement of the differential cross section for coherent scattering of neutrinos and antineutrinos on carbon. We find poor agreement with the predicted kinematics in neutrino generators used by current oscillation experiments.
We report on multinucleon effects in low momentum transfer (<0.8 GeV/c) antineutrino interactions on plastic (CH) scintillator. These data are from the 2010-2011 antineutrino phase of the MINERvA ...experiment at Fermilab. The hadronic energy spectrum of this inclusive sample is well described when a screening effect at a low energy transfer and a two-nucleon knockout process are added to a relativistic Fermi gas model of quasielastic, Δ resonance, and higher resonance processes. In this analysis, model elements introduced to describe previously published neutrino results have quantitatively similar benefits for this antineutrino sample. We present the results as a double-differential cross section to accelerate the investigation of alternate models for antineutrino scattering off nuclei.
The MINERvA Collaboration reports a novel study of neutrino-nucleus charged-current deep inelastic scattering (DIS) using the same neutrino beam incident on targets of polystyrene, graphite, iron, ...and lead. Results are presented as ratios of C, Fe, and Pb to CH. The ratios of total DIS cross sections as a function of neutrino energy and flux-integrated differential cross sections as a function of the Bjorken scaling variable x are presented in the neutrino-energy range of 5-50 GeV. Based on the predictions of charged-lepton scattering ratios, good agreement is found between the data and prediction at medium x and low neutrino energy. However, the ratios appear to be below predictions in the vicinity of the nuclear shadowing region, x<0.1. This apparent deficit, reflected in the DIS cross-section ratio at high E sub(nu), is consistent with previous MINERvA observations B. Tice et al.(MINERvA Collaboration), Phys. Rev. Lett. 112, 231801 (2014). and with the predicted onset of nuclear shadowing with the axial-vector current in neutrino scattering.
The aim of this research is to evaluate the co-digestion of cow and llama manure combined with sheep manure, in psychrophilic conditions and real field low cost tubular digesters adapted to cold ...climate. Four digesters were monitored in cold climate conditions; one fed with cow manure, a second one with llama manure, the third one with co-digestion of cow-sheep manure and the fourth one was fed with llama-sheep manure. The slurry had a mean temperature of 16.6 °C, the organic load rate was 0.44 kgvs m(-3) d(-1) and the hydraulic retention time was 80 days. After one hundred days biogas production was stable, as was the methane content and the pH of the effluent. The co-digestion of cow-sheep manure results in a biogas production increase of 100% compared to the mono-digestion of cow manure, while co-digestion of llama-sheep manure results in a decrease of 50% in biogas production with respect to mono-digestion of llama manure.
The development of materials with special microstructures is a continuing theme of research due to their improved physical and mechanical properties. Rapid solidification can lead to amorphous or ...partially crystalline microstructures, depending on the imposed processing conditions and compositions allowing the production of composite materials. Al–Ni-Sm alloys can be easily obtained in the amorphous state by rapid solidification processing so that composites could be generated depending on the cooling conditions. In this work, new results are shown for Al-based amorphous matrix composites produced during rapid solidification of Al83.5Ni10Sm6.5, Al81.5Ni10Sm6.5Pb2 and Al83.5Ni8Sm6.5Pb2 (atom percent) alloys. Lead (Pb) was added to the amorphous forming composition alloy due to its immiscibility with the main alloying elements. Ribbon samples were obtained by melt spinning technique. Besides the use of ordinary characterization techniques, such as X-ray diffraction (XRD), differential scanning calorimetry (DSC) and scanning electron microscopy (SEM), advanced transmission electron microscopy (TEM) was employed to reveal the special microstructure features forming these alloys. A strong influence of the Pb addition on crystallization behavior has been identified. Also, the size and morphology of the nanocrystalline Pb-rich minority phase within the amorphous matrix is examined. These effects were explained based on the immiscibility of Pb with the alloy components.
We present measurements of ν(μ) charged-current cross section ratios on carbon, iron, and lead relative to a scintillator (CH) using the fine-grained MINERvA detector exposed to the NuMI neutrino ...beam at Fermilab. The measurements utilize events of energies 2<E(ν)<20 GeV, with ⟨E(ν)⟩ = 8 GeV, which have a reconstructed μ(-) scattering angle less than 17° to extract ratios of inclusive total cross sections as a function of neutrino energy E(ν) and flux-integrated differential cross sections with respect to the Bjorken scaling variable x. These results provide the first high-statistics direct measurements of nuclear effects in neutrino scattering using different targets in the same neutrino beam. Measured cross section ratios exhibit a relative depletion at low x and enhancement at large x. Both become more pronounced as the nucleon number of the target nucleus increases. The data are not reproduced by GENIE, a conventional neutrino-nucleus scattering simulation, or by the alternative models for the nuclear dependence of inelastic scattering that are considered.
The anelastic spectra of Cu39.5Zr51Al9.5, Cu54Zr40Al6 and Cu47.75Zr47.75Al4.5 bulk metallic glasses were obtained by Mechanical Spectroscopy Technique at different frequencies. Studies below room ...temperature show two principal mechanisms: one broad peak, similar to β relaxation, and another sharp peak, similar to α relaxation. This observation suggests that the interaction between amorphous structure and mechanical waves demonstrates that β′ relaxation can be decomposed in two principal peaks, one due to the movement of flow units and another owing to the growth of molecular-like structures. The behaviour of hybridization among Al-Cu atoms tends to decrease the free space in the amorphous matrix, increasing internal friction. The mechanical excitation and interaction among nanostructures such as cluster type, supercluster and molecular-like structure, can lead to growth of microalloy which may give rise to the medium-range order structure as well as increasing the small nanocrystalline region scattered into the glassy structure. In addition, the effect of mechanical vibration and cryogenic temperature acting on the amorphous structure can promote interaction among nanostructures resulting in a barrier that tends to block the propagation of shear bands in the metallic glasses.
