The use of biopharmaceuticals dates from the 19th century and within 5–10 years, up to 50% of all drugs in development will be biopharmaceuticals. In the 1980s, the biopharmaceutical industry ...experienced a significant growth in the production and approval of recombinant proteins such as interferons (IFN α, β, and γ) and growth hormones. The production of biopharmaceuticals, known as bioprocess, involves a wide range of techniques. In this review, we discuss the technology involved in the bioprocess and describe the available strategies and main advances in microbial fermentation and purification process to obtain biopharmaceuticals.
This paper contributes to a new Lagrangian vortex method for the statistical control of turbulence in two-dimensional flow configurations around a rough circular cylinder in ground effect when ...considering higher subcritical Reynolds numbers, namely 3 × 104 ≤ Re ≤ 2 × 105. A smoothed moving wall (active control technique) is used to include the blockage effect in association with the variation in cylinder surface roughness (passive control technique), characterizing a hybrid approach. In contrast with the previous approaches of our research group, the rough cylinder surface is here geometrically constructed, and a new momentum source term is introduced and calculated for the investigated problem. The methodology is structured by coupling the random Discrete Vortex Method, the Lagrangian Dynamic Roughness Model, and the Large Eddy Simulation with turbulence closure using the truncated Second-Order Velocity Structure Function model. This methodological option has the advantage of dispensing with the use of both a refined near-wall mesh and wall functions. The disadvantage of costly processing is readily solved with Open Multi-Processing. The results reveal that intermediate and high roughness values are most efficient for Reynolds numbers on the orders of 105 and 104, respectively. In employing a moving wall, the transition from the large-gap to the intermediate-gap regime is satisfactorily characterized. For the conditions studied with the hybrid technique, it was concluded that the effect of roughness is preponderant and acts to anticipate the characteristics of a lower gap-to-diameter ratio regime, especially with regard to intermittency.
l-asparaginase (l-asparagine amino hydrolase, E.C.3.5.1.1) is an enzyme clinically accepted as an antitumor agent to treat acute lymphoblastic leukemia and lymphosarcoma. It catalyzes l-asparagine ...(Asn) hydrolysis to l-aspartate and ammonia, and Asn effective depletion results in cytotoxicity to leukemic cells. Microbial l-asparaginase (ASNase) production has attracted considerable attention owing to its cost effectiveness and eco-friendliness. The focus of this review is to provide a thorough review on microbial ASNase production, with special emphasis to microbial producers, conditions of enzyme production, protein engineering, downstream processes, biochemical characteristics, enzyme stability, bioavailability, toxicity and allergy potential. Some issues are also highlighted that will have to be addressed to achieve better therapeutic results and less side effects of ASNase use in cancer treatment: (a) search for new sources of this enzyme to increase its availability as a drug; (b) production of new ASNases with improved pharmacodynamics, pharmacokinetics and toxicological profiles, and (c) improvement of ASNase production by recombinant microorganisms. In this regard, rational protein engineering, directed mutagenesis, metabolic flux analysis and optimization of purification protocols are expected to play a paramount role in the near future.

Multiple drug resistance (MDR) bacterial strains are responsible by 1.2 million of human deaths all over the world. The pathogens possess efficient enzymes which are able to mitigate the toxicity of ...reactive oxygen species (ROS) produced by some antibiotics and the host immune cells. Among them, the bacterial peroxiredoxin alkyl hydroperoxide reductase C (AhpC) is able to decompose efficiently several kinds of hydroperoxides. To decompose their substrates AhpC use a reactive cysteine residue (peroxidatic cysteine-CysP) that together with two other polar residues (Thr/Ser and Arg) comprise the catalytic triad of these enzymes and are involved in the substrate targeting/stabilization to allow a bimolecular nucleophilic substitution (SN2) reaction. Additionally to the high efficiency the AhpC is very abundant in the cells and present virulent properties in some bacterial species. Despite the importance of AhpC in bacteria, few studies aimed at using natural compounds as inhibitors of this class of enzymes. Some natural products were identified as human isoforms, presenting as common characteristics a bulk hydrophobic moiety and an α, β-unsaturated carbonylic system able to perform a thiol-Michael reaction. In this work, we evaluated two chemically related natural products: 1,4-dihydroxy-2-(3',7'-dimethyl-1'-oxo-2'E,6'-octadienyl) benzene (C1) and 4-hydroxy-2-(3',7'-dimethyl-1'-oxo-2'E,6'-octadienyl) benzoic acid (C2), both were isolated from branches Piper crassinervium (Piperaceae), over the peroxidase activity of AhpC from Pseudomonas aeruginosa (PaAhpC) and Staphylococcus epidermidis (SeAhpC). By biochemical assays we show that although both compounds can perform the Michael addition reaction, only compound C2 was able to inhibit the PaAhpC peroxidase activity but not SeAhpC, presenting IC50 = 20.3 μM. SDS-PAGE analysis revealed that the compound was not able to perform a thiol-Michael addition, suggesting another inhibition behavior. Using computer-assisted simulations, we also show that an acidic group present in the structure of compound C2 may be involved in the stabilization by polar interactions with the Thr and Arg residues from the catalytic triad and several apolar interactions with hydrophobic residues. Finally, C2 was not able to interfere in the peroxidase activity of the isoform Prx2 from humans or even the thiol proteins of the Trx reducing system from Escherichia coli (EcTrx and EcTrxR), indicating specificity for P. aeruginosa AhpC.

The present paper introduces the key ideas of a purely Lagrangian temperature particle method, which includes preheating effects on fluid flow nearest a heated wall. The numerical approach is then ...applied for the study of mixed heat transfer on aircraft wake vortices from wing tips in the vicinity of a heated ground plane, a situation commonly found during landing or takeoff operations at airports around the world. It was found in the literature experimental results of an investigation without the effects of heat transfer and crosswind, which were useful for a comparison with some present numerical results. Other numerical results are also discussed, focusing on the physics of the effects of mixed convection heat transfer and crosswind. As a contribution, the Richardson number is defined in terms of both aircraft wingspan and constant ground plane temperature, being the most important dimensionless group to capture the effects of laminar ascending mixed convection flow. The present methodology presents potentialities for predicting the transport and decay of primary vortical structures (under buoyancy forces), including their interaction with secondary vortical structures generated from a ground plane.

The present study focuses on the fractionation of cashew apple bagasse via a pretreatment using acetic acid as a delignifying agent and sulfuric acid as an external catalyst. As expected, the concentrations of both acids and the incubation time dramatically affected delignification and hemicellulose solubilization. Under the optimal pretreatment conditions, recycling of the spent liquor had no apparent impact on the chemical composition of the pretreated material, yield of sugar produced via enzymatic hydrolysis (∼37 g/L reducing sugars at 7.5% (w/v) solid loading), or yield of ethanol obtained via fermentation with Saccharomyces cerevisiae (∼16 g/L at 10% (w/v) solid loading). The lignin recovered from the spent liquor showed a good ultraviolet protective effect; the addition of 5% (w/w) of the biopolymer increased the sun protection factor of a commercial sunscreen lotion from 21.62 to 40.71. The combined use of hydrogen peroxide and ultraviolet radiation reduced the organosolv lignin color (absorbance at 450 nm was four times lower) owing to aromatic ring cleavage, but cosmetics containing whitened organosolv lignin had low sun protection factor values. In summary, the results obtained in this study demonstrate the utility of organic acid pretreatment in the valorization of lignocellulosic materials.
A discrete vortex method is implemented with a hybrid control technique of vortex shedding to solve the problem of the two-dimensional flow past a slightly rough circular cylinder in the vicinity of ...a moving wall. In the present approach, the passive control technique is inspired on the fundamental principle of surface roughness, promoting modifications on the cylinder geometry to affect the vortex shedding formation. A relative roughness size of ε*/d* = 0.001 (ε* is the average roughness and d* is the outer cylinder diameter) is chosen for the test cases. On the other hand, the active control technique uses a wall plane, which runs at the same speed as the free stream velocity to contribute with external energy affecting the fluid flow. The gap-to-diameter varies in the range from h*/d* = 0.05 to 0.80 (h* is the gap between the moving wall and the cylinder bottom). A detailed account of the time history of pressure distributions, simultaneously investigated with the time evolution of forces, Strouhal number behavior, and boundary layer separation are reported at upper-subcritical Reynolds number flows of Re = 1.0 × 105. The saturation state of the numerical simulations is demonstrated through the analysis of the Strouhal number behavior obtained from temporal history of the aerodynamic loads. The present work provides an improvement in the prediction of Strouhal number than other studies no using roughness model. The aerodynamic characteristics of the cylinder, as well as the control of intermittence and complete interruption of von Kármán-type vortex shedding have been better clarified.
Memory gaps in intensive care unit (ICU) survivors are associated with psychiatric disorders. The ICU diaries improve the patient's factual memory of the ICU, but it is not clear if they reduce the ...incidence of psychiatric disorders in patients and relatives after hospital discharge. The aim of this study is to evaluate the literature on the effect of ICU diaries for patients admitted in ICU and their relatives.
Two authors independently searched the online databases PubMed, OVID, Embase, EBSCO host, and PsycINFO from inception to July 2019. Studies were included if the intervention group (ICU diary) was compared with a group with no diaries and the sample was comprised patients ≥ 18 years old admitted in the ICU for more than 24 h and their relatives. Randomized clinical trials, observational studies, letter with original data, and abstracts were included, irrespective of the language. The search was not limited by any specific outcome. Review articles, commentaries, editorials, and studies without a control group were excluded. Structured tools were used to assess the methodological quality ("Risk Of Bias In Non-randomized Studies of Interventions (ROBINS-I)" for cohort studies and the "Cochrane Risk of Bias tool" for included RCTs and before/after studies). A random-effects model was employed considering the anticipated variability between the studies.
Seven hundred eighty-five titles were identified for screening. Two additional studies were selected after a reference search, and after a full-text review, a total of 12 studies were included. When pooling the results, ICU diary was associated with lower risk of depression (RR 0.41, 95% CI 0.23-0.75) and better quality of life (10.3 points higher in SF-36 general health score, 95% CI 0.79-19.8), without a decrease in anxiety or post-traumatic stress disorder (PTSD). For the relatives receiving an ICU diary, there was no difference in the incidence of PTSD, anxiety, or depression.
This systematic review and meta-analysis supports the use of ICU diaries to reduce the risk of depression and preserve the quality of life of patients after ICU admission. ICU diaries do not seem to have any beneficial effect on the relatives of the patients.

American visceral leishmaniosis (AVL) or visceral leishmaniosis (VL) has been known as the most severe form of the disease. However, despite the growing number of people exposed to the infection risk and the great effort done by the scientific community worldwide to significantly increase the knowledge about these diseases, there is no vaccine capable of preventing VL in humans. In this short review, we present some of the plasmids used for the expression of recombinant protein by
Escherichia coli
strains used mainly for the second generation of vaccines for leishmaniosis. It can be emphasized that currently, these vectors and hosts play an important role in developing vaccine strategies against the disease. Indeed, use of the
E. coli
BL21 (DE) strain is remarkable mainly due to its characteristics for being a stable protein producer as well as the use of histidine tags for antigen purification.

Propolis is a balsamic product obtained from vegetable resins by exotic Africanized bees Apis mellifera L., transported and processed by them, originating from the activity that explores and ...maintains these individuals. Because of its vegetable and natural origins, propolis is a complex mixture of different compound classes; among them are the volatile compounds present in the aroma. In this sense, in the present study we evaluated the volatile fraction of propolis present in the aroma obtained by distillation and simultaneous extraction, and its chemical composition was determined using coupled gas chromatography, mass spectrometry, and flame ionization detection. The majority of compounds were sesquiterpene and hydrocarbons, comprising 8.2–22.19% α-copaene and 6.2–21.7% β-caryophyllene, with additional compounds identified in greater concentrations. Multivariate analysis showed that samples collected from one region may have different chemical compositions, which may be related to the location of the resin’s production. This may be related to other bee products.
