Abstract Background and aims The positive and negative health effects of dietary carbohydrates are of interest to both researchers and consumers. Methods International experts on carbohydrate ...research held a scientific summit in Stresa, Italy, in June 2013 to discuss controversies surrounding the utility of the glycemic index (GI), glycemic load (GL) and glycemic response (GR). Results The outcome was a scientific consensus statement which recognized the importance of postprandial glycemia in overall health, and the GI as a valid and reproducible method of classifying carbohydrate foods for this purpose. There was consensus that diets low in GI and GL were relevant to the prevention and management of diabetes and coronary heart disease, and probably obesity. Moderate to weak associations were observed for selected cancers. The group affirmed that diets low in GI and GL should always be considered in the context of diets otherwise understood as healthy, complementing additional ways of characterizing carbohydrate foods, such as fiber and whole grain content. Diets of low GI and GL were considered particularly important in individuals with insulin resistance. Conclusions Given the high prevalence of diabetes and pre-diabetes worldwide and the consistency of the scientific evidence reviewed, the expert panel confirmed an urgent need to communicate information on GI and GL to the general public and health professionals, through channels such as national dietary guidelines, food composition tables and food labels.
Degenerative disorders of motor neurons include a range of progressive fatal diseases such as amyotrophic lateral sclerosis (ALS), spinal-bulbar muscular atrophy (SBMA), and spinal muscular atrophy ...(SMA). Although the causative genetic alterations are known for some cases, the molecular basis of many SMA and SBMA-like syndromes and most ALS cases is unknown. Here we show that missense point mutations in the cytoplasmic dynein heavy chain result in progressive motor neuron degeneration in heterozygous mice, and in homozygotes this is accompanied by the formation of Lewy-like inclusion bodies, thus resembling key features of human pathology. These mutations exclusively perturb neuron-specific functions of dynein.

In the parton shower framework, data favour the Lund model ansatz and corresponding values of its parameters have been determined within PYTHIA 6.156 from DELPHI data:

A detailed study of hadronic interactions is presented using data recorded with the highly granular CALICE silicon–tungsten electromagnetic calorimeter. Approximately 350,000 selected π− events at ...energies between 2 and 10GeV have been studied. The predictions of several physics models available within the GEANT4 simulation tool kit are compared to this data. A reasonable overall description of the data is observed; the Monte Carlo predictions are within 20% of the data, and for many observables much closer. The largest quantitative discrepancies are found in the longitudinal and transverse distributions of reconstructed energy.
Insulin resistance of skeletal muscle glucose disposal underlies the pathogenesis of NIDDM and is associated with hypertension, obesity, and dyslipidemia. Angiotensin-converting enzyme (ACE) ...inhibitors are used primarily in antihypertensive therapy but also are known to improve whole-body insulin-mediated glucose disposal. However, the exact site of action is not well characterized. We have used the isolated epitrochlearis muscle from a well-established animal model of skeletal muscle insulin resistance, the obese Zucker rat, to test the effect of oral administration of ACE inhibitors on insulin-sensitive muscle glucose transport activity. Both acute and chronic administration of a sulfhydryl-containing ACE inhibitor (captopril) or a non-sulfhydryl-containing ACE inhibitor (tran-dolapril) significantly enhanced in vitro insulin-mediated muscle glucose transport activity. In addition, the acute effect of oral captopril administration was completely abolished by pretreatment of the animal with a bradykinin B2 receptor antagonist (HOE 140). These findings indicate that ACE inhibitors may improve whole-body glucose metabolism by acting on the insulin-sensitive skeletal muscle glucose transport system. In addition, bradykinin or one of its metabolites may be involved in the action of the ACE inhibitor captopril on insulin-resistant muscle.

An international multidisciplinary panel of experts prepared evidenced-based World Society of Emergency Surgery (WSES) guidelines for management of CDI in surgical patients.
Cleavage of amyloid precursor protein (APP) by the Alzheimer's β-secretase (BACE1) is a key step in generating amyloid β-peptide, the main component of amyloid plaques. Here we report evidence that ...heparan sulfate (HS) interacts with β-site APP-cleaving enzyme (BACE) 1 and regulates its cleavage of APP. We show that HS and heparin interact directly with BACE1 and inhibit in vitro processing of peptide and APP substrates. Inhibitory activity is dependent on saccharide size and specific structural characteristics, and the mechanism of action involves blocking access of substrate to the active site. In cellular assays, HS specifically inhibits BACE1 cleavage of APP but not alternative cleavage by α-secretase. Endogenous HS immunoprecipitates with BACE1 and colocalizes with BACE1 in the Golgi complex and at the cell surface, two of its putative sites of action. Furthermore, inhibition of cellular HS synthesis results in enhanced BACE1 activity. Our findings identify HS as a natural regulator of BACE1 and suggest a novel mechanism for control of APP processing.
Seafloor backscatter mosaics are now routinely produced from multibeam echosounder data and used in a wide range of marine applications. However, large differences (>5 dB) can often be observed ...between the mosaics produced by different software packages processing the same dataset. Without transparency of the processing pipeline and the lack of consistency between software packages raises concerns about the validity of the final results. To recognize the source(s) of inconsistency between software, it is necessary to understand at which stage(s) of the data processing chain the differences become substantial. To this end, willing commercial and academic software developers were invited to generate intermediate processed backscatter results from a common dataset, for cross-comparison. The first phase of the study requested intermediate processed results consisting of two stages of the processing sequence: the one-value-per-beam level obtained after reading the raw data and the level obtained after radiometric corrections but before compensation of the angular dependence. Both of these intermediate results showed large differences between software solutions. This study explores the possible reasons for these differences and highlights the need for collaborative efforts between software developers and their users to improve the consistency and transparency of the backscatter data processing sequence.
Ruminant livestock systems contribute to global warming through the emission of nitrous oxide (N
2O), methane (CH
4) and carbon dioxide (CO
2). This paper discusses a general framework for a ...whole-farm approach to develop cost-effective GHG mitigation strategies. A dairy farm is a complex system with different interacting components. Generally, whole-farm approaches distinguish at least an animal component and a soil–crop component. Whole-farm models should be able to give an accurate representation of the internal cycling of materials and its constituents as well as the exchange between the farming system and its environment. The paper gives an overview of current whole-farm models that are able to simulate GHG emissions for dairy farms. These models are DairySim, FarmGHG, SIMS
DAIRY and FarmSim. All models are able to calculate CH
4 and N
2O emissions, but differences appear in the ability to calculate CO
2 emissions, economics and other parameters. The effects of selected mitigation strategies are demonstrated with some of the models. It is concluded that a whole-farm approach is a powerful tool for the development of cost-effective GHG mitigation options as it reveals relevant interactions between farm components. Model calculations underlined the relationship between farm gate N surplus and GHG emissions, and thus the possibility to use N surpluses as an indicator for GHG emissions.
While spirometry and particularly airflow limitation is still considered as an important tool in therapeutic decision making, it poorly reflects the heterogeneity of respiratory impairment in chronic ...obstructive pulmonary disease (COPD). The aims of this study were to identify pathophysiological clusters in COPD based on an integrated set of standard lung function attributes and to investigate whether these clusters can predict patient-related outcomes and differ in clinical characteristics.
Clinically stable COPD patients referred for pulmonary rehabilitation underwent an integrated assessment including clinical characteristics, dyspnea score, exercise performance, mood and health status, and lung function measurements (post-bronchodilator spirometry, body plethysmography, diffusing capacity, mouth pressures and arterial blood gases). Self-organizing maps were used to generate lung function based clusters.
Clustering of lung function attributes of 518 patients with mild to very severe COPD identified seven different lung function clusters. Cluster 1 includes patients with better lung function attributes compared to the other clusters. Airflow limitation is attenuated in clusters 1 to 4 but more pronounced in clusters 5 to 7. Static hyperinflation is more dominant in clusters 5 to 7. A different pattern occurs for carbon monoxide diffusing capacity, mouth pressures and for arterial blood gases. Related to the different lung function profiles, clusters 1 and 4 demonstrate the best functional performance and health status while this is worst for clusters 6 and 7. All clusters show differences in dyspnea score, proportion of men/women, age, number of exacerbations and hospitalizations, proportion of patients using long-term oxygen and number of comorbidities.
Based on an integrated assessment of lung function variables, seven pathophysiological clusters can be identified in COPD patients. These clusters poorly predict functional performance and health status.
