The aim of the study was to evaluate the effects of low to moderate oral exposure to the
Fusarium
toxin deoxynivalenol (DON; derived from culture material) on performance, water intake, and carcass ...parameters of broilers during early and late developmental phases. A total of 160 Ross 308 broilers were randomly allocated to four different feeding groups (
n
= 40/group) including 0 (control), 2.5, 5, and 10 mg DON/kg wheat-soybean meal-based feed. Three consecutive replicates of the experiment were performed. Half of the broilers were slaughtered in week 3 of the trial whereas the other half were slaughtered in week 5. Dry matter intake (DMI) and water intake (WI) were recorded on a daily basis and the body weight (BW) and BW gain (BWG) were determined weekly. The following carcass traits were recorded and calculated in absolute and relative data: dressed carcass weight, breast muscle weight, leg weight, and liver weight. Data showed that BW (
P
< 0.001), BWG (
P =
0.005), and DMI (
P
< 0.001) were reduced by DON-feeding during the entire feeding period. The ratio of DMI to body weight gain (DMI/BWG) was not affected by the treatment. However, the ratio of water to DMI (WI/DMI) increased in DON-treated birds (
P =
0.021). Contrast analysis showed that DON tendentially reduced slaughter weight (
P
= 0.082) and decreased leg yield (
P
= 0.037) in DON-fed chickens in week 5 of the experiment. Liver organ weight decreased in the 3-week-old DON-fed broilers compared to that in the control-fed birds (
P
= 0.037). In conclusion, the study suggests that DMI and BW were negatively affected under the experimental conditions at DON levels lower than the current guidance value in the European Union of 5 mg/kg feed. The study also indicates that broilers fed on low to moderate level DON-contaminated diets showed increased WI/DMI ratio which might have negative influence on wet litter syndrome.
ABSTRACT
Although acute mycotoxicoses are rare in poultry production, chronic exposure to low levels of mycotoxins is responsible for reduced productivity and increased susceptibility to infectious ...diseases. Deoxynivalenol (DON) is known to modulate immune function, but only a few studies have investigated the effect of DON on the vaccinal immune response. In addition, the effects of Mycofix select (Biomin GmbH, Herzogenburg, Austria) supplementation to DON-contaminated broiler diets have not yet been demonstrated. Therefore, an experiment with 1-d-old male broilers (Ross 308) was carried out to examine the effects of feeding DON-contaminated low-protein grower diets on performance, serum biochemical parameters, lymphoid organ weight, and antibody titers to infectious bronchitis vaccination in serum and to evaluate the effects of Mycofix select dietary supplementation in either the presence or absence of DON in broilers. In total, thirty-two 1-d-old broiler chicks were randomly assigned to 1 of the 4 dietary treatments for 5 wk. The dietary treatments were 1) control; 2) artificially contaminated diets with 10 mg of DON/kg of diet; 3) DON-contaminated diets supplemented with Mycofix select; and 4) control diet supplemented with Mycofix select. Feeding of contaminated diets decreased (P = 0.000) the feed intake, BW (P = 0.001), BW gain (P = 0.044), and feed efficiency during the grower phase. Deoxynivalenol affected the blood biochemistry, whereas plasma total protein and uric acid concentrations in birds fed contaminated grains were decreased compared with those of the controls. Moreover, in birds fed contaminated feeds, there was a tendency to reduce triglycerides in the plasma (P = 0.090), suggesting that DON in the diets affected protein and lipid metabolism in broiler chickens. The feeding of contaminated diets altered the immune response in broilers by reducing the total lymphocyte count. Similarly, the antibody response against infectious bronchitis vaccination antigens was decreased (P = 0.003) after feeding contaminated diets, compared with the controls. Moreover, contamination of the broiler diet with DON increased the heteropil:lymphocyte ratio (stress index), suggesting that DON elevated the physiological stress responses of broilers. However, feeding of DON-containing diets did not alter the other plasma constituents, including activities of enzymes. Mycofix select addition to the DON-contaminated feed led to normal immunological and physiological functions in broilers that were comparable with those of the control group, indicating that the addition of the additive to the DON-contaminated feed of the broilers effectively alleviated the alterations caused by DON. It was concluded that broiler performance and some blood and immunological parameters were adversely affected by feeding diets contaminated with the Fusarium mycotoxin DON. However, the dietary Mycofix select supplementation as a detoxifying agent was successful in overcoming the mycotoxin-related effects.
The present trial was conducted to study some morphological, digestive, and electrophysiological variables of the small intestine during chronic exposure of broilers to aflatoxin B(1) (AFB(1)). Ross ...308 male chicks (7 d old) were randomly allotted to control (no AFB(1)), low AFB(1) (0.07 mg of AFB(1)/kg), or high AFB(1) (0.75 mg of AFB(1)/kg) diet. The high AFB(1) diet resulted in reduced (P ≤ 0.002) bird performance during the first 4 wk of exposure, whereas the low AFB(1) diet temporarily reduced (P = 0.034) the bird performance during wk 3 of exposure. During wk 4 of exposure, a linear (P ≤ 0.013) decrease in the unit weight of both the duodenum and jejunum was observed with increasing levels of AFB(1). This reduction in unit weight appeared to progress from the proximal (duodenum) to the distal (jejunum) small intestine with increase in the length of exposure and was not accompanied by modulation of electrophysiological variables in jejunal epithelium. Response from amiloride, a specific blocker of epithelial sodium channel, was also similar among jejunal epithelia of birds under different treatments. Interestingly, a compensatory linear (P ≤ 0.002) increase in the length of the duodenum and jejunum under high AFB(1) diets was noted to occur during wk 4 of exposure. Thus, retention of DM and nitrogen was not negatively affected by the AFB(1) diets. These data indicate that the intestine in broilers may adapt to an ongoing dietary challenge to AFB(1).
It is shown that the puzzling experimental observation of suppressed molecular ionization, in intense laser fields, of O2, and its absence in N2, is a symmetry induced dynamical effect. More ...generally, it is predicted that the ionization signal of the class of homonuclear diatomic molecules having valence orbitals with an antibonding symmetry (e.g., sigma(u), pi(g)) would be suppressed, but not those with a bonding symmetry (e.g., sigma(g), pi(u)). The suppression effect can be visualized as due to an effective destructive interference between the two subwaves of the ionizing electron emerging from the two atomic centers.
The role of dried distillers grains plus solubles (DDGS) and associative effects of different levels of grape seed meal (GSM) fortified in DDGS, used as both protein and energy sources in the diet, ...on ruminal fermentation and microbiota were investigated using rumen-simulation technique. All diets consisted of hay and concentrate mixture with a ratio of 48:52 dry matter (DM) basis, but were different in the concentrate composition. The control diet contained soybean meal (13.5% of diet DM) and barley grain (37%), whereas DDGS treatments, unfortified DDGS (19.5% of diet DM), or DDGS fortified with GSM, either at 1, 5, 10, or 20% were used entirely in place of soybean meal and part of barley grain at a 19.5 to 25% inclusion level. All diets had similar DM, organic matter, and crude protein contents, but consisted of increasing neutral detergent fiber and decreasing nonfiber carbohydrates levels with DDGS-GSM inclusion. Compared with the soy-based control diet, the unfortified DDGS treatment elevated ammonia concentration (19.1%) of rumen fluid associated with greater crude protein degradation (~19.5%). Methane formation decreased with increasing GSM fortification levels (≥5%) in DDGS by which the methane concentration significantly decreased by 18.9 to 23.4 and 12.8 to 17.6% compared with control and unfortified DDGS, respectively. Compared with control, unfortified DDGS decreased butyrate proportion, and GSM fortification in the diet further decreased this variable. The proportions of genus Prevotella and Clostridium cluster XIVa were enhanced by the presence of DDGS without any associative effect of GSM fortification. The abundance of methanogenic archaea was similar, but their composition differed among treatments; whereas Methanosphaera spp. remained unchanged, proportion of Methanobrevibacter spp. decreased in DDGS-based diets, being the lowest with 20% GSM inclusion. The abundance of Ruminococcus flavefaciens, anaerobic fungi, and protozoa were decreased by the GSM inclusion. As revealed by principal component analysis, these variables were the microorganisms associated with the methane formation. Grape seed meal fortification level in the diet decreased DM and organic matter degradation, but this effect was more related to a depression of nonfiber carbohydrates degradation. It can be concluded that DDGS fortified with GSM can favorably modulate ruminal fermentation.
Chicory (Cichorium intybus) belongs to plants of the Compositae family accumulating energy in the form of inulin fructan. Chicory, a prebiotic, is a fermentable oligosaccharide and oligofructose that ...may affect the intestinal mucosal architecture and the electrophysiological parameters. Therefore, this study was conducted to evaluate the effectiveness of adding chicory fructans in feed on the intestinal morphology and electrogenic transport of glucose in broilers. Four hundred, 1 day old broiler chicks were randomly divided into two groups (200 bird per group) for 5 weeks. The dietary treatments were (i) control, (ii) basal diets supplemented with the dried, grinded ground chicory pulp containing inulin (1 kg of chicory/ton of the starter and grower diets). In duodenum, dietary chicory increased the villus height and villus width and villus height to crypt depth ratio (p < 0.05), but the duodenal crypt depth remained unaffected (p > 0.05). However, in jejunum, the villus height, crypt depth and villus height to crypt depth ratio were decreased by dietary chicory compared with control birds (p < 0.05). In ileum, the villus height and villus crypt depth was decreased by dietary chicory supplementation compared with control (p < 0.05), but, the villus height to crypt depth ratio was increased (p < 0.05). Moreover, dietary chicory relatively affected the electrophysiological parameters of the intestine but did not reach significance. The amount of ΔIsc after d-glucose addition to the jejunal mucosa was numerically higher for chicory fed birds (19 μA/cm²) than control birds (10 μA/cm²). The percentage of increase in the Isc after d-glucose addition (ΔIsc %) was higher for chicory group upto (90%) of the control group. In colon, the actual Isc value and Isc after d-glucose addition was numerically higher for chicory fed birds than control birds (p > 0.05). Moreover, the conductance of jejunal and colonic tissues after d-glucose addition remained unaffected by the dietary chicory. In conclusion, addition of chicory to broilers diet increased the duodenal villus height, villus width and villus height to crypt depth ratio and decreased the villus height and crypt depth in both jejenum and ileum. Furthermore, dietary chicory relatively modified the small intestinal electrogenic transport of glucose in broilers.
Background and Aims:
The principle of complete mesocolic excision for colon cancer has been introduced to improve oncologic outcome. However, this approach is scantily discussed for laparoscopic ...surgery and there is a lack of randomized trials. This study examined oncologic and clinical outcome after laparoscopic wide mesocolic excision and central vascular ligation for colon cancer.
Material and Methods:
This is a review of prospectively gathered data from a single-institution colorectal cancer database. This study was conducted in the Central Hospital of Central Finland. From January 2003 to December 2011, 222 patients underwent laparoscopic colonic resections with wide mesocolic excision and central vascular ligation in the multimodal setting. The main measures of outcome were cancer recurrence and survival, with early recovery, 30d-mortality and morbidity, reoperation, readmission, and late complications as secondary outcomes.
Results:
The median follow-up was 5.5 (interquartile range (IQR) = 3.7–8.0) years. The 5-year overall survival for all 222 patients was 80.2% and disease-specific survival was 87.5%, and for those 210 R0-patients with stage I–III disease, 83.9% and 91.3%, respectively. The 5-year disease-free survival was 85.8%: stage I was 94.7%, stage II was 90.8%, and stage III was 75.6% (p = 0.004). Increasing lymph node ratio significantly decreased the 5-year disease-free survival. Conversion rate to open surgery was 12.2%. Thirty-day mortality was 1.3% and morbidity, 19.7%. Median postoperative hospital stay was 5 (IQR = 3–7) days.
Conclusion:
Laparoscopic wide mesocolic excision and central vascular ligation for colon cancer resulted in good long-term oncologic outcome. Randomized trials are needed to show that laparoscopic complete mesocolic excision technique would become the standard of care for the carcinoma of the colon.
Probiotics might be one of the solutions to reduce the effects of the recent ban on antimicrobial growth promoters in feed. However, the mode of action of probiotics still not fully understood. ...Therefore, evaluating probiotics (microbial feed additives) is essential. Thus the objective of this work was to investigate the efficacy of a new microbial feed additive (Lactobacillus salivarius and Lactobacillus reuteri) in broiler nutrition. The body weight (BW), average daily weight gain was relatively increased by the dietary inclusion of Lactobacillus sp. in broiler diets. Furthermore, the Lactobacillus feed additive influenced the histomorphological measurements of small intestinal villi. The addition of Lactobacillus sp. increased (p < 0.05) the villus height (VH)/crypt depth ratio and the VH was numerically increased in duodenum. The duodenal crypt depth remained unaffected (p > 0.05), while the ileal crypt depth was decreased by dietary supplementation of Lactobacillus sp. compared with the control. At the end of the feeding period, the basal and glucose stimulated short-circuit current (Isc) and electrical tissue conductivity were measured in the isolated gut mucosa to characterize the electrical properties of the gut. The addition of glucose on the mucosal side in Ussing chamber produced a significant increase (p = 0.001) in Isc in both jejunum and colon relative to the basal values in Lactobacillus probiotic group. This increase in Isc for probiotic group in jejunum is equivalent to an increase of about two times that for the basal values, while in the control group is about half fold that for the basal value. In addition, the ΔIsc after glucose addition to the large intestine was greater than the ΔIsc in the small intestine in both control and probiotic group. Moreover in both jejunum and colon, the increase in Isc for birds fed Lactobacillus was higher than their control counterparts (p less-than or equal to 0.1). This result suggests that the addition of Lactobacillus sp. to broiler diets increased the glucose transport. Additionally, the results indicated that the conductivity of jejunal and colonic tissues remained unaffected by the dietary inclusion of Lactobacillus and support the concept that this additive enhances the maintenance and function of the epithelial barrier. In conclusion, dietary inclusion of a microbial feed additive (L. salivarius and L. reuteri) slightly increased the growth performance and improved intestinal nutrient absorption which was in association with the intestinal architecture improvement.
Recent data suggest that Fusarium trichothecenes may reduce broiler performance at levels previously thought not to affect this variable in chickens. In the present study, we investigated the effects ...of deoxynivalenol (DON), a type-B trichothecene, on broilers. Male broilers at 7 d of age were fed either a basal diet (0.265 ± 0.048 mg of DON; 0.013 ± 0.001 mg of zearalenone/kg), a low DON diet (1.68 mg of DON/kg; 0.145 ± 0.007 mg of zearalenone/kg), or a high DON diet (12.209 ± 1.149 mg of DON/kg; 1.094 ± 0.244 mg of zearalenone/kg). Increasing levels of DON decreased the weekly weight gain linearly (P ≤ 0.041) during the first 3 wk of exposure; there were no significant differences in the weight gain of the birds after wk 3. With increasing levels of DON, the titers against Newcastle disease virus increased linearly during wk 2 (P = 0.022) and wk 4 (P = 0.033) of exposure, whereas the titers against infectious bronchitis virus decreased linearly (P = 0.006) during wk 5 of exposure. The serum protein concentration increased linearly (P = 0.017) during wk 2 and quadratically (P = 0.002) during wk 4 of exposure. Under these experimental conditions, the performance and vaccine response of the broilers were modulated to varying degrees at concentrations of DON that are currently permitted (up to 5 mg/kg of diet) in many countries. Further studies are therefore required to clarify the implications of these results on the welfare of chickens.

The
AUSTRAL
observing program was started in 2011, performing geodetic and astrometric very long baseline interferometry (VLBI) sessions using the new Australian AuScope VLBI antennas at Hobart, ...Katherine, and Yarragadee, with contribution from the Warkworth (New Zealand) 12 m and Hartebeesthoek (South Africa) 15 m antennas to make a southern hemisphere array of telescopes with similar design and capability. Designed in the style of the next-generation VLBI system, these small and fast antennas allow for a new way of observing, comprising higher data rates and more observations than the standard observing sessions coordinated by the International VLBI Service for Geodesy and Astrometry (IVS). In this contribution, the continuous development of the
AUSTRAL
sessions is described, leading to an improvement of the results in terms of baseline length repeatabilities by a factor of two since the start of this program. The focus is on the scheduling strategy and increased number of observations, aspects of automated operation, and data logistics, as well as results of the 151
AUSTRAL
sessions performed so far. The high number of the
AUSTRAL
sessions makes them an important contributor to VLBI end-products, such as the terrestrial and celestial reference frames and Earth orientation parameters. We compare
AUSTRAL
results with other IVS sessions and discuss their suitability for the determination of baselines, station coordinates, source coordinates, and Earth orientation parameters.
