Endurance exercise training studies frequently show modest changes in VO2max with training and very limited responses in some subjects. By contrast, studies using interval training (IT) or combined ...IT and continuous training (CT) have reported mean increases in VO2max of up to ~1.0 L · min(-1). This raises questions about the role of exercise intensity and the trainability of VO2max. To address this topic we analyzed IT and IT/CT studies published in English from 1965-2012. Inclusion criteria were: 1)≥ 3 healthy sedentary/recreationally active humans <45 yrs old, 2) training duration 6-13 weeks, 3) ≥ 3 days/week, 4) ≥ 10 minutes of high intensity work, 5) ≥ 1:1 work/rest ratio, and 6) results reported as mean ± SD or SE, ranges of change, or individual data. Due to heterogeneity (I(2) value of 70), statistical synthesis of the data used a random effects model. The summary statistic of interest was the change in VO2max. A total of 334 subjects (120 women) from 37 studies were identified. Participants were grouped into 40 distinct training groups, so the unit of analysis was 40 rather than 37. An increase in VO2max of 0.51 L · min(-1) (95% CI: 0.43 to 0.60 L · min(-1)) was observed. A subset of 9 studies, with 72 subjects, that featured longer intervals showed even larger (~0.8-0.9 L · min(-1)) changes in VO2max with evidence of a marked response in all subjects. These results suggest that ideas about trainability and VO2max should be further evaluated with standardized IT or IT/CT training programs.

Escape responses are used by many animal species as their main defence against predator attacks. Escape success is determined by a number of variables; important are the directionality (the ...percentage of responses directed away from the threat) and the escape trajectories (ETs) measured relative to the threat. Although logic would suggest that animals should always turn away from a predator, work on various species shows that these away responses occur only approximately 50-90% of the time. A small proportion of towards responses may introduce some unpredictability and may be an adaptive feature of the escape system. Similar issues apply to ETs. Theoretically, an optimal ET can be modelled on the geometry of predator-prey encounters. However, unpredictability (and hence high variability) in trajectories may be necessary for preventing predators from learning a simple escape pattern. This review discusses the emerging trends in escape trajectories, as well as the modulating key factors, such as the surroundings and body design. The main ET patterns identified are: (1) high ET variability within a limited angular sector (mainly 90-180 deg away from the threat; this variability is in some cases based on multiple peaks of ETs), (2) ETs that allow sensory tracking of the threat and (3) ETs towards a shelter. These characteristic features are observed across various taxa and, therefore, their expression may be mainly related to taxon-independent animal design features and to the environmental context in which prey live - for example whether the immediate surroundings of the prey provide potential refuges.
The Johnston's Organ neurons (JONs) form chemical and electrical synapses onto the giant fiber neuron (GF), as part of the neuronal circuit that mediates the GF escape response in Drosophila ...melanogaster. The purpose of this study was to identify which of the 8 Drosophila innexins (invertebrate gap junction proteins) mediates the electrical connection at this synapse. The GF is known to express Shaking B (ShakB), specifically the ShakB(N+16) isoform only, at its output synapses in the thorax. The shakB2 mutation disrupts these GF outputs and also abolishes JON-GF synaptic transmission. However, the identity of the innexin that forms the presynaptic hemichannels in the JONs remains unknown. We used electrophysiology, immunocytochemistry and dye injection, along with presynaptically-driven RNA interference, to investigate this question. The amplitude of the compound action potential recorded in response to sound from the base of the antenna (sound-evoked potential, or SEP) was reduced by RNAi of the innexins Ogre, Inx3, Inx6 and, to a lesser extent Inx2, suggesting that they could be required in JONs for proper development, excitability, or synchronization of action potentials. The strength of the JON-GF connection itself was reduced to background levels only by RNAi of shakB, not of the other seven innexins. ShakB knockdown prevented Neurobiotin coupling between GF and JONs and removed the plaques of ShakB protein immunoreactivity that are present at the region of contact. Specific shakB RNAi lines that are predicted to target the ShakB(L) or ShakB(N) isoforms alone did not reduce the synaptic strength, implying that it is ShakB(N+16) that is required in the presynaptic neurons. Overexpression of ShakB(N+16) in JONs caused the formation of ectopic dye coupling, whereas ShakB(N) prevented it altogether, supporting this conclusion and also suggesting that gap junction proteins may have an instructive role in synaptic target choice.

Escape trajectories (ETs; measured as the angle relative to the direction of the threat) have been studied in many taxa using a variety of methodologies and definitions. Here, we provide a review of ...methodological issues followed by a survey of ET studies across animal taxa, including insects, crustaceans, molluscs, lizards, fish, amphibians, birds and mammals. Variability in ETs is examined in terms of ecological significance and morpho-physiological constraints. The survey shows that certain escape strategies (single ETs and highly variable ETs within a limited angular sector) are found in most taxa reviewed here, suggesting that at least some of these ET distributions are the result of convergent evolution. High variability in ETs is found to be associated with multiple preferred trajectories in species from all taxa, and is suggested to provide unpredictability in the escape response. Random ETs are relatively rare and may be related to constraints in the manoeuvrability of the prey. Similarly, reports of the effect of refuges in the immediate environment are relatively uncommon, and mainly confined to lizards and mammals. This may be related to the fact that work on ETs carried out in laboratory settings has rarely provided shelters. Although there are a relatively large number of examples in the literature that suggest trends in the distribution of ETs, our understanding of animal escape strategies would benefit from a standardization of the analytical approach in the study of ETs, using circular statistics and related tests, in addition to the generation of large data sets.
We undertook a systematic literature review as a background to the European League Against Rheumatism (EULAR) recommendations for conducting clinical trials in anti-neutrophil cytoplasm antibody ...associated vasculitis (AAV), and to assess the quality of evidence for outcome measures in AAV.
Using a systematic Medline search, we categorised the identified studies according to diagnoses. Factors affecting remission, relapse, renal function and overall survival were identified.
A total of 44 papers were reviewed from 502 identified by our search criteria. There was considerable inconsistency in definitions of end points. Remission rates varied from 30% to 93% in Wegener granulomatosis (WG), 75% to 89% in microscopic polyangiitis (MPA) and 81% to 91% in Churg-Strauss syndrome (CSS). The 5-year survival for WG, MPA and CSS was 74-91%, 45-76% and 60-97%. Relapse (variably defined) was common in the first 2 years but the frequency varied: 18% to 60% in WG, 8% in MPA, and 35% in CSS. The rate of renal survival in WG varied from 23% at 15 months to 23% at 120 months.

Antipredator behavior is vital for most animals and calls for accurate timing and swift motion. Whereas fast reaction times 1 and predictable, context-dependent escape-initiation distances 2 are ...common features of most escape systems, previous work has highlighted the need for unpredictability in escape directions, in order to prevent predators from learning a repeated, fixed pattern 3–5. Ultimate unpredictability would result from random escape trajectories. Although this strategy would deny any predictive power to the predator, it would also result in some escape trajectories toward the threat. Previous work has shown that escape trajectories are in fact generally directed away from the threat, although with a high variability 5–8. However, the rules governing this variability are largely unknown. Here, we demonstrate that individual cockroaches (Periplaneta americana, a much-studied model prey species 9–14) keep each escape unpredictable by running along one of a set of preferred trajectories at fixed angles from the direction of the threatening stimulus. These results provide a new paradigm for understanding the behavioral strategies for escape responses, underscoring the need to revisit the neural mechanisms controlling escape directions in the cockroach and similar animal models, and the evolutionary forces driving unpredictable, or “protean”3, antipredator behavior.
Gravitational-wave astronomy is established with direct observation of gravitational wave from merging binary black holes and binary neutron stars during the first and second observing run of LIGO ...and Virgo detectors. The gravitational-wave transient searches mainly separate into two families: modeled and modeled-independent searches. The modeled searches are based on matched filtering techniques, and model-independent searches are based on the extraction of excess power from time-frequency representations. We have proposed a hybrid method, called wavegraph that mixes the two approaches. It uses astrophysical information at the extraction stage of model-independent search using a mathematical graph. In this work, we assess the performance of wavegraph clustering in real LIGO and Virgo noise (the sixth science run and the first observing run) and using the coherent WaveBurst transient search as a backbone. Further, we propose a new signal consistency test for this algorithm. This test uses the amplitude profile information to distinguish between the gravitational-wave transients from the noisy glitches. This test is able to remove a large fraction of loud glitches, which thus results in additional overall sensitivity in the context of searches for binary black-hole mergers in the low-mass range.
Health planners use forecasts of key metrics associated with influenza-like illness (ILI); near-term weekly incidence, week of season onset, week of peak, and intensity of peak. Here, we describe our ...participation in a weekly prospective ILI forecasting challenge for the United States for the 2016-17 season and subsequent evaluation of our performance. We implemented a metapopulation model framework with 32 model variants. Variants differed from each other in their assumptions about: the force-of-infection (FOI); use of uninformative priors; the use of discounted historical data for not-yet-observed time points; and the treatment of regions as either independent or coupled. Individual model variants were chosen subjectively as the basis for our weekly forecasts; however, a subset of coupled models were only available part way through the season. Most frequently, during the 2016-17 season, we chose; FOI variants with both school vacations and humidity terms; uninformative priors; the inclusion of discounted historical data for not-yet-observed time points; and coupled regions (when available). Our near-term weekly forecasts substantially over-estimated incidence early in the season when coupled models were not available. However, our forecast accuracy improved in absolute terms and relative to other teams once coupled solutions were available. In retrospective analysis, we found that the 2016-17 season was not typical: on average, coupled models performed better when fit without historically augmented data. Also, we tested a simple ensemble model for the 2016-17 season and found that it underperformed our subjective choice for all forecast targets. In this study, we were able to improve accuracy during a prospective forecasting exercise by coupling dynamics between regions. Although reduction of forecast subjectivity should be a long-term goal, some degree of human intervention is likely to improve forecast accuracy in the medium-term in parallel with the systematic consideration of more sophisticated ensemble approaches.

Increasing water abstraction is severely degrading the world’s wetlands, which is frequently reflected in the condition of flood dependent organisms. Understanding minimum requirements under which ...thresholds are crossed from desired to undesired states can improve managing for resilience and avoid catastrophic ecological consequences. The Macquarie Marshes, a Ramsar-listed wetland in eastern Australia is located in a catchment with a long history of water resource development. It is also a catchment in which there has been a strong focus on wetland recovery using environmental flows. We investigated changes in the condition of 212 river red gum trees (Eucalyptus camaldulensis), a long-lived flood dependent species, at 17 sites, over 18years (1993–2011). Four variables were measured for each tree: crown density, crown size, dead branches and epicormic growth. More than half (56.13%, 119) of the healthy trees first measured in 1993, showed no signs of life by 2011. Using historic inundation mapping, we identified that condition declined with reduced flooding. The probability of inundation in the previous five years had the strongest explanatory power, with strong increasing threshold responses of persistence and recovery associated with probabilities of flooding exceeding 5years in the previous 10years. We extended our modelling to the river red gum forests of the Macquarie Marshes floodplain, showing that 37% of the area had a predicted survival probability lower than P=0.6 while similar probabilities were present in only 12% of the area within the Macquarie Marshes Nature Reserve. The condition of river red gum forests, using the relationships identified, provide a useful measure of ecosystem health, particularly if extended to understanding of reproduction, germination and recruitment in relation to different scenarios for environmental flows. Embedding such a monitoring strategy in an adaptive management framework provides considerable promise for ongoing learning and improvement of management, not just for river red gums but also other indicators. There is a possibility that the current poor ecological condition of the Macquarie Marshes can be redressed with increased environmental flows restoring some of the river red gum forests that are a key characteristic of the wetland.
