Lack of requisite genetic variation in Turkish okra has necessitated the use of different types of markers for estimating the genetic diversity and identifying the source of variation. Transposable ...elements, present abundantly in plant genomes, generate genomic diversity through their replication and are thus an excellent source of molecular markers. We hypothesized that inter-primer binding site (iPBS)-retrotransposons could be the source of variation because of their genome plasticity nature. In the present study, genetic diversity of 66 okra landraces was analyzed using iPBS-retrotransposon markers. iPBS-retrotransposons detected 88 bands with 40.2% polymorphism and an average of 6.8 bands per primer. Gene diversity and Shannon's information index ranged from 0.01 to 0.13 and 0.02 to 0.21 for iPBS-retrotransposons and from 0.06 to 0.46 and 0.14 to 0.65 for simple sequence repeat (SSR) markers, respectively. Polymorphism information content value for retrotransposons varied between 0.12 and 0.99, while that for SSR was from 0.52 to 0.81. Neighbor joining analysis based on retrotransposons and SSRs divided all the accessions into four clusters; however, SSR markers were more efficient in clustering the landraces based on their origin. Using the STRUCTURE software for determining population structure, and two populations (at the number of hypothetical subpopulations, K = 2) were identified among the landraces. Low genetic diversity in Turkish okra highlights the need for the introduction of plants from countries with greater genetic diversity for these crops. This study also demonstrates the utility and role of iPBS-retrotransposons, a dominant and ubiquitous part of eukaryotic genomes, for diversity studies in okra.

We studied the genetic variability due to mutation induced by γ-rays (10, 15, and 20 Kr) on various traits of twelve rice genotypes. Mutated and non-mutated seeds were sown in the field between July ...2013 and 2014 using a split plot design. Yield and yield-related trait data was recorded, which showed significant (P < 0.05) genotypic and irradiation effects. Gamma radiation exerted non-significant effects on the panicle length of all plants, indicating the uniformity of performance of this character at different radiation levels. The plant height, grains per main panicle, panicle length, 1000-grain weight, grain weight per main panicle, and fertility percentage was minimum in Basmati-198, Basmati-Pak, Shaheen-2000, Super Basmati, Basmati-385, and Super Basmati, respectively, when exposed to radiation at 20 Kr. However, Basmati-370 attained maximum flag leaf area at this level of radiation. Broad sense heritability ranged from 72.0 to 97.7%, indicating the possibility of selection during earlier generations. Phenotypic correlation was positive and significant between grain weight per main panicle with panicle length and flag leaf area. Number of grains per main panicle and genotypic correlation were positive among grain weight per main panicle, panicle length, fertility percentage, 1000-grain weight, and plant height, indicating the effectiveness of these traits in the selection for yield. Phylogenetic analyses revealed a pair-wise similarity ranging from 0.51-0.76 before mutation and 0.39-0.89 after mutation. This study proved that γ-rays at 20 Kr would be most effective in creating genetic variability in the existing germplasm.
A field experiment was conducted to investigate the effect of soil application of zinc fertilizer on yield and yield components of wheat (Triticum aestivum L. cv. Inqlab 91) grown on calcareous soil ...in Pakistan. The levels of zinc sulphate were 0 (control), 5, 10, 15, 20, 25 and 30 kg ha â2 and the zinc sulphate was combine-drilled at the time of sowing. Zinc sulphate increased the Leaf Area Index, the total number of fertile tillers m â2 , number of spikelets spike â2 , spike length, grain spike â2 , thousand grain weight, grain yield, straw yield and biological yield and decreased harvest index. Most of the response trends were curvilinear although the decrease in harvest index was linear. All applications of zinc sulphate gave economic increases in margins over costs but the application of 5 kg ha â2 gave the highest marginal rate of return. It is recommended that under such calcareous soil conditions growers can expect good returns from the application of 5 kg zinc sulphate ha â2 at the time of sowing but if the grain price were to increase or the price of zinc sulphate were reduced economic responses could be expected from higher levels of zinc sulphate.
Protection against SARS-CoV-2 is mediated by humoral and T cell responses. Pakistan faced relatively low morbidity and mortality from COVID-19 through the pandemic. To examine the role of prior ...immunity in the population, we studied IgG antibody response levels, virus neutralizing activity and T cell reactivity to Spike protein in a healthy control group (HG) as compared with COVID-19 cases and individuals from the pre-pandemic period (PP). HG and COVID-19 participants were recruited between October 2020 and May 2021. Pre-pandemic sera was collected before 2018. IgG antibodies against Spike and its Receptor Binding Domain (RBD) were determined by ELISA. Virus neutralization activity was determined using a PCR-based micro-neutralization assay. T cell - IFN-gamma activation was assessed by ELISpot. Overall, the magnitude of anti-Spike IgG antibody levels as well as seropositivity was greatest in COVID-19 cases (90%) as compared with HG (39.8%) and PP (12.2%). During the study period, Pakistan experienced three COVID-19 waves. We observed that IgG seropositivity to Spike in HG increased from 10.3 to 83.5% during the study, whilst seropositivity to RBD increased from 7.5 to 33.3%. IgG antibodies to Spike and RBD were correlated positively in all three study groups. Virus neutralizing activity was identified in sera of COVID-19, HG and PP. Spike reactive T cells were present in COVID-19, HG and PP groups. Individuals with reactive T cells included those with and without IgG antibodies to Spike. Antibody and T cell responses to Spike protein in individuals from the pre-pandemic period suggest prior immunity against SARS-CoV-2, most likely from cross-reactive responses. The rising seroprevalence observed in healthy individuals through the pandemic without known COVID-19 may be due to the activation of adaptive immunity from cross-reactive memory B and T cells. This may explain the more favourable COVID-19 outcomes observed in this population.

Vernalization and photoperiod response genes play a significant role in the geographical adaptation, agronomic performance and yield potential of crops. Therefore, understanding the distribution ...pattern and allelic diversity for vernalization and photoperiod genes are important in any wheat breeding program. In this study, we screened 63 bread wheat cultivars and 7 bread wheat landraces from Turkey for photoperiod (Ppd-D1) and vernalization genes (Vrn-A1, Vrn-B1, Vrn-D1 and Vrn-B3) using diagnostic molecular markers. The photoperiod insensitive dominant allele, Ppd-D1a, was present in 60% of wheat cultivars and 42% of landraces, whereas, all other genotypes carried the photoperiod sensitive allele Ppd-D1b as recessive allele. Twenty-four cultivars and two landraces contained recessive alleles for all four VRN loci, whereas 39 wheat cultivars and 6 landraces contained one or more dominant VRN alleles. The highest percentage of Turkish wheat cultivars contained the dominant Vrn-B1 allele followed by Vrn-D1 and Vrn-A1. Information for vernalization and photoperiod alleles in Turkish germplasm will facilitate the planning and implementation of molecular markers in wheat breeding programs. This information will be helpful to develop elite wheat cultivars carrying suitable vernalization and photoperiod alleles with higher grain yield potential and better quality suitable for different production environments through marker assisted selection.
This study examines the correlation between oil price fluctuation and absolute business development in Pakistan. Our study focusses on three economic sectors, agriculture and livestock, manufacturing ...and electricity production and transportation from 1980 to 2018 using the autoregressive distributed lag, with linear regression to evaluate the (time series or panel) data (please elaborate the frequency of data as well either it is daily, weekly, monthly, quarterly or yearly data). Our findings reveal negative impact of oil price on the economic development overall, and manufacturing, electricity production and livestock sectors individually; while, there is positive relationship observed with communication and transport sectors. There is need for policymaker’s attention on highly oil-dependent sectors to run their operations. Empirical findings suggest a 30% shortage of oil supply responsible for the highest fluctuated structure of oil pricing, which suddenly increases the projected welfare loss through a 40% reduction in gross domestic product. This study suggests that the country should maintain a minimum 100-day strategic petroleum reserves to hedge any adverse effect of oil price fluctuation on economic and social welfare losses.
The advent of multidrug resistance among pathogenic bacteria is imperiling the worth of antibiotics, which have previously transformed medical sciences. The crisis of antimicrobial resistance has ...been ascribed to the misuse of these agents and due to unavailability of newer drugs attributable to exigent regulatory requirements and reduced financial inducements. Comprehensive efforts are needed to minimize the pace of resistance by studying emergent microorganisms, resistance mechanisms, and antimicrobial agents. Multidisciplinary approaches are required across health care settings as well as environment and agriculture sectors. Progressive alternate approaches including probiotics, antibodies, and vaccines have shown promising results in trials that suggest the role of these alternatives as preventive or adjunct therapies in future.
