An electromagnetic calorimeter made of undoped CsI crystals is used in the J-PARC KOTO experiment to search for new physics beyond the standard model with the KL→π0νν̄ decay. The calorimeter is ...designed to operate in vacuum of 0.1 Pa and in a high-rate environment where the counting rate due to KL decays is O(100) kHz. A special method to calibrate the calorimeter during the data taking without using a tracking system for charged particles is reported. The energy, position, and timing resolutions of the calorimeter were evaluated in several beam tests, and the resolutions satisfy the required performance. The energy resolution with the total energy E is 0.66⊕1.81∕EGeV% in the inner region of the calorimeter.
Botrytis cinerea field isolates collected in Japan were screened for resistance to Qo inhibitor fungicides (QoIs). Of the 198 isolates screened, six grew well on a medium containing azoxystrobin, a ...QoI, when salicylhydroxamic acid, an alternative oxidase inhibitor, was present. The resistance mutation in the cytochrome b gene (cytb) was characterized. All QoI-resistant isolates had the same mutation (GGT to G CT) in cytb that led to the substitution of glycine by alanine at position 143 of cytochrome b, which is known to confer QoI resistance in plant pathogens. To detect this mutation, a hybridization probe assay based on real-time PCR amplification and melting curve analysis was developed. Using DNA samples prepared from aubergines coinfected with QoI-resistant and QoI-sensitive B. cinerea isolates, two similar peak profiles with their corresponding melting temperatures were obtained. This result suggests that QoI-resistant and QoI-sensitive isolates may compete equally in terms of pathogenicity, and the assay may be used to assess the population ratio of mutant and wild-type isolates. However, the hybridization probe did not anneal to PCR products derived from the DNA samples of some QoI-sensitive isolates. Structural analysis of cytb revealed that B. cinerea field isolates could be classified into two groups: one with three introns and the other with an additional intron (Bcbi-143/144 intron) inserted between the 143rd and 144th codons. All 88 isolates possessing the Bcbi-143/144 intron were azoxystrobin-sensitive, suggesting that the QoI-resistant mutation at codon 143 in cytb prevents self-splicing of the Bcbi-143/144 intron, as proposed in some other plant pathogens.
The pumpellyite–actinolite facies proposed by Hashimoto is defined by the common occurrence of the pumpellyite–actinolite assemblage in basic schists. It can help characterize the paragenesis of ...basic and intermediate bulk compositions, which are common constituents of various low‐grade metamorphic areas. The dataset of mutually consistent thermodynamic properties of minerals gives a positive slope for the boundary between the pumpellyite–actinolite and prehnite–pumpellyite facies in P–T space. In the Sanbagawa belt in Japan, the mineral parageneses of hematite‐bearing and ‐free basic schists, as well as pelitic schists have been well documented. The higher temperature limit of this facies is defined by the disappearance of the pumpellyite+epidote+actinolite+chlorite assemblage in hematite‐free basic schists with XFe3+ of epidote around 0.20–0.25 and the appearance of epidote+actinolite+chlorite assemblage with XEpFe3+≤0.20. In hematite‐bearing basic schists, there is a continuous change of paragenesis to higher grade, epidote–glaucophane or epidote–blueschist facies. In pelitic schists, the albite+lawsonite+chlorite assemblage does occur but only rarely, and its assemblage cannot be used to determine the regional thermal structure. The lower temperature equivalence of the pumpellyite–actinolite assemblage is not observed in the field. The Mikabu Greenstone complex and the northern margin of the Chichibu complex, which are located to the south of the Sanbagawa belt, are characterized by clinopyroxene+chlorite or lawsonite+actinolite assemblages, which are lower temperature assemblages than the pumpellyite+actinolite assemblage. These three metamorphic complexes belong to the same subduction‐metamorphic complex. The pumpellyite–actinolite facies or subfacies can be useful to help reveal the field thermal structure of metamorphic complexes
We searched for the CP-violating rare decay of the neutral kaon, KL→π0νν¯, in data from the first 100 hours of physics running in 2013 of the J-PARC KOTO experiment. One candidate event was observed ...while 0.34±0.16 background events were expected. We set an upper limit of 5.1×10−8 for the branching fraction at the 90% confidence level (C.L.). An upper limit of 3.7×10−8 at the 90% C.L. for the KL→π0X0 decay was also set for the first time, where X0 is an invisible particle with a mass of 135 MeV/c2.
MUM1/IRF4 is a myeloma-associated oncogene transcriptionally activated as a result of t(6;14)(p25,q32) chromosomal translocation and by virtue of its juxtaposition to the immunoglobulin heavy chain ...gene (IgH) locus. When this oncogene becomes non-functional, no activated B/T lymphocytes and Ig secreting plasma cells are observed, suggesting that MUM1/IRF4 is crucial for lymphoid development. Its expression was analyzed in both reactive lymphoid and lymphoma tissues by means of an immunohistochemical technique using specific goat antiserum against MUM1/IRF4. This analysis detected a 50 kDa MUM1 product whose localization was restricted to the nuclei of the lymphocytes. The MUM1+ cells in reactive lymph nodes were found to consist of plasma cells and a small fraction (approximately 7.9%) of B cells harboring CD20+CD38+, which were located in the light zone of the germinal center. MUM1 expression in peripheral blood B/T lymphocytes was upregulated by mitogenic stimuli, suggesting that MUM1 positivity represents the activated state of the B/T cells. In B cell non-Hodgkin's lymphoma (NHL), MUM1 expression was observed in 73.2% (30/41) of diffuse large B cell lymphoma (DLBCL), 20% (1/5) of marginal zone lymphoma (MZL) and 43% (3/7) of small lymphocytic lymphoma (SLL) cases, whereas it was not seen in any cases of mantle cell lymphoma (MCL) or follicle center lymphoma (FCL). Also, MUM1 was stained at high intensity in various types of T cell lymphomas including adult T cell leukemia/lymphoma (ATL/L) and anaplastic large cell lymphoma (ALCL) and in the majority of Hodgkin's diseases. Our results suggest that a major proportion of lymphomas comprise either physiologically or aberrantly activated neoplastic lymphocytes expressing the MUM1 protein.
Granulitized coesite-bearing eclogite from Weihai, northeastern part of the Shandong peninsula, eastern China was studied in detail to reveal the modification of mineral chemistry during ...decompression metamorphism. Considerable modification of chemical composition is recorded in clinopyroxene that occurs both as inclusions in garnet and as a matrix mineral. Careful examination of chemical variation with the change in microstructure made it possible to estimate the equilibrium composition of minerals at the coesite eclogite and garnet granulite stages. We were able to define three reference points on the
P–
T path, namely, coesite eclogite (3 GPa, 660±40°C), granulite (1 GPa, 700±30°C) and amphibolite (0.9 GPa, 600±20°C). The path thus obtained is similar to those obtained by previous workers and supports nearly isothermal decompression of coesite eclogite.
Objective: Rheumatoid arthritis (RA) is a chronic inflammatory disease and synovial cells, antigen presenting cells, lymphocytes, and their cytokines might be associated with the disease. ...Transforming growth factor β1 (TGFβ1) has been reported to have important roles in unresolved inflammation, immune suppression, fibrosing processes, and angiogenesis. TGFβ1 is highly expressed in joints in RA and is considered to be a regulator of anti-inflammation in RA. Polymorphisms of TGFβ1 have been reported to be associated with the production of TGFβ1 protein, and to increase the risk of acquiring several diseases. It was speculated that these polymorphisms might also be involved in RA, and therefore the TGFβ1 codon 10 T869C polymorphism in a series of patients and controls was investigated. Method: A total of 155 patients with RA and 110 healthy subjects were studied. DNA was extracted from peripheral leucocytes and TGFβ1 codon 10 T869C polymorphism was determined by polymerase chain reaction restriction fragment polymorphism. Results: A significantly higher proportion of patients with RA with the T allele (CT type or TT type) was found compared with the CC type (p=0.039). Conclusion: The T allele, previously reported to be linked with production of TGFβ1, may be associated with an increased risk of RA.

at Tokai) experiment aims to observe the CP-violating rare decay
$K_L \rightarrow \pi ^0 \nu \bar {\nu }$
by using a long-lived neutral-kaon beam produced by the 30 GeV proton beam ...at the Japan Proton Accelerator Research Complex. The
$K_L$
flux is an essential parameter for the measurement of the branching fraction. Three
$K_L$
neutral decay modes,
$K_L \rightarrow 3\pi ^0$
,
$K_L \rightarrow 2\pi ^0$
, and
$K_L \rightarrow 2\gamma $
, were used to measure the
$K_L$
flux in the beam line in the 2013 KOTO engineering run. A Monte Carlo simulation was used to estimate the detector acceptance for these decays. Agreement was found between the simulation model and the experimental data, and the remaining systematic uncertainty was estimated at the 1.4% level. The
$K_L$
flux was measured as

$K_L$
per

protons on a 66-mm-long Au target.
A cyclic AMP (cAMP)-dependent protein kinase pathway has been shown to reg-ulate growth, morphogenesis and virulence in filamentous fungi. However, the precise mechanisms of regulation through the ...pathway remain poorly understood. In Neurospora crassa, the cr-1 adenylate cyclase mutant exhibits colonial growth with short aerial hyphae bearing conidia, and the mcb mutant, a mutant of the regulatory subunit of cAMP-dependent protein kinase (PKA), shows the loss of growth polarity at the restrictive temperature. In the present study, we isolated mutants of the catalytic subunit of the PKA gene pkac-1 through the process of repeat-induced point mutation (RIP). PKA activity of the mutants obtained through RIP was undetectable. The genome sequence predicts two distinct catalytic subunit genes of PKA, named pkac-1 (NCU06240.1, AAF75276) and pkac-2 (NCU00682.1), as is the case in most filamentous fungi. The results sug-gest that PKAC-1 works as the major PKA in N. crassa. The phenotype of the pkac-1 mutants included colonial growth, short aerial hyphae, premature conidiation on solid medium, inappropriate conidiation in submerged culture, and increased thermotolerance. This phenotype of pkac-1 mutants resembled to that of cr-1 mutants, except that the addition of cAMP did not rescue the abnormal morphology of pkac-1 mutants. The loss of growth polarity at the restrictive temperature in the mcb mutant was suppressed by pkac-1 mutation. These results suggest that the signal transduction pathway mediated by PKAC-1 plays an important role in regulation of aerial hyphae formation, conidiation, and hyphal growth with polarity.
