Rail power conditioner (RPC) has the ability to improve the power quality in AC railway power grids. This power conditioner can increase the loading capacity of traction substations, balance the ...active power between the feeder load sections, and compensate for reactive power and current harmonics. At present, there is increasing use of multilevel converter topologies, which provide scalability and robust performance under different conditions. In this framework, modular multilevel converter (MMC) is emerging as a prominent solution for medium-voltage applications. Serving that purpose, this paper focuses on the implementation, testing, and validation of a reduced-scale laboratory prototype of a proposed RPC based on an MMC. The developed laboratory prototype, designed to be compact, reliable, and adaptable to multipurpose applications, is presented, highlighting the main control and power circuit boards of the MMC. In addition, MMC parameter design of the filter inductor and submodule capacitor is also explained. Experimental analysis and validation of a reduced-scale prototype RPC based on MMC topology, are provided to verify the power quality improvement in electrified railway power grids. Thus, two experimental case studies are presented: (1) when both of the load sections are unequally loaded; (2) when only one load section is loaded. Experimental results confirm the RPC based on MMC is effective in reducing the harmonic contents, solving the problem of three-phase current imbalance and compensating reactive power.
AbstractThe red piranha, Pygocentrus nattereri, is an important resource for artisanal and commercial fisheries. The present study determines the genetic differentiation among P. nattereri ...populations from the northeastern Brazilian state of Maranhão. The DNA was isolated using a standard phenol-chloroform protocol and the Control Region was amplified by PCR. The PCR products were sequenced using the didesoxyterminal method. A sequence of 1039 bps was obtained from the Control Region of 60 specimens, which presented 33 polymorphic sites, 41 haplotypes, һ =0.978 and π =0.009. The neutrality tests (D and Fs) were significant (P < 0.05) for most of the populations analyzed. The AMOVA indicated that most of the molecular variation (72%) arises between groups. The fixation index was highly significant (FST = 0.707, P < 0.00001). The phylogenetic analyses indicated that the specimens represented a monophyletic group. Genetic distances between populations varied from 0.8% to 1.9%, and were <0.5% within populations. The degree of genetic differentiation found among the stocks of P. nattereriindicates the need for the development of independent management plans for the different river basins in order to preserve the genetic variability of their populations.
Mutations in SF3B1 have been identified across several cancer types. Here, we demonstrate that depletion of SF3B1 specifically compromises homologous recombination (HR) and is epistatic with loss of BRCA1. More importantly, the most prevalent cancer-associated mutation in SF3B1, K700E, also affects HR efficiency and as a consequence, increases the cellular sensitivity to ionizing radiation and a variety of chemotherapeutic agents, including PARP inhibitors. In addition, the SF3B1 K700E mutation induced unscheduled R-loop formation, replication fork stalling, increased fork degradation, and defective replication fork restart. Taken together, these data suggest that tumor-associated mutations in SF3B1 induce a BRCA-like cellular phenotype that confers synthetic lethality to DNA-damaging agents and PARP inhibitors, which can be exploited therapeutically.

Purpose To investigate the in vitro effect of pH, osmolarity, solvent, and light interaction on currently used and novel dyes to minimize dye-related retinal toxicity. Design Laboratory ...investigation. Methods Retinal pigment epithelium (RPE) human cells (ARPE-19) were exposed for 10 minutes to different pH solutions (4, 5, 6, 7, 7.5, 8, and 9) and glucose solutions (2.5%, 5.0%, 10%, 20%, 40%, and 50%) with osmolarity from 142 to 2530 mOsm, with and without 0.5 mg/mL trypan blue. R28 cells were also incubated with glucose (150, 310, and 1000 mOsm) and mannitol used as an osmotic control agent in both experiments. Dye-light interaction was assessed by incubating ARPE-19 for 10 minutes with trypan blue, brilliant blue, bromophenol blue, fast green, light green, or indigo carmine (0.05 mg/mL diluted in balanced saline solution) in the presence of high-brightness xenon and mercury vapor light sources. Results Solutions with nonphysiologic pH, below 7 and above 7.5, proved to be remarkably toxic to RPE cells with or without trypan blue. Also, all glucose solutions were deleterious to RPE ( P < .001) even in iso-osmolar range. No harmful effect was found with mannitol solutions. Among the dyes tested, only light green and fast green were toxic to ARPE-19 ( P < .001). Light exposure did not increase RPE toxicity either with xenon light or mercury vapor lamp. Conclusions Solutions containing glucose as a dye solvent or nonphysiologic pH should be used with care in surgical situations where the RPE is exposed. Light exposure under present assay conditions did not increase the RPE toxicity.
Abstract High-voltage electric injuries have many manifestations, and an important complication is the damage of the central/peripheral nervous system. The purpose of this work was to assess the ...upper limb dysfunction in patients injured by high-voltage current. The evaluation consisted of analysis of patients’ records, cutaneous-sensibility threshold, handgrip and pinch strength and a specific questionnaire about upper limb dysfunctions (DASH) in 18 subjects. All subjects were men; the average age at the time of the injury was 38 years. Of these, 72% changed job/retired after the injury. The current entrance was the hand in 94% and grounding in the lower limb in 78%. The average burned surface area (BSA) was 8.6%. The handgrip strength of the injured limb was reduced ( p < 0.05) and so also that of the three pinch types. The relationship between the handgrip strength and the DASH was statistically significant ( p < 0.001) as well as the relationship between the three pinch types ( p ≤ 0.02) to the injured limb. The ability to perceive cutaneous touch/pressure was decreased in the burnt hand, principally in the median nerve area. These data indicate a reduction of the hand muscular strength and sensibility, reducing the function of the upper limb in patients who received high-voltage electrical shock.

Background
Excessive sodium consumption (>2g/day) is one of the risk factors for the development of cardiovascular diseases, which impact the world's public health systems. Rapid ...urbanization, busy life, the high availability of ultra-processed foods and the short time for meals, especially inside the home, are factors that interfere in the population's food choices.
Objective
To estimate sodium intake according to the place of meal consumption and its association with demographic, socioeconomic and salt-related variables.
Methods
Cross-sectional population-based study, carried out with 901 adolescents aged 10 to 19 years, who answered the Food Consumption and Nutritional Status Survey, ISACamp-Nutri 2015. The mean sodium intake was estimated by the 24-hour recall and the association with the independent variables, using linear regression.
Results
Sodium consumption among adolescents was 3.52 g/day (95% CI 3.42-3.63). Those who consumed at least one meal away from home had an excess of 0.19 g/day (P = 0.012) of sodium, which corresponded to a 6.1% increase compared to adolescents who had eaten exclusively at home. Of the total meals eaten by adolescents, 15.9% are taken outside the home, with 6.6% eating at school and 9.3% in different places. There was a significant increase in sodium consumption among adolescents who had breakfast CR = 0.16g (p = 0.032), afternoon snack CR = 0.09g (p = 0.002) and dinner CR = 0.30g (p = 0.029) outside the home.
Conclusions
sodium consumption by adolescents exceeded the WHO recommendation. Eating at least one meal away from home was associated with a higher sodium intake among these individuals. Regulatory measures in the food industry and strategies for food and nutrition education are relevant to reduce sodium consumption in this age group.

In this work, multivariate optimization techniques were used to develop a method based on the ultrasound-assisted extraction for copper, manganese, nickel and zinc determination from rations for ...chicken nutrition using flame atomic absorption spectrometry. The proportions of extracting components (2.0 mol.L-1 nitric, hydrochloric and acetic acid solutions) were optimized using centroid-simplex mixture design. The optimum proportions of this mixture taken as percentage of each component were respectively 20%, 37% and 43%. Variables of method (sample mass, sonication time and final acid concentration) were optimized using Doehlert design. The optimum values found for these variables were respectively 0.24 g, 18s and 3.6 mol.L-1. The developed method allows copper, manganese, nickel and zinc determination with quantification limits of 2.82; 4.52; 10.7; e 9.69 µg.g-1, and precision expressed as relative standard deviation (%RSD, 25 µg.g-1, N = 5) of 5.30; 2.13; 0.88; and 0.83%, respectively. This method was applied in the analytes determination from chicken rations collected from specialized commerce in Jequié city (Bahia State/Brazil). Application of paired t-test at the obtained results, in a confidence level of 95%, does not show significant difference between the proposed method and the microwave-assisted digestion.
Identification of human papillomavirus (HPV) association in head and neck squamous cell carcinoma (HNSCC) is important to identify patients with favorable disease course. However, molecular HPV ...detection is not universally available. p16 has been proposed as a surrogate marker for HPV infection in HNSCC but, use on its own may result in wrong assignment of some cases to the group of HPV-associated tumors. We have therefore studied 424 HNSCC cases with known p16 and HPV DNA polymerase chain reaction (PCR) status for expression of retinoblastoma protein (pRb) and CyclinD1 by immunohistochemistry using 6-tiered scales (0 to 5) and a combined score (0 to 10). Sixty-one of 424 cases showed overexpression of p16. Of these, 52 cases were HPV DNA-PCR-positive. HPV association strongly correlated with low expression scores for pRb and CyclinD1 individually (scores ≤2) or combined (score sum ≤4), whereas HPV-negative carcinomas showed widely distributed expression scores. High expression scores for pRb or for pRb/CyclinD1 were observed exclusively in HPV DNA-PCR-negative cases. Three of 9 p16-positive/HPV DNA-PCR-negative cases showed high expression of pRb and displayed a high combined pRb/CyclinD1 score. We conclude that HPV-positive HNSCC are characterized by p16 overexpression and low scores for pRb, CyclinD1, and a low combined pRb/CyclinD1 score. High pRb or combined pRb/CyclinD1 scores are strong indicators for HPV-negativity and may justify excluding these cases from further molecular HPV testing. Furthermore p16-positive/HPV DNA-PCR-negative cases show heterogeneous expression of pRb and CyclinD1, including high pRb or high combined pRb/CyclinD1 scores suggesting that at least some of these cases are truly HPV negative.
Annual suicide figures are critical in identifying trends and guiding research, yet challenges arising from significant lags in reporting can delay and complicate real-time interventions. In this ...paper, we utilized Google Trends search volumes for behavioral forecasting of national suicide rates in Ireland between 2004 and 2015. Official suicide rates are recorded by the Central Statistics Office in Ireland. While similar investigations using Google trends data have been carried out in other jurisdictions (e.g., United Kingdom, United Stated of America), such research had not yet been completed in Ireland. We compiled a collection of suicide- and depression-related search terms suggested by Google Trends and manually sourced from the literature. Monthly search rate terms at different lags were compared with suicide occurrences to determine the degree of correlation. Following two approaches based on vector autoregression and neural network autoregression, we achieved mean absolute error values between 4.14 and 9.61 when incorporating search query data, with the highest performance for the neural network approach. The application of this process to United Kingdom suicide and search query data showed similar results, supporting the benefit of Google Trends, neural network approach, and the applied search terms to forecast suicide risk increase. Overall, the combination of societal data and online behavior provide a good indication of societal risks; building on past research, our improvements led to robust models integrating search query and unemployment data for suicide risk forecasting in Ireland.
