The female rabbit was used to study (i) accumulation of lipophilic chlorinated hydrocarbons in genital tract tissues and (ii) subsequent morphological and functional effects after long-term low-dose ...exposure. Polychlorinated biphenyl (PCB), 1,1-di(p-chlorophenyl)-2,2,2-trichloroethane (DDT) and gamma-hexachlorocyclohexane (gamma-HCH) (dosages: 4, 3 and 0.8 mg per kg body weight, respectively) and a combination of these three components (and dosages) were administered to sexually mature rabbits over a period of 12-15 weeks. The animals were killed shortly before and at various times after ovulation. Accumulation of chlorinated hydrocarbons was high in ovarian, oviductal and uterine tissues, in follicular fluid and clearly detectable in uterine secretions. In follicular fluid, the concentration and patterns of congeners and isomers of PCB and DDT were distinctly different from serum. DDT- and gamma-HCH-treated animals showed a significantly reduced ovulation rate (P < 0.002 and 0.05, respectively). During early pregnancy DDT decreased serum progesterone levels and changed the protein pattern of uterine secretion. Functional effects, however, were much less expressed compared with the highly significant accumulation of the persistent organochlorines in the genital tract.
We present new geochemical and isotopic data for rock samples from two island arc volcanoes, Erromango and Vulcan Seamount, and from a 500 m thick stratigraphic profile of lava flows exposed on the ...SW flank of Vate Trough back‐arc rift of the New Hebrides Island Arc (NHIA). The basalts from the SW rift flank of Vate Trough have ages of ~0.5 Ma but are geochemically similar to those erupting along the active back‐arc rift. The weak subduction component in the back‐arc basalts implies formation by decompression melting during early rifting and rifting initiation by tectonic processes rather than by lithosphere weakening by arc magma. Melting beneath Vate Trough is probably caused by chemically heterogeneous and hot mantle that flows in from the North Fiji Basin in the east. The melting zone beneath Vate Trough back‐arc is separate from that of the arc front, but a weak slab component suggests fluid transport from the slab. Immobile incompatible element ratios in South NHIA lavas overlap with those of the Vate Trough depleted back‐arc basalts, suggesting that enriched mantle components are depleted by back‐arc melting during mantle flow. The slab component varies from hydrous melts of subducted sediments in the Central NHIA to fluids from altered basalts in the South NHIA. The volcanism of Erromango shows constant compositions for 5 million years, that is, there is no sign for variable depletion of the mantle or for a change of slab components due to collision of the D'Entrecasteaux Ridge as in lava successions further north.

Results are reported from a joint analysis of Phase I and Phase II data from the Sudbury Neutrino Observatory. The effective electron kinetic energy threshold used is T{sub eff}=3.5 MeV, the lowest ...analysis threshold yet achieved with water Cherenkov detector data. In units of 10{sup 6} cm{sup -2} s{sup -1}, the total flux of active-flavor neutrinos from {sup 8}B decay in the Sun measured using the neutral current (NC) reaction of neutrinos on deuterons, with no constraint on the {sup 8}B neutrino energy spectrum, is found to be PHI{sub NC}=5.140{sub -0.158}{sup +0.160}(stat){sub -0.117}{sup +0.132}(syst). These uncertainties are more than a factor of 2 smaller than previously published results. Also presented are the spectra of recoil electrons from the charged current reaction of neutrinos on deuterons and the elastic scattering of electrons. A fit to the Sudbury Neutrino Observatory data in which the free parameters directly describe the total {sup 8}B neutrino flux and the energy-dependent nu{sub e} survival probability provides a measure of the total {sup 8}B neutrino flux PHI{sub {sup 8}{sub B}}=5.046{sub -0.152}{sup +0.159}(stat){sub -0.123}{sup +0.107}(syst). Combining these new results with results of all other solar experiments and the KamLAND reactor experiment yields best-fit values of the mixing parameters of theta{sub 12}=34.06{sub -0.84}{sup +1.16} degrees and DELTAm{sub 21}{sup 2}=7.59{sub -0.21}{sup +0.20}x10{sup -5} eV{sup 2}. The global value of PHI{sup 8}{sub B} is extracted to a precision of {sub -2.95}{sup +2.38}%. In a three-flavor analysis the best fit value of sin{sup 2}theta{sub 13} is 2.00{sub -1.63}{sup +2.09}x10{sup -2}. This implies an upper bound of sin{sup 2}theta{sub 13}<0.057 (95% C.L.).
A search has been made for neutrinos from the hep reaction in the Sun and from the diffuse supernova neutrino background (DSNB) using data collected during the first operational phase of the Sudbury ...Neutrino Observatory, with an exposure of 0.65 ktons yr. For the hep neutrino search, two events are observed in the effective electron energy range of 14.3 MeV < T sub(eff) < 20 MeV, where 3.1 background events are expected. After accounting for neutrino oscillations, an upper limit of 2.3 x 10 super(4) cm super(-2) s super(-1) at the 90% confidence level is inferred on the integral total flux of hep neutrinos. For DSNB neutrinos, no events are observed in the effective electron energy range of 21 MeV < T sub(eff) < 35 MeV, and, consequently, an upper limit on the u sub(e) component of the DSNB flux in the neutrino energy range of 22.9 MeV < E sub(u)< 36.9 MeV of 70 cm super(-2) s super(-1) is inferred at the 90% confidence level. This is an improvement by a factor of 6.5 on the previous best upper limit on the hep neutrino flux and by 2 orders of magnitude on the previous upper limit on the u sub(e) component of the DSNB flux.
Conservation practitioners have long recognized ecological connectivity as a global priority for preserving biodiversity and ecosystem function. In the early years of conservation science, ecologists ...extended principles of island biogeography to assess connectivity based on source patch proximity and other metrics derived from binary maps of habitat. From 2006 to 2008, the late Brad McRae introduced circuit theory as an alternative approach to model gene flow and the dispersal or movement routes of organisms. He posited concepts and metrics from electrical circuit theory as a robust way to quantify movement across multiple possible paths in a landscape, not just a single least‐cost path or corridor. Circuit theory offers many theoretical, conceptual, and practical linkages to conservation science. We reviewed 459 recent studies citing circuit theory or the open‐source software Circuitscape. We focused on applications of circuit theory to the science and practice of connectivity conservation, including topics in landscape and population genetics, movement and dispersal paths of organisms, anthropogenic barriers to connectivity, fire behavior, water flow, and ecosystem services. Circuit theory is likely to have an effect on conservation science and practitioners through improved insights into landscape dynamics, animal movement, and habitat‐use studies and through the development of new software tools for data analysis and visualization. The influence of circuit theory on conservation comes from the theoretical basis and elegance of the approach and the powerful collaborations and active user community that have emerged. Circuit theory provides a springboard for ecological understanding and will remain an important conservation tool for researchers and practitioners around the globe.

Quienes practican la conservación han reconocido durante mucho tiempo que la conectividad ecológica es una prioridad mundial para la preservación de la biodiversidad y el funcionamiento del ecosistema. Durante los primeros años de la ciencia de la conservación los ecólogos difundieron los principios de la biografía de islas para evaluar la conectividad con base en la proximidad entre el origen y el fragmento, así como otras medidas derivadas de los mapas binarios de los hábitats. Entre 2006 y 2008 el fallecido Brad McRae introdujo la teoría de circuitos como una estrategia alternativa para modelar el flujo génico y la dispersión o las rutas de movimiento de los organismos. McRae propuso conceptos y medidas de la teoría de circuitos eléctricos como una manera robusta para cuantificar el movimiento a lo largo de múltiples caminos posibles en un paisaje, no solamente a lo largo de un camino o corredor de menor costo. Military helicopter training over the Lincoln National Forest (LNF) in southcentral New Mexico has been severely limited to protect nesting Mexican spotted owls (Strix occidentalis lucida). To ...evaluate nesting and nonnesting spotted owl responses to helicopter noise, we measured flush frequency, flush distance, alert behavior, response duration, prey delivery rates, female trips from the nest, and nest attentiveness during manipulated and nonmanipulated periods, 1995-96. Chain saws were included in our manipulations to increase experimental options and to facilitate comparative results. We analyzed stimulus events by measuring noise levels as unweighted one-third-octave band levels, applying frequency weighting to the resultant spectra, and calculating the sound exposure level for total sound energy (SEL) and the 0.5-sec equivalent maximum energy level$(\text{LEQ}_{\text{max}\ 0.5\text{-sec}})$for helicopters, and the 10-sec equivalent average energy level$(\text{LEQ}_{\text{avg.}\ 10\text{-sec}})$for chain saws. An owl-weighting (dBO) curve was estimated to emphasize the middle frequency range where strigiform owls have the highest hearing sensitivity. Manipulated and nonmanipulated nest sites did not differ in reproductive success (P = 0.59) or the number of young fledged (P = 0.12). As stimulus distance decreased, spotted owl flush frequency increased, regardless of stimulus type or season. We recorded no spotted owl flushes when noise stimuli were >105 m away. Spotted owls returned to predisturbance behavior within 10-15 min after a stimulus event. All adult flushes during the nesting season occurred after juveniles had left the nest. Spotted owl flush rates in response to helicopters did not differ between nonnesting (13.3%) and nesting seasons (13.6%; P = 0.34). Spotted owls did not flush when the SEL noise level for helicopters was ≤102 dBO (92 dBA) and the LEQ level for chain saws was ≤59 dBO (46 dBA). Chain saws were more disturbing to spotted owls than helicopter flights at comparable distances. Our data indicate a 105-m buffer zone for helicopter overflights on the LNF would minimize spotted owl flush response and any potential effects on nesting activity.
Current interest in Foxp3+ T-regulatory (Treg) cells as therapeutic targets in transplantation is largely focused on their harvesting pre-transplant, expansion and infusion post-transplantation. An ...alternate strategy of pharmacologic modulation of Treg function using histone/protein deacetylase inhibitors (HDACi) may allow more titratable and longer-term dosing. However, the effects of broadly acting HDACi vary, such that HDAC isoform-selective targeting is likely required. We report data from mice with constitutive or conditional deletion of HDAC11 within Foxp3+ Treg cells, and their use, along with small molecule HDAC11 inhibitors, in allograft models. Global HDAC11 deletion had no effect on health or development, and compared to WT controls, Foxp3+ Tregs lacking HDAC11 showed increased suppressive function, and increased expression of Foxp3 and TGF-β. Likewise, compared to WT recipients, conditional deletion of HDAC11 within Tregs led to long-term survival of fully MHC-mismatched cardiac allografts, and prevented development of transplant arteriosclerosis in an MHC class II-mismatched allograft model. The translational significance of HDAC11 targeting was shown by the ability of an HDAC11i to promote long-term allograft allografts in fully MHC-disparate strains. These data are powerful stimuli for the further development and testing of HDAC11-selective pharmacologic inhibitors, and may ultimately provide new therapies for transplantation and autoimmune diseases.
Invariant natural killer T (iNKT) cells comprise a unique subset of lymphocytes that are primed for activation and possess innate NK-like functional features. Currently, iNKT cell-based ...immunotherapies remain in early clinical stages, and little is known about the ability of these cells to survive and retain effector functions within the solid tumor microenvironment (TME) long-term. In conventional T cells (T
), cellular metabolism is linked to effector functions and their ability to adapt to the nutrient-poor TME. In contrast, the bioenergetic requirements of iNKT cells - particularly those of human iNKT cells - at baseline and upon stimulation are not well understood; neither is how these requirements affect effector functions such as production of cytokines and cytolytic proteins. We find that unlike T
, human iNKT cells are not dependent upon glucose or glutamine for these effector functions upon stimulation with anti-CD3 and anti-CD28. Additionally, transcriptional profiling revealed that stimulated human iNKT cells are less glycolytic than T
and display higher expression of fatty acid oxidation (FAO) and adenosine monophosphate-activated protein kinase (AMPK) pathway genes. Furthermore, stimulated iNKT cells displayed higher mitochondrial mass and membrane potential relative to T

Landscape connectivity is crucial for many ecological processes, including dispersal, gene flow, demographic rescue, and movement in response to climate change. As a result, governmental and ...non-governmental organizations are focusing efforts to map and conserve areas that facilitate movement to maintain population connectivity and promote climate adaptation. In contrast, little focus has been placed on identifying barriers-landscape features which impede movement between ecologically important areas-where restoration could most improve connectivity. Yet knowing where barriers most strongly reduce connectivity can complement traditional analyses aimed at mapping best movement routes. We introduce a novel method to detect important barriers and provide example applications. Our method uses GIS neighborhood analyses in conjunction with effective distance analyses to detect barriers that, if removed, would significantly improve connectivity. Applicable in least-cost, circuit-theoretic, and simulation modeling frameworks, the method detects both complete (impermeable) barriers and those that impede but do not completely block movement. Barrier mapping complements corridor mapping by broadening the range of connectivity conservation alternatives available to practitioners. The method can help practitioners move beyond maintaining currently important areas to restoring and enhancing connectivity through active barrier removal. It can inform decisions on trade-offs between restoration and protection; for example, purchasing an intact corridor may be substantially more costly than restoring a barrier that blocks an alternative corridor. And it extends the concept of centrality to barriers, highlighting areas that most diminish connectivity across broad networks. Identifying which modeled barriers have the greatest impact can also help prioritize error checking of land cover data and collection of field data to improve connectivity maps. Barrier detection provides a different way to view the landscape, broadening thinking about connectivity and fragmentation while increasing conservation options.

Advanced endometrial transformation often occurs in IVF and embryo transfer therapy after ovarian stimulation with gonadotrophins. One reason is probably the early rise in peripheral progesterone ...concentration after ovulation induction. Consequently, we studied in a rabbit model, whether the post-ovulatory application of the progesterone receptor antagonist, onapristone, could prevent such an advancement of endometrial transformation after stimulation with different gonadotrophin preparations. The inhibitory effect of onapristone on the endometrium is dependent upon the strength of ovarian stimulation. In unstimulated animals or animals treated with recombinant LH (nine corpora lutea/animal in both groups), secretory differentiation and proliferation of the endometrium was strongly inhibited by onapristone. After weak ovarian stimulation with a 3:1 mixture of FSH and LH (22 corpora lutea/animal), secretory differentiation was strongly inhibited, while proliferation was enhanced. After strong stimulation with either a 1:1 mixture of FSH and LH, or human menopausal gonadotrophin (HMG; >40 corpora lutea/animal), only limited inhibitory effects of onapristone on secretory transformation or proliferation could be detected. In conclusion, these graded effects of onapristone after stimulation with gonadotrophins, resemble the basic observations from which a therapeutic strategy emerges, to modulate the advanced endometrial transformation which occurs in many IVF patients after ovarian stimulation.
