Electrons can be transferred from microorganisms to multivalent metal ions that are associated with minerals and vice versa. As the microbial cell envelope is neither physically permeable to minerals ...nor electrically conductive, microorganisms have evolved strategies to exchange electrons with extracellular minerals. In this Review, we discuss the molecular mechanisms that underlie the ability of microorganisms to exchange electrons, such as c-type cytochromes and microbial nanowires, with extracellular minerals and with microorganisms of the same or different species. Microorganisms that have extracellular electron transfer capability can be used for biotechnological applications, including bioremediation, biomining and the production of biofuels and nanomaterials.

Sediment microbial fuel cells (SMFCs) are expected to be used as a renewable power source for remote environmental monitoring; therefore, evaluation of their long-term power performance is critical for their usability. In this paper, we present novel data needed to understand the long-term performance of SMFCs. We used 3-D Microemulsion (3DMe)™ doped anodes, which slowly release lactate and its fermented products. During our tests, anode-limited SMFCs with and without 3DMe-doped anodes were operated for more than 18months with a load simulating a sensor operation. We found that doping an anode with an electron donor reduced startup time and increased maximum power (55±2μW compared to 46±2μW) in the control systems. We found that the long-term steady power performance is approximately 33% of the maximum power (∼18μW). Finally, our small-sized SMFCs generated higher power densities than those in the literature (28mW/m2 versus 4mW/m2). Using electron donor doped anodes can be practical when a short startup time and initial high power are needed. However, if long-term power is critical, the addition of an electron donor does not provide a practical advantage. In addition, in long-term operation enrichment of the anode surface with electrochemically active bacteria does not provide any advantage.
This review examines the electrochemical techniques used to study extracellular electron transfer in the electrochemically active biofilms that are used in microbial fuel cells and other ...bioelectrochemical systems. Electrochemically active biofilms are defined as biofilms that exchange electrons with conductive surfaces: electrodes. Following the electrochemical conventions, and recognizing that electrodes can be considered reactants in these bioelectrochemical processes, biofilms that deliver electrons to the biofilm electrode are called anodic, ie electrode-reducing, biofilms, while biofilms that accept electrons from the biofilm electrode are called cathodic, ie electrode-oxidizing, biofilms. How to grow these electrochemically active biofilms in bioelectrochemical systems is discussed and also the critical choices made in the experimental setup that affect the experimental results. The reactor configurations used in bioelectrochemical systems research are also described and the authors demonstrate how to use selected voltammetric techniques to study extracellular electron transfer in bioelectrochemical systems. Finally, some critical concerns with the proposed electron transfer mechanisms in bioelectrochemical systems are addressed together with the prospects of bioelectrochemical systems as energy-converting and energy-harvesting devices.
Implant-related infection is one of the key concerns in total joint hip arthroplasties. To reduce bacterial adhesion, we used silver (Ag)/silver oxide (Ag2O) doping in plasma sprayed hydroxyapatite ...(HA) coating on titanium substrate. HA powder was doped with 2.0, 4.0, and 6.0 wt % Ag, heat-treated at 800 °C and used for plasma spray coating using a 30 kW plasma spray system, equipped with supersonic nozzle. Application of supersonic plasma nozzle significantly reduced phase decomposition and amorphous phase formation in the HA coatings as evident by X-ray diffraction (XRD) study and Fourier transformed infrared spectroscopic (FTIR) analysis. Adhesive bond strength of more than 15 MPa ensured the mechanical integrity of the coatings. Resistance against bacterial adhesion of the coatings was determined by challenging them against Pseudomonas aeruginosa (PAO1). Live/dead staining of the adherent bacteria on the coating surfaces indicated a significant reduction in bacterial adhesion due to the presence of Ag. In vitro cell-material interactions and alkaline phosphatase (ALP) protein expressions were evaluated by culturing human fetal osteoblast cells (hFOB). Our results suggest that the plasma-sprayed HA coatings doped with an optimum amount of Ag can have excellent antimicrobial property without altering mechanical property of the Ag-doped HA coatings.

To understand electron transport in electrochemically active biofilms, it is necessary to elucidate the heterogeneous electron transport across the biofilm/electrode interface and in the interior of G. sulfurreducens biofilms bridging gaps of varying widths. The conductivity of Geobacter sulfurreducens biofilm bridging nonconductive gaps with widths of 5µm, 10µm, 20µm and 50µm is investigated. Results of electrochemical gating measurement show that biofilm conductivity peaks at the potential of −0.35V vs. Ag/AgCl. The biofilm conductivity increases with gap width (10.4±0.2µScm−1 in 5µm gap, 13.3±0.2µScm−1 in 10µm gap, 16.7±1.4µScm−1 in 20µm gap and 41.8±2.02µScm−1 in 50µm gap). These results revealed that electron transfer in G. sulfurreducens biofilm is a redox-driven. In addition, higher biofilm conductivities and lower charge transfer resistances are observed in all gaps under a turnover condition than in those under a non-turnover condition. Our results offer insights into the spatial heterogeneity of biofilm structure and extracellular electron transfer in electrochemically active biofilms.

Here we describe a form of metabolism linking anoxygenic ...photosynthesis to anaerobic respiration that we call 'syntrophic anaerobic photosynthesis'. We show that photoautotrophy in the green sulfur bacterium Prosthecochloris aestaurii can be driven by either electrons from a solid electrode or acetate oxidation via direct interspecies electron transfer from a heterotrophic partner bacterium, Geobacter sulfurreducens. Photosynthetic growth of P. aestuarii using reductant provided by either an electrode or syntrophy is robust and light-dependent. In contrast, P. aestuarii does not grow in co-culture with a G. sulfurreducens mutant lacking a trans-outer membrane porin-cytochrome protein complex required for direct intercellular electron transfer. Syntrophic anaerobic photosynthesis is therefore a carbon cycling process that could take place in anoxic environments. This process could be exploited for biotechnological applications, such as waste treatment and bioenergy production, using engineered phototrophic microbial communities.
We have operated a microbial fuel cell in which glucose was oxidized by Klebsiella pneumoniae in the anodic compartment, and biomineralized manganese oxides, deposited by Leptothrix discophora, were ...electrochemically reduced in the cathodic compartment. In the anodic compartment, to facilitate the electron transfer from glucose to the graphite electrode, we added a redox mediator, 2-hydroxy-1,4-naphthoquinone. We did not add any redox mediator to the cathodic compartment because the biomineralized manganese oxides were deposited on the surface of a graphite electrode and were reduced directly by electrons from the electrode. We have demonstrated that biomineralized manganese oxides are superior to oxygen when used as cathodic reactants in microbial fuel cells. The current density delivered by using biomineralized manganese oxides as the cathodic reactant was almost 2 orders of magnitude higher than that delivered using oxygen. Several fuel cells were operated for 500 h, reaching anodic potentials of −441.5 ± 31 mVSCE and cathodic potentials of +384.5 ± 64 mVSCE. When the electrodes were connected by a 50 Ω resistor, the fuel cell delivered the peak power density of 126.7 ± 31.5 mW/m2.
The goal of this work was to develop a microbiosensor to measure acetate concentration profiles inside biofilms in situ. The working principle of the microbiosensor was based on the correlation ...between the acetate concentration and the current generated during acetate oxidation by Geobacter sulfurreducens. The microbiosensor consisted of a 30-µm carbon microelectrode with an open tip as a working electrode, with G. sulfurreducens biofilm on the tip and a pseudo Ag/AgCl reference electrode, all enclosed in a glass outer case with a 30-µm tip diameter. The microbiosensor showed a linear response in the 0–1.6mM acetate concentration range with a 79±8µM limit of detection (S/N=2). We quantified the stirring effect and found it negligible. However, the interfering effect of alternative electron donors (lactate, formate, pyruvate, or hydrogen) was found to be significant. The usefulness of the acetate microbiosensor was demonstrated by measuring acetate concentration depth profiles within a G. sulfurreducens biofilm. The acetate concentration remained at bulk values throughout the biofilm when no current was passed, but it decreased from the bulk values to below the detection limit within 200µm when current was allowed to pass. The zero acetate concentration at the bottom of the biofilm showed that the biofilm was acetate-limited.

Biofilm formation causes prolonged wound infections due to the dense biofilm structure, differential gene regulation to combat stress, and production of extracellular polymeric substances. ...Acinetobacter baumannii, Staphylococcus aureus, and Pseudomonas aeruginosa are three difficult-to-treat biofilm-forming bacteria frequently found in wound infections. This work describes a novel wound dressing in the form of an electrochemical scaffold (e-scaffold) that generates controlled, low concentrations of hypochlorous acid (HOCl) suitable for killing biofilm communities without substantially damaging host tissue. Production of HOCl near the e-scaffold surface was verified by measuring its concentration using needle-type microelectrodes. E-scaffolds producing 17, 10 and 7 mM HOCl completely eradicated S. aureus, A. baumannii, and P. aeruginosa biofilms after 3 hours, 2 hours, and 1 hour, respectively. Cytotoxicity and histopathological assessment showed no discernible harm to host tissues when e-scaffolds were applied to explant biofilms. The described strategy may provide a novel antibiotic-free strategy for treating persistent biofilm-associated infections, such as wound infections.
