To investigate the clinicopathological features and molecular characteristics of β-catenin-deficient colorectal cancer.
The clinical, pathological and molecular features of 11 colorectal cancers with ...β-catenin protein loss diagnosed at the 960th Hospital of People's Liberation Army of China, from January 2012 to November 2022 were analyzed.
Among the 11 patients, 3 were males and 8 were females. Their age ranged from 43 to 74 years, with the median age of 59 years. Six were in the left colon and 5 were in the right colon. One of the 11 cases had lymph node metastasis, 10 cases were well and moderately differentiated adenocarcinoma, and 1 was mucinous adenocarcinoma. Eight cases were of TNM stage T4, 2 of T1 stage and 1 of Tis stage. β-catenin protein was not detected using immunohistochemistry. Sanger sequencing revealed the presence of fragment-deletion mutation in exon 3 of CTNNB1 gene, resulting in loss of β-catenin protein expression.
β-catenin deficiency is present in a small number of colorectal cancers
The receptor activator of nuclear factor κB ligand (RANKL)/receptor activator of nuclear factor κB (RANK)/osteoprotegerin (OPG) system plays a key role in rheumatoid arthritis (RA)-associated bone ...erosion. The upregulation of the RANKL/OPG ratio promotes bone erosion. The objective of this study is to explore the effects of iguratimod, a small-molecule disease-modifying antirheumatic drug (DMARD), alone or in combination with methotrexate (MTX), on RANKL and OPG expression in RA. We performed an enzyme-linked immunosorbent assay (ELISA) to investigate the modulatory effects of iguratimod, MTX, or their combination on serum RANKL and OPG levels of patients with RA before and after treatment for 12 and 24 weeks. Furthermore, fibroblast-like synoviocytes (FLS) from patients with RA were interleukin (IL)-1β-stimulated and then treated with different concentrations of iguratimod, MTX, or both, and RANKL and OPG expressions were investigated by using ELISA, quantitative real-time polymerase chain reaction (qPCR) and western blot analysis. We found that RANKL levels and the RANKL/OPG ratio significantly decreased in both serum and IL-1β-induced RA FLS after treatment. Moreover, combination therapy with iguratimod and MTX showed an even stronger inhibition than each drug alone did. Our results suggest that iguratimod and MTX, especially in combination, efficaciously protected against bone erosion by suppressing the production of RANKL.
In the automatic sorting process, overlapping translucent and flexible workpieces on the conveyor belt, blurring the imaging edge features of translucent and flexible workpieces is a challenge to ...locate the upper and lower workpieces spatially, we propose a method for locating translucent and flexible workpieces spatially under the overlapping environment in conjunction with the most common automatic sorting of translucent and flexible workpieces such as infusion tube drip buckets. Firstly, we propose a rectangular surface light source based on 650 nm band and monocular CCD for imaging translucent workpieces such as infusion tube drip buckets and optimize the imaging parameters. Secondly, we study a feature matching recognition algorithm for flexible workpieces that are prone to deformation, construct a mapping relationship between the position of overlapping layers and imaging quality of translucent and flexible workpieces such as infusion tube drip buckets based on clarity and information entropy, and establish The mapping relationship between the position of the overlapping layers and the imaging quality of translucent and flexible workpieces such as infusion tube drip buckets is constructed based on clarity and information entropy, and a local spatial coordinate conversion model is established. Finally, the spatial positioning coordinates of overlapping and non-overlapping translucent and flexible workpieces in the local coordinate system are identified, and the results show that the imaging method and theory can be effectively applied to the identification of overlapping and spatial positioning coordinates in the automatic sorting of translucent workpieces such as infusion tube drip buckets.
To investigate damaging mechanisms of chilling and salt stress to peanut (Arachis hypogaea L.) leaves, LuHua 14 was used in the present work upon exposure to chilling temperature (4°C) accompanied by ...high irradiance (1,200 μmol m⁻² s⁻¹) (CH), salt stress accompanied by high irradiance (1,200 μmol m⁻² s⁻¹) (SH), and high-irradiance stress (1,200 μmol m⁻² s⁻¹) at room temperature (25°C) (NH), respectively. Additionally, plants under low irradiance (100 μmol m⁻² s⁻¹) at room temperature (25°C) were used as control plants (CK). Relative to CK and NH treatments, both the maximal photochemical efficiency of PSII (Fv/Fm) and the absorbance at 820 nm decreased greatly in peanut leaves under CH and SH stress, which indicated that severe photoinhibition occurred in peanut leaves under such conditions. Initial fluorescence (Fo), 1 − qP and nonphotochemical quenching (NPQ) in peanut leaves significantly increased under CH- and SH stress. Additionally, the activity of superoxide dismutase (SOD), one of the key enzymes of water-water cycle, decreased greatly, the accumulation of malondialdehyde (MDA) and membrane permeability increased. These results suggested that damages to peanut photosystems might be related to the accumulation of reactive oxygen species (ROS) induced by excess energy, and the water-water cycle could not dissipate energy efficiently under the stress of CH and SH, which caused the accumulation of ROS greatly. CH and SH had similar damaging effects on peanut photosystems, except that CH has more severe effects. All the results showed that CH- and SH stress has similar damaging site and mechanisms in peanut leaves.
Tanshinone IIA (TSIIA) is a major active triterpenoid isolated from Salvia miltiorrhiza. The purposes of this study were to investigate various preclinical factors that determined the ...pharmacokinetics of TSIIA. After oral dosing at 6.7, 20, and 60 mg kg−1, TSIIA was detected mainly as glucuronidated conjugate (TSIIAG) with only small amounts of the unchanged in the plasma. TSIIA was predominantly excreted into the bile and faeces as TSIIAG, and urine to a minor extent. The Cmax and AUC0−t of TSIIAG after i.p. administration were significantly lower than those after intragastric administration. The plasma concentration-time profiles of TSIIA following oral dosing of TSIIA showed multiple peaks. The Cmax and AUC0−t of TSIIA and its glucuronides in rats with intact bile duct were significantly lower than those of rats with bile duct cannulation. Studies from the linked-rat model and intraduodenal injection of bile containing TSIIA and its metabolites indicate that TSIIA glucuronides underwent hydrolysis and the aglycone was reabsorbed from the gut and excreted into the bile as conjugates. TSIIA had a wide tissue distribution, with a very high accumulation in the lung, but very limited penetration into the brain and testes. TSIIA was metabolized by rat CYP2C, 3A and 2D, as ticlopidine, ketoconazole and quinidine all inhibited TSIIA metabolism in rat liver microsomes. Taken collectively, these findings indicate that multiple factors play important roles in determining the pharmacokinetics of TSIIA.

Magnetoelectric (ME) Ni–PZT–Ni cylindrical layered composites have been prepared by electroless deposition. The ME effect was measured by applying both constant and alternating magnetic fields parallel and perpendicular to the cylinder axis direction. The ME voltage coefficient increases with cylinder diameter. Both experimental and calculated data indicate that the resonance frequency of Ni–PZT–Ni cylindrical layered composites decreases with increasing cylinder diameter. We can apply this characteristic of resonance frequency to design a kind of ME composites with the controllable resonance ME coupling.
The objective of this study was to investigate the expression and relationship of LL-37, plasmacytoid dendritic cells (pDCs) and interferon-alpha (IFN-α) in skin in systemic lupus erythematosus ...(SLE), and their role in SLE pathogenesis. Skin biopsies were taken from nine SLE patients and six healthy volunteers. Expression of LL-37, pDCs and IFN-α in skin specimens and consecutive sections of skin was detected with an immunohistochemical technique (IH); the expression of LL-37 and pDCs in the samples was detected with in situ hybridization (ISH). The expression levels of LL-37, pDCs and IFN-α were significantly higher in SLE skin than in that of healthy controls (p < 0.001) with either the IH or the ISH technique, and the location of positive expression in consecutive sections was similar. Correlation analysis showed that the expression levels of LL-37, pDCs and IFN-α correlated positively with each other. In conclusion, the expression of LL-37, pDCs and IFN-α was increased in the skin of patients with active SLE. It is necessary to study further the role of LL-37 in the pathogenesis of SLE, and the exact relationship among LL-37, pDCs and IFN-α.

Myosins and kinesins are molecular motors that hydrolyse ATP to track along
actin filaments and microtubules, respectively. Although the kinesin family
includes motors that move towards either the ...plus or minus ends of microtubules, all characterized myosin motors move towards the barbed (+) end
of actin filaments. Crystal structures of myosin II (refs 3,4,5,6) have shown that small movements within the myosin motor
core are transmitted through the 'converter domain' to a 'lever
arm' consisting of a light-chain-binding helix and associated light
chains. The lever arm further amplifies the motions of the
converter domain into large directed movements. Here
we report that myosin VI, an unconventional myosin,
moves towards the pointed (-) end of actin. We visualized the myosin VI construct
bound to actin using cryo-electron microscopy and image analysis, and found
that an ADP-mediated conformational change in the domain distal to the motor,
a structure likely to be the effective lever arm, is in the opposite direction
to that observed for other myosins. Thus, it appears that myosin VI achieves
reverse-direction movement by rotating its lever arm in the opposite direction
to conventional myosin lever arm movement.

BACKGROUNDThe female gluteal morphology is an important secondary sexual characteristic and helps accentuate and define the female body shape. Despite the increasing prevalence of gluteal ...augmentations in our country, little data exist regarding the morphology of the gluteal region in young Chinese females. This study analyzed a convenience sample of Chinese females and their gluteal region.
METHODSData, measurements, and photos of the following parameters were takenage, weight, height, and body mass index (BMI). Morphology was classified into four typesA shape, V shape, round shape, and square shape. Aesthetic characteristics of each buttock were recorded. Height, weight, BMI, and gluteal projection ratio differences in different shape groups were analyzed.
RESULTSThe study sample consisted of 103 women, with ages from 23 to 27 years. The V-shaped buttock was not found in this sample. The A shape, square shape, and round shape occupied 55.3%, 38.8%, and 5.8% accordingly. Short infragluteal fold, supragluteal fossettes, lateral depression, and V-shaped crease appeared in 42.7%, 14.6%, 11.7%, and 2.9% of samples, respectively. Of the 103 samples, 39 (37.9%) fulfilled the aesthetic gluteal projection(ratio ≥ 2). The average BMI was statistically significant between each gluteal shape (P = 0.009). Height, weight, and gluteal projection ratio showed no significant difference with the buttock shapes.
CONCLUSIONSWe are the first to describe gluteal morphology in young Chinese woman using gluteal anthropometrics. The findings of the study may provide a guideline for plastic surgeons who perform gluteal augmentation in Asian patients.
