Skeletal disorders can seriously threaten the health and the performance of poultry, such as tibial dyschondroplasia (TD) and osteoporosis (OP). Oligomeric proanthocyanidins (OPC) are naturally ...occurring polyphenolic flavonoid compounds that can be used as potential substances to improve the bone health and the growth performance of poultry. Eighty 7-day-old green-eggshell yellow feather layer chickens were randomly divided into 4 groups: basal diet and basal diet supplementation with 25, 50, and 100 mg/kg OPC. The results have indicated that the growth performance and bone parameters of chickens were significantly improved supplementation with OPC in vivo, including the bone volume (BV), the bone mineral density (BMD) and the activities of antioxidative enzymes, but ratio of osteoprotegerin (OPG)/receptor activator of NF-κB (RANK) ligand (RANKL) was decreased. Furthermore, primary bone marrow mesenchymal stem cells (BMSCs) and bone marrow monocytes/macrophages (BMMs) were successfully isolated from femur and tibia of chickens, and co-cultured to differentiate into osteoclasts in vitro. The osteogenic differentiation derived from BMSCs was promoted treatment with high concentrations of OPC (10, 20, and 40 µmol/L) groups in vitro, but emerging the inhibition of osteoclastogenesis by increasing the ratio of OPG/RANKL. In contrary, the osteogenic differentiation was also promoted treatment with low concentrations of OPC (2.5, 5, and 10 µmol/L) groups, but osteoclastogenesis was enhanced by decreasing the ratio of OPG/RANKL in vitro. In addition, OPG inhibits the differentiation and activity of osteoclasts by increasing the autophagy in vitro. Dietary supplementation of OPC can improve the growth performance of bone and alter the balance of osteoblasts and osteoclasts, thereby improving the bone health of chickens.
Gap junction protein connexin 43 (Cx43) plays a critical role in gap junction communication in rat hepatocytes. However, those located between hepatocytes are easily internalized following exposure ...to poisons. Herein, we investigated the potential of buffalo rat liver 3A (BRL 3A) cells to generate annular gap junctions (AGJs) proficient at alleviating cadmium (Cd) cytotoxic injury through degradation via an endosome-lysosome pathway. Our results showed that Cd-induced damage of liver microtubules promoted Cx43 internalization and increased Cx43 phosphorylation at Ser373 site. Furthermore, we established that Cd induced AGJs generation in BRL 3A cells, and AGJs were subsequently degraded through the endosome-lysosome pathway. Overall, our results suggested that Cx43 internalization and the generation of AGJs were cellular protective mechanisms to alleviate Cd toxicity in rat hepatocytes.
Satellite observations show that carbon monoxide (CO) concentration centers exist in the tropopause region of the Tibetan Plateau, while their sources and formation mechanism still remain uncertain. ...In this paper, the 3-D chemical transport model GEOS-Chem is used to conduct sensitivity analysis in 2016. Combined with the analysis data and satellite data, the contribution of three important emission sources (South Asia, East Asia and Southeast Asia) and two important chemical reaction species (CH4 and nonmethane volatile organic compounds (NMVOCs)) to CO in the upper troposphere and lower stratosphere (UTLS) are studied. The results show that in the Asian monsoon region CO emissions originating from the surface are transported to the upper troposphere via a deep convection process and then enter the Asian Summer Monsoon (ASM) anticyclone. The strong ASM anticyclone isolates the mixing process of air inside and outside the anticyclone, upon entry of carbon monoxide-rich air. In the lower stratosphere, the intensity of the ASM anticyclone declines and the air within the anticyclone flows southwestward with monsoon circulation. We found that in the summer Asian monsoon region, South Asia exhibited the highest carbon monoxide concentration transported to the UTLS. CH4 imposed the greatest influence on the CO concentration in the UTLS region. According to the model simulation results, the CO concentrations in the Asian monsoon region at 100 hPa altitudes were higher than those in other regions at the same latitudes. Regarding effects, 43.18% originated from CH4 chemical reactions, 20.81% originated from NMVOC chemical reactions, and 63.33% originated from surface CO emissions, while sinks yielded a negative contribution of −27.32%. Regarding surface CO emissions, East Asia contributed 13.56%, South Asia contributed 39.27%, and Southeast Asia contributed 7.15%.
Osteoprotegerin (OPG) is known to inhibit differentiation and activation of osteoclasts (OCs) by functioning as a decoy receptor blocking interactions between RANK and RANKL. However, the exact role ...of OPG in the survival/apoptosis of OCs remains unclear. OPG caused increased rates of apoptosis of both OCs and osteoclast precursor cells (OPCs). The expression of Fas and activated caspase-8 was increased by both 20 ng/mL and 40 ng/mL of OPG, but was markedly decreased at 80 ng/mL. Interestingly, we noted that while levels of Fas ligand (FasL) increased with increasing doses of OPG, the soluble form of FasL in the supernatant decreased. The results of a co-immunoprecipitation assay suggested that the decrease of sFasL might be caused by the binding of OPG. This would block the inhibition of the apoptosis of OCs and OPCs. Furthermore, changes in expression levels of Bax/Bcl-2, cleaved-caspase-9, cleaved-caspased-3 and the translocation of cytochrome c, illustrated that OPG induced apoptosis of OCs and OPCs via the classic Fas/FasL apoptosis pathway, and was mediated by mitochondria. Altogether, our results demonstrate that OPG induces OCs and OPCs apoptosis partly by the Fas/FasL signaling pathway.

The Asian summer monsoon (ASM) anticyclone circulation system is recognized to be a significant transport pathway for water vapor and pollutants to enter the stratosphere. The observational evidence, ...however, is largely based on satellite retrievals. We report the first coincident in situ measurements of water vapor and ozone within the ASM anticyclone. The combined water vapor and ozonesondes were launched from Kunming, China in August 2009 and Lhasa, China in August 2010. In total, 11 and 12 sondes were launched in Kunming and Lhasa, respectively. We present the key characteristics of these measurements, and provide a comparison to similar measurements from an equatorial tropical location, during the Tropical Composition, Cloud and Climate Coupling (TC4) campaign in July and August of 2007. Results show that the ASM anticyclone region has higher water vapor and lower ozone concentrations in the upper troposphere and lower stratosphere than the TC4 observations. The results also show that the cold point tropopause in the ASM region has a higher average height and potential temperature. The in situ observations therefore support the satellite‐based conclusion that the ASM is an effective transport pathway for water vapor to enter stratosphere.

Abstract
Ozonesonde launches were routinely performed in Beijing from March 2001 to February 2019 to generate a unique long-term (18 years) vertical ozone profile dataset over mainland China. This ...study elucidates the vertical ozone structure on various temporal scales during this 18 years period by using the entire ozonesonde data product for the first time. Moreover, the long-term variability in the integrated ozone column over the North China Plain (NCP) is also explored by comparing the retrievals from ozonesonde at the Beijing urban site and a Dobson ozone spectrometer at the Xianghe suburban site. Our results indicate that vertical ozone exhibited clear monthly variability characterized by high values of tropospheric ozone during warm seasons and high values of stratospheric ozone during cold seasons. Stratospheric intrusions frequently occurred during spring and effectively transported cold air masses with high ozone from the lower stratosphere downward into the upper troposphere. Evident interannual variability in the lower troposphere and in ozone-rich areas of the stratosphere was revealed by vertical ozone distributions. The integrated total ozone columns retrieved from ozonesonde and Dobson bear close resemblance and exhibit strong sinusoidal monthly variations. In the troposphere and boundary layer, the integrated ozone column presented a significant positive trend during 2001–2012 in Beijing; a sudden decline occurred between 2011 and 2013, which was followed by a slow and insignificant increase after the implementation of the Clean Air Action plan in 2013 on the NCP.
Aerosol extinction profiles at 550 nm were retrieved by applying multi-axis differential optical absorption spectroscopy (MAX-DOAS) and lookup table. Then the tropospheric NO2 and HCHO vertical ...column densities were retrieved using a two-step method from 28 July to 5 August of 2015 in Shanghai. The retrieved results were compared with the satellite products, and then their diurnal variation was observed. A consistency check was performed before the inversion to obtain a correction factor. Based on the sensitivity of geometric angles to oxygen dimer air mass factor (O4 AMF, AMF is the ratio of the slanted column density to the vertical column density), the parameterization scheme of geometric angles in the lookup table is optimized. The results show that the aerosol increased significantly in the afternoon. The diurnal variation of tropospheric NO2 and HCHO vertical column densities (VCDs) are bimodal and unimodal patterns respectively, and their values are higher than those of GOME-2 and OMI satellite products. A process of aerosol reduction and recovery are related to ground particulates and meteorological elements. The chemical sensitivity of local ozone production also has a clear diurnal variation.
Cadmium (Cd) is a highly neurotoxic environmental pollutant. Puerarin (Pur) is a natural antioxidant isolated from Kudzu root that exhibits a powerful neuroprotective effect. Herein, we illustrated ...the mechanism underlying the protective effect of Pur on Cd-induced rat neurocyte injury in an in vivo rat model as well as in vitro using PC12 cells and primary rat cerebral cortical neurons. First, the results showed that Pur alleviated Cd-induced cerebral cortical pathological damage and decreased the viability of neurocytes. Furthermore, Cd activated the nuclear factor erythroid 2-related factor 2 (Nrf2) signaling pathway, which plays a negative role in Cd-induced rat neurocyte injury. In addition, Pur alleviated Cd-induced oxidative stress by enhancing antioxidant defense, reducing reactive oxygen species (ROS) accumulation and lipid peroxidation, and inhibiting activation of the Nrf2 signaling pathway in rat neurocytes. Moreover, Pur inhibited the Cd-induced mitochondrial unfolded protein response (UPRmt) in rat neurocytes. Overall, Pur alleviated Cd-induced rat neurocyte injury by alleviating Nrf2-mediated oxidative stress and inhibiting UPRmt.

Our previous studies have confirmed that cadmium (Cd) exposure causes hepatotoxicity; it also induces autophagy and blocks the autophagy flux. Therefore, we hypothesized that Cd hepatotoxicity could ...be alleviated through nutritional intervention. Taurine (Tau) has various biological functions such as acting as an antioxidant, acting as an anti-inflammatory, and stabilizing cell membranes. In order to explore the protective effect and internal mechanism of Tau on Cd-induced hepatotoxicity, normal rat liver cell line BRL3A cells were treated with Cd alone or in combination with Tau to detect cell injury and autophagy-related indexes in this study. We found that Tau can alleviate Cd-induced cell-proliferation decline and morphological changes in the cell. In addition, Tau activates autophagy and alleviates the blockage of Cd-induced autophagy flux. In this process, lysosome acidification and degradation were enhanced, and autophagosomes were further fused with lysosomes. Then, we found that Tau alleviated autophagic flux block by promoting the transfer of membrane fusion proteins STX17 and SNAP29 to autophagosomes and the translocation of VAMP8 to lysosomes, which in turn attenuated the hepatocyte injury induced by Cd exposure. This will further reveal the hepatotoxicity mechanism of Cd and provide the theoretical basis for the prevention and treatment of Cd poisoning.
