Drylands are predicted to become more arid and saline due to increasing global temperature and drought. Although species from the Caatinga, a Brazilian tropical dry forest, are tolerant to these ...conditions, the capacity for germination to withstand extreme soil temperature and water deficit associated with climate change remains to be quantified. We aimed to evaluate how germination will be affected under future climate change scenarios of limited water and increased temperature. Seeds of three species were germinated at different temperatures and osmotic potentials. Thermal time and hydrotime model parameters were established and thresholds for germination calculated. Germination performance in 2055 was predicted, by combining temperature and osmotic/salt stress thresholds, considering soil temperature and moisture following rainfall events. The most pessimistic climate scenario predicts an increase of 3.9 °C in soil temperature and 30% decrease in rainfall. Under this scenario, soil temperature is never lower than the minimum and seldomly higher than maximum temperature thresholds for germination. As long as the soil moisture (0.139 cm³ cm³) requirements are met, germination can be achieved in 1 day. According to the base water potential and soil characteristics, the minimum weekly rainfall for germination is estimated to be 17.5 mm. Currently, the required minimum rainfall occurs in 14 weeks of the year but will be reduced to 4 weeks by 2055. This may not be sufficient for seedling recruitment of some species in the natural environment. Thus, in future climate scenarios, rainfall rather than temperature will be extremely limiting for seed germination.
Insular land crabs (Gecarcinidae) can transit between terrestrial and aquatic environments and inhabit vacant ecological niches that other species do not occupy in oceanic islands. During the ...reproductive period, these crabs migrate between residential and reproductive areas; this is a critical moment because individuals are more vulnerable to stressful conditions, especially species occupying anthropized islands. Currently, many insular crab species are considered threatened; yet few studies have evaluated the biology of this group, especially the size at which individuals reach sexual maturity. Here, we evaluate the size at the onset of morphological, physiological, and functional maturity for the insular land crab Johngarthia lagostoma in Trindade Island (Brazil) and assess the chronology of the events underlying those processes. Males and females exhibited the same order of occurrence of the different maturity processes, starting by being morphologically, physiologically, and, finally, functionally mature at similar sizes (about 56 mm carapace width). This value corresponds to at least half of the maximum size that J. lagostoma reaches in Trindade Island and is close to the average relative value registered to other Gecarcinidae species. Considering the current decline in the population of insular crabs, such estimates can be used in management programs, mainly for the definition and protection of breeding and recruitment areas. Specifically, our results can be used toward the conservation of J. lagostoma, which is currently classified as endangered in Brazil, especially in the isolated population of Trindade Island.
This paper reports on an NMR metabolomics study of lipophilic extracts of
clams exposed to the hormonal contaminant 17-α-ethinylestradiol (EE2), at 17 °C and 21 °C. The results reveal that exposure ...at 17 °C triggers a weak response at low EE2 concentrations, suggestive of a slight increase in membrane rigidity, followed by lipid metabolic stability at higher EE2 concentrations. On the other hand, at 21 °C, lipid metabolism begins to respond at 125 ng/L EE2, with antioxidant docosahexaenoic acid (DHA) helping to tackle high-oxidative-stress conditions, in tandem with enhanced storage of triglycerides. Exposure to 625 ng/L EE2 (highest concentration) enhances phosphatidylcholine (PtdCho) and polyunsaturated fatty acid (PUFA) levels, their direct intercorrelation suggesting PUFA incorporation in new membrane phospholipids. This should lead to increased membrane fluidity, probably aided by a decrease in cholesterol. PUFA levels, considered a measure of membrane fluidity, were strongly (and positively) correlated to intracellular glycine levels, thus identifying glycine as the main osmolyte entering the cells under high stress. Membrane fluidity also seems to elicit the loss of taurine. This work contributes to the understanding of the mechanisms of response of
clams to EE2 in tandem with warming while unveiling novel potential markers of stress mitigation, namely high levels of PtdCho, PUFAs (or PtdCho/glycerophosphocholine and PtdCho/acetylcholine ratios) and linoleic acid and low PUFA/glycine ratios.

Carbapenem-resistant Klebsiella pneumoniae (CR-KP) are a public health concern, causing infections with a high mortality rate, limited therapeutic options and challenging infection control ...strategies. In Portugal, the CR-KP rate has increased sharply, but the factors associated with this increase are poorly explored. In order to address this question, phylogenetic and resistome analysis were used to compare the draft genomes of 200 CR-KP isolates collected in 2017–2019 from five hospitals in the Lisbon region, Portugal.
Most CR-KP belonged to sequence type (ST) 13 (29%), ST17 (15%), ST348 (13%), ST231 (12%) and ST147 (7%). Carbapenem resistance was conferred mostly by the presence of KPC-3 (74%) or OXA-181 (18%), which were associated with IncF/IncN and IncX plasmids, respectively. Almost all isolates were multi-drug resistant, harbouring resistance determinants to aminoglycosides, beta-lactams, trimethoprim, fosfomycin, quinolones and sulphonamides. In addition, 11% of isolates were resistant to colistin. Colonizing and infecting isolates were highly related, and most colonized patients (89%) reported a previous hospitalization. Moreover, among the 171 events of cross-dissemination identified by core genome multi-locus sequence typing data analysis (fewer than five allelic differences), 41 occurred between different hospitals and 130 occurred within the same hospital.
The results suggest that CR-KP dissemination in the Lisbon region results from acquisition of carbapenemases in mobile genetic elements, influx of CR-KP into the hospitals by colonized ambulatory patients, and transmission of CR-KP within and between hospitals. Prudent use of carbapenems, patient screening at hospital entry, and improvement of infection control are needed to decrease the burden of CR-KP infection in Portugal.
This study investigated the influence of sodium restriction and antihypertensive drugs on atherogenesis utilizing hypertensive (H) low-density lipoprotein-receptor knockout mice treated or not with ...losartan (Los) or hydralazine (Hyd) and fed low-sodium (LS) or normal-sodium (NS) chow. Despite reducing the blood pressure (BP) of H-LS mice, the LS diet caused arterial lipid infiltration due to increased plasma total cholesterol (TC) and triglycerides (TG). Los and Hyd reduced the BP of H-LS mice, and Los effectively prevented arterial injury, likely by reducing plasma TG and nonesterified fatty acids. Aortic lipid infiltration was lower in Los-treated H-LS mice (H-LS+Los) than in normotensive (N)-LS and H-LS mice. Aortic angiotensin II type 1 (AT1) receptor content was greater in H-NS than H-LS mice and in H-LS+Hyd than H-LS+Los mice. Carboxymethyl-lysine (CML) and receptor for advanced glycation end products (RAGE) immunostaining was greater in H-LS than H-NS mice. CML and RAGE levels were lower in LS animals treated with antihypertensive drugs, and Hyd enhanced the AT1 receptor level. Hyd also increased the gene expression of F4/80 but not tumor necrosis factor-α, interleukin (IL)-1β, IL-6, IL-10, intercellular adhesion molecule-1 or cluster of differentiation 66. The novelty of the current study is that in a murine model of simultaneous hypertension and hyperlipidemia, the pleiotropic effect of chronic, severe sodium restriction elicited aortic damage even with reduced BP. These negative effects on the arterial wall were reduced by AT1 receptor antagonism, demonstrating the influence of angiotensin II in atherogenesis induced by a severely LS diet.

This paper describes an untargeted NMR metabolomics study to identify potential intracellular donor-dependent and donor-independent metabolic markers of proliferation and osteogenic differentiation ...of human adipose mesenchymal stem cells (hAMSCs). The hAMSCs of two donors with distinct proliferating/osteogenic characteristics were fully characterized regarding their polar endometabolome during proliferation and osteogenesis. An 18-metabolites signature (including changes in alanine, aspartate, proline, tyrosine, ATP, and ADP, among others) was suggested to be potentially descriptive of cell proliferation, independently of the donor. In addition, a set of 11 metabolites was proposed to compose a possible donor-independent signature of osteogenesis, mostly involving changes in taurine, glutathione, methylguanidine, adenosine, inosine, uridine, and creatine/phosphocreatine, choline/phosphocholine and ethanolamine/phosphocholine ratios. The proposed signatures were validated for a third donor, although they require further validation in a larger donor cohort. We believe that this proof of concept paves the way to exploit metabolic markers to monitor (and potentially predict) cell proliferation and the osteogenic ability of different donors.
A highly exothermic nitrogen generation system (NGS) can be achieved by mixing solutions of sodium nitrite and ammonium chloride, a process used by the oil and gas industry to dissolve paraffin wax ...and gas hydrates. Although its main products are nitrogen gas and a sodium chloride brine, the NGS has a side reaction that produces nitrogen oxides. To optimize this process to ensure the greatest and fastest heat generation with the lowest oxide production, this reaction was checked by infrared spectroscopy and calorimetry. The factors temperature, pH, and initial concentration of nitrite and ammonium were evaluated, and the optimal conditions of the NGS were determined by the constructed models to predict heat and NO x generation. These conditions were a ratio of ammonium/nitrite equal to 1 and a catalyst concentration of 0.07 mol·L–1 (for a case in which the temperature is 5 °C).
Hepatic disorders such as steatosis and alcoholic steatohepatitis are common diseases that affect thousands of people around the globe. This study aims to identify the main phenol compounds using a ...new HPLC-ESI+-MS/MS method, to evaluate some oxidative stress parameters and the hepatoprotective action of green dwarf coconut water, caffeic and ascorbic acids on the liver and serum of rats treated with ethanol. The results showed five polyphenols in the lyophilized coconut water spiked with standards: chlorogenic acid (0.18 µM), caffeic acid (1.1 µM), methyl caffeate (0.03 µM), quercetin (0.08 µM) and ferulic acid (0.02 µM) isomers. In the animals, the activity of the serum γ-glutamyltranspeptidase (γ-GT) was reduced to 1.8 I.U/L in the coconut water group, 3.6 I.U/L in the ascorbic acid group and 2.9 I.U/L in the caffeic acid groups, when compared with the ethanol group (5.1 I.U/L, p<0.05). Still in liver, the DNA analysis demonstrated a decrease of oxidized bases compared to ethanol group of 36.2% and 48.0% for pretreated and post treated coconut water group respectively, 42.5% for the caffeic acid group, and 34.5% for the ascorbic acid group. The ascorbic acid was efficient in inhibiting the thiobarbituric acid reactive substances (TBARS) in the liver by 16.5% in comparison with the ethanol group. These data indicate that the green dwarf coconut water, caffeic and ascorbic acids have antioxidant, hepatoprotective and reduced DNA damage properties, thus decreasing the oxidative stress induced by ethanol metabolism.
