The presence of natural organic matter (NOM) in drinking water treatment presents many challenges. Integrated treatment processes combining oxidation and biofiltration have been demonstrated to be ...very effective at reducing NOM, specifically biodegradable organics. Laboratory bench-scale experiments were carried out to investigate the effect of oxidation by ozonation or UV/H2O2 on NOM. Specifically the rate of biodegradation was studied by performing bench-scale biodegradation experiments using acclimatized biological activated carbon (BAC). For the source water investigated, oxidation did not preferentially react with the biodegradable or non-biodegradable NOM. In addition, the type or dose of oxidation applied did not affect the observed rate of biodegradation. The rate kinetics for biodegradation were constant for all oxidation conditions investigated. Oxidation prior to biofiltration increased the overall removal of organic matter, but did not affect the rate of biodegradation of NOM.
RATIONALE:Familial recurrence studies provide strong evidence for a genetic component to the predisposition to sporadic, nonsyndromic Tetralogy of Fallot (TOF), the most common cyanotic congenital ...heart disease phenotype. Rare genetic variants have been identified as important contributors to the risk of congenital heart disease, but relatively small numbers of TOF cases have been studied to date.
OBJECTIVE:We used whole exome sequencing to assess the prevalence of unique, deleterious variants in the largest cohort of nonsyndromic TOF patients reported to date.
METHODS AND RESULTS:Eight hundred twenty-nine TOF patients underwent whole exome sequencing. The presence of unique, deleterious variants was determined; defined by their absence in the Genome Aggregation Database and a scaled combined annotation-dependent depletion score of ≥20. The clustering of variants in 2 genes, NOTCH1 and FLT4, surpassed thresholds for genome-wide significance (assigned as P<5×10) after correction for multiple comparisons. NOTCH1 was most frequently found to harbor unique, deleterious variants. Thirty-one changes were observed in 37 probands (4.5%; 95% CI, 3.2%–6.1%) and included 7 loss-of-function variants 22 missense variants and 2 in-frame indels. Sanger sequencing of the unaffected parents of 7 cases identified 5 de novo variants. Three NOTCH1 variants (p.G200R, p.C607Y, and p.N1875S) were subjected to functional evaluation, and 2 showed a reduction in Jagged1-induced NOTCH signaling. FLT4 variants were found in 2.4% (95% CI, 1.6%–3.8%) of TOF patients, with 21 patients harboring 22 unique, deleterious variants. The variants identified were distinct to those that cause the congenital lymphoedema syndrome Milroy disease. In addition to NOTCH1, FLT4 and the well-established TOF gene, TBX1, we identified potential association with variants in several other candidates, including RYR1, ZFPM1, CAMTA2, DLX6, and PCM1.
CONCLUSIONS:The NOTCH1 locus is the most frequent site of genetic variants predisposing to nonsyndromic TOF, followed by FLT4. Together, variants in these genes are found in almost 7% of TOF patients.
A novel polysaccharide EtGIPL1a was purified from fruiting bodies of Echinodontium tinctorium, a fungus unique to western North America. EtGIPL1a has an estimated weight average molecular weight of ...275 kDa and is composed of glucose (54.3%), galactose (19.6%), mannose (11.1%), fucose (10.3%), glucuronic acid (4%), and rhamnose (0.6%). It has multiple glycosidic linkages, with 3-Glcp (28.9%), 6-Glcp (18.3%), 3,6-Glcp (13%), 4-GlcpA (9.2%), 6-Galp (3.9%), 2,6-Galp (2.6%), 3-Fucp (2.5%), 6-Manp (2.4%) being the most prominent, and unsubstituted glucose (15.3%), mannose (1.3%) and fucose (0.9%) as major terminal sugars. EtGIPL1a has a backbone containing mostly 3-substituted β-glucopyranose with 4-substituted glucopyranosyluronic acid. EtGIPL1a showed anti-proliferative activity against multiple cancer cell lines, with IC
ranging from 50.6 to 1446 nM. Flow cytometry analyses confirmed that apoptosis induction is one mechanism for its anti-proliferative activity. EtGIPL1a should be further investigated for its potential anti-cancer activity in animal models, and for its possible utility in differentiation cancer therapy.
ABSTRACT
HiPERCAM is a portable, quintuple-beam optical imager that saw first light on the 10.4-m Gran Telescopio Canarias (GTC) in 2018. The instrument uses re-imaging optics and four dichroic ...beamsplitters to record $u_{\rm s}\, g_{\rm s}\, r_{\rm s}\, i_{\rm s}\, z_{\rm s}$ (320–1060 nm) images simultaneously on its five CCD cameras, each of 3.1-arcmin (diagonal) field of view. The detectors in HiPERCAM are frame-transfer devices cooled thermo-electrically to 183 K, thereby allowing both long-exposure, deep imaging of faint targets, as well as high-speed (over 1000 windowed frames per second) imaging of rapidly varying targets. A comparison-star pick-off system in the telescope focal plane increases the effective field of view to 6.7 arcmin for differential photometry. Combining HiPERCAM with the world’s largest optical telescope enables the detection of astronomical sources to gs ∼ 23 in 1 s and gs ∼ 28 in 1 h. In this paper, we describe the scientific motivation behind HiPERCAM, present its design, report on its measured performance, and outline some planned enhancements.
Population connectivity, which is essential for the persistence of benthic marine metapopulations, depends on how life history traits and the environment interact to influence larval production, ...dispersal and survival. Although we have made significant advances in our understanding of the spatial and temporal dynamics of these individual processes, developing an approach that integrates the entire population connectivity process from reproduction, through dispersal, and to the recruitment of individuals has been difficult. We present a population connectivity modelling framework and diagnostic approach for quantifying the impact of i) life histories, ii) demographics, iii) larval dispersal, and iv) the physical seascape, on the structure of connectivity and metapopulation dynamics. We illustrate this approach using the subtidal rocky reef ecosystem of Port Phillip Bay, were we provide a broadly-applicable framework of population connectivity and quantitative methodology for evaluating the relative importance of individual factors in determining local and system outcomes.
The spatial characteristics of marine population connectivity are primarily influenced by larval mortality, the duration of the pelagic larval stage, and the settlement competency characteristics, with significant variability imposed by the geographic setting and the timing of larval release. The relative influence and the direction and strength of the main effects were strongly consistent among 10 connectivity-based metrics.
These important intrinsic factors (mortality, length of the pelagic larval stage, and the extent of the precompetency window) and the spatial and temporal variability represent key research priorities for advancing our understanding of the connectivity process and metapopulation outcomes.
Survival of larval fish is often linked to production of preferred prey such as copepods, both inter- and intra-annually. In turn, copepod production depends not only the quantity of food, but also ...on the nutritional quality, edibility and/or toxicity of their micro-algal food. Hence, larval fish survival can become de-coupled from levels of nutrient input depending on the resulting composition of the plankton. Here we use a plankton dynamics model to study nutrient input, phytoplankton composition and copepod, Paracalanus, production in relation to interannual variation in recruitment of snapper, Chrysophrys auratus, in Port Phillip Bay, Australia. The model was able to simulate the ratio of diatoms to flagellates in the plume of the main river entering Port Phillip Bay. Interannual variability in the copepod, Paracalanus, abundance during the C. auratus spawning period over 5 years was accurately predicted. The seasonal peak in Paracalanus production depended on the timing and magnitude (match-mismatch) of nutrient inputs and how these were reflected in temporal change in the diatom:flagellate ratio. In turn, the model-predicted Paracalanus abundance was strongly related to inter-annaul variability in abundance of snapper, C. auratus, larvae over 7 years. Years of highest larval C. auratus abundance coincided with a matching of the spawning period with the peak in Paracalanus abundance. High freshwater flows and nutrient inputs led to an early seasonal dominance of diatoms, and consequently reduced abundances of copepods over the C. auratus spawning period with correspondingly low abundances of larvae. Conversely years of very low rainfall and nutrient input also led to low phytoplankton and copepod concentrations and larval C. auratus abundances. Highest abundances of larval C. auratus occurred in years of low to intermediate rainfall and nutrient inputs, particularly when pulses of nutrients occurred in the spring period, the latter supporting the match-mismatch hypothesis.
Neisseria gonorrhoeae culture is required for antimicrobial susceptibility testing, but recovering isolates from clinical specimens is challenging. Although many variables influence culture recovery, ...studies evaluating the impact of culture specimen collection timing and patient symptom status are limited. This study analyzed urogenital and extragenital culture recovery data from Centers for Disease Control and Prevention's Strengthening the US Response to Resistant Gonorrhea (SURRG) program, a multisite project, which enhances local N. gonorrhoeae culture and antimicrobial susceptibility testing capacity.
Eight SURRG jurisdictions collected gonococcal cultures from patients with N. gonorrhoeae-positive nucleic acid amplification test (NAAT) results attending sexually transmitted disease and community clinics. Matched NAAT and culture specimens from the same anatomic site were collected, and culture recovery was assessed. Time between NAAT and culture specimen collection was categorized as same day, 1 to 7 days, 8 to 14 days, or ≥15 days, and patient symptoms were matched to the anatomic site where culture specimens were collected.
From 2018 to 2019, among persons with N. gonorrhoeae-positive NAAT, urethral infections resulted in the highest culture recovery (5927 of 6515 91.0%), followed by endocervical (222 of 363 61.2%), vaginal (63 of 133 47.4%), rectal (1117 of 2805 39.8%), and pharyngeal (1019 of 3678 27.7%) infections. Culture recovery was highest when specimens were collected on the same day as NAAT specimens and significantly decreased after 7 days. Symptoms were significantly associated with culture recovery at urethral (P = <0.0001) and rectal (P = <0.0001) sites of infection but not endocervical, vaginal, or pharyngeal sites.
Culture specimen collection timing and patient symptomatic status can impact culture recovery. These findings can guide decisions about culture collection protocols to maximize culture recovery and strengthen detection of antimicrobial-resistant infections.
This research offers a contextual snapshot—via the applied tensional analysis (ATA) method—of common barriers that educators face when taking on community engagement work. A six-person research team ...interviewed 22 faculty and staff actively pursuing community partnerships at three different institutions of higher education in a single region. Participants’ responses were coded to identify common barriers to engagement and the strategies that participants used to overcome those barriers. We draw on ATA (Mease, 2019) as a methodological approach that acknowledges tensions as critical entry points for understanding both the experiences of individual actors and the malleable structures from which those tensions emerge. This article does not attempt to offer one-size-fits-all strategies for community engagement. Instead, it offers ATA as a means for contextualizing specific tensions and strategies within local community engagement settings and for reframing those tensions as generative spaces for community engagement work.
